New series of pyrazolo[3,4-b]pyridines were prepared and evaluated for antimicrobial activity toward six selected microorganisms. Compounds 2a, 3b, 3d and 3e exhibited good activity toward B. cereus. On the other hand, 2a and 3b evinced interesting activity over C. albicans, whereas 2a, 3b and 3e displayed promising activity over A. fumigatus. Antiquorum-sensing effectiveness of the new members over C. violaceum was also assessed, where compounds 2a and 3b exhibited higher activity than that of the reference compound, indole. In vitro antiproliferative assessment toward HepG2, HCT-116 and MCF-7 cancer cells evidenced that 2f has notable effectiveness on all examined cell lines, whereas 3a–c were active but to a lower extent. In vivo antitumor activity of 2f and 3a–c against EAC cells was also esteemed, where 2f and 3c showed considerable activity comparable to that of doxorubicin. Cytotoxicity screening over WI38 and WISH normal cells evinced that 2f and 3a–c are less cytotoxic than doxorubicin. Compounds 2a, 2f, 3a–c and 3e were evaluated for DNA-binding affinity and topoisomerase IIβ inhibitory activity. Analogs 2a, 2f, 3a and 3b illustrated strong DNA-binding affinity, whereas 2a, 2f and 3a exhibited interesting topoisomerase IIβ inhibitory activity. Compounds 2a and 2f were docked into topoisomerase IIβ, where 2f showed preferential binding to topoisomerase IIβ. Computational studies articulated that the new members are in compliance with Veber's standards and Lipinski’s rule.  相似文献

Three-component reactions involving azaarenes (quinoline, isoquinoline, and phenanthridine) with acetylenic esters such as methyl propiolate or ethyl propiolate in the presence of NH-heterocyclic compounds (carbazole, maleimide, 5-nitroindazole, 2-benzoxazolinone, indole, and 2-methylindole) or 1,3-dicarbonyl compounds involving acetylacetone and N,N′-dimethylbar bituric acid are described. The reactions proceeded smoothly at room temperature without a catalyst and with excellent yields. This method is very useful for functionalizing aza-aromatic compounds in a one-pot operation.  相似文献

Condensation of 4-acetyl-5-methyl-2-phenyl-2,4-dihydropyrazol-3-one (1) with hydrazine derivatives (2a–d) afforded hydrazone derivatives (3a–d), which reacted with alkyl halides 4a–c to give bis(alkylthio)methylene derivatives (5a–e). Also, 3a,b reacted with hydrazonyl halides 6a–d to give 1,3,4-thiadiazole (7a–d). Cyclization of 3c with ethyl bromoacetate and haloketones gave thiazolidinone and thiazole derivatives (8, 10a,b) respectively. Treatment of hydrazone (3d) with benzylidine malononitrile 13a,b gave pyridine (14a,b). In addition, cyclocondensation of 3d with phenolic aldehydes furnished coumarin derivatives (16a–c). Coupling of 3d with heterocyclic diazonium salts gave triazol[5,1-c]triazine (20) and pyrazolo[5,1-c]triazine (22). Some of the prepared products showed potent antimicrobial activity.  相似文献

In this study, a theoretical work on magnetocaloric properties of ceramic and sol–gel La0.67Ca0.33MnO3 samples was presented. The value of the magnetocaloric effect has been determined from theoretical work on magnetization as a function of temperature. It is showed that ceramic sample shows better magnetocaloric properties than the sol–gel one. Though the maximum magnetic entropy change ?S M of ceramic sample upon 5 kOe applied field variation which is about 76 % of that of a pure Gd metal upon 5 kOe, ?S M distribution of the La0.67Ca0.33MnO3 is much more uniform than that of Gd. This feature is desirable for an Ericson-cycle magnetic refrigerator.  相似文献

A new, simple, precise, accurate and selective high-performance thin-layer chromatographic (HPTLC) method has been developed and validated for the simultaneous determination of ledipasvir and sofosbuvir in their tablet dosage form. Chromatographic separation was carried out on Merck TLC aluminum sheets of silica gel 60 F254 using ethyl acetate:hexane:methanol in the ratio of 8:1.25:0.75 (% v/v) as the mobile phase followed by densitometric measurement at 256 nm. The method was validated in terms of linearity, accuracy, precision, limit of detection, limit of quantification and specificity in accordance with the International Conference on Harmonization (ICH) guidelines. The calibration curve was found to be linear between 60 to 1980 and 45 to 3600 ng/band for ledipasvir and sofosbuvir, respectively, with significantly high value of regression coefficient (r2 > 0.9999) with linear and homoscedastic residuals. The limits of detection and quantification were found to be 16.5 and 50 ng/band, respectively, for ledipasvir and 13 and 39.5 ng/band, respectively, for sofosbuvir. Comparative study was performed between the developed HPTLC method and the reported high-performance liquid chromatography (HPLC) method. The quantitative results of the two analytical methods did not show statistically significant difference, whereas the developed HPTLC method is both time- and cost-effective.

A method is proposed and explored for speciation of Cr(III) and Cr(VI), selective extraction and preconcentration in various water samples based on dynamic and static techniques. Three newly designed alumina phases-physically adsorbed-2-pyridenecarboxyladehyde-thiosemicarbazone [Al-2PC-TSC (I–III)] were synthesised and characterised. Stability tests and application of [Al-2PC-TSC (I–III)] as inorganic ion exchangers and chelating solid sorbents for various metal ions were studied and evaluated. The distribution coefficient and separation factors of chromium species versus other interfering metal ions were determined to examine the incorporated selectivity into these alumina phases. Quantitative recovery of Cr(VI) was accomplished by [Al-2PC-TSC (I–III)] at pH = 1.0 while Cr(III) was found to be quantitatively recovered on these sorbents at pH = 7.0 with minimal or no interference between these two species under the studied buffering conditions. Selective solid phase speciation and preconcentration of Cr(III) and Cr(VI) in various real water samples were successfully performed and accomplished via a micro-column, with 200 as a preconcentration factor. Selective speciation of Cr(VI) and Cr(III) via preconcentration from seawater and industrial water samples by alumina phases in pH = 1 was found to give percentage recovery values of Cr(VI) in the range 93.5–97.3 ± 3.0–5.0% and 94.0–97.5 ± 3.0–4.0%, for seawater and industrial water samples, respectively.  相似文献
