Aptamers are RNA/DNA oligonucleotide molecules that specifically bind to a targeted complementary molecule. As potential recognition elements with promising diagnostic and therapeutic applications, aptamers, such as monoclonal antibodies, could provide many treatment and diagnostic options for blood diseases. Aptamers present several superior features over antibodies, including a simple in vitro selection and production, ease of modification and conjugation, high stability, and low immunogenicity. Emerging as promising alternatives to antibodies, aptamers could overcome the present limitations of monoclonal antibody therapy to provide novel diagnostic, therapeutic, and preventive treatments for blood diseases. Researchers in several biomedical areas, such as biomarker detection, diagnosis, imaging, and targeted therapy, have widely investigated aptamers, and several aptamers have been developed over the past two decades. One of these is the pegaptanib sodium injection, an aptamer-based therapeutic that functions as an anti-angiogenic medicine, and it is the first aptamer approved by the U.S. Food and Drug Administration (FDA) for therapeutic use. Several other aptamers are now in clinical trials. In this review, we highlight the current state of aptamers in the clinical trial program and introduce some promising aptamers currently in pre-clinical development for blood diseases.  相似文献

Novel screen-printed electrodes (SPEs) were constructed for the quantitation of nicorandil (NIC) in its pharmaceutical formulations. Different ion-exchangers and plasticizers were investigated, but the optimal potentiometric response was obtained using nicorandil-phosphotungstate (NIC-PTA) ion associate and tricresyl phosphate as a plasticizer. A Nernstian response of 58.80±1 mV/decade was obtained over a concentration range of (1×10−6–1×10−2) M with 1×10−5 M as a detection limit. Sensor morphology was characterized using scanning electron microscopy (SEM). The method was validated for the assay of NIC with high selectivity, accuracy (average recovery=100.54 %), and precision (%RSD≤2).  相似文献

The spectral properties of etoricoxib (ETR) at pH 2.0, 6.0 and 10.0 in the presence of cyclodextrins (CDs) were investigated. The absorption spectrum of ETR in acidic medium exhibited two bands centered at 236 and 273 nm, while in basic medium it exhibited two bands centered at 236 and 285 nm. No change in the spectrum was observed in the presence of CDs. The fluorescence emission spectra of ETR in acidic and basic media exhibited one band at 380 nm and another one at 484 nm. The emission band at 484 nm was enhanced when ETR was complexed with β-CD and γ-CD at pH 2.0, 6.0 and 10.0, while the band at 380 nm was enhanced selectively when ETR was complexed with α-CD at pH 2.0. Molecular dynamics simulations computations revealed that at pH 2.0, the sulfonyl moiety of H2ETR2+ is preferentially included within the α-CD cavity, which is believed to cause the enhancement of the band at 380 nm. Moreover, at pH 6.0 and 10.0, the enhancement of the band at 484 nm was related to the inclusion of the chloropyridinyl and methylpyridinyl groups of the bipyridine moiety of HETR+ and ETR within β-CD and γ-CD cavities. Benesi–Hildebrand analysis showed that the ETR/β-CD complex adopts a 1:1 stoichiometry with association constant of K 11?=?64.8 at pH 2.0, K 11?=?105.4 at pH 6.0 and K 11?=?520.5 at pH 10.0.  相似文献

The kinetics of oxidation of the diaqua(nitrilotriacetato)chromium(III) complex [CrIIInta(H2O)2], by N-bromosuccinimide (NBS) to chromium(VI) in aqueous solution obeys the equation: where k1 is the rate constant for the electron-transfer process, K1 the equilibrium constant for the dissociation of [CrIIInta(H2O)2] to [CrIIInta(H2O)(OH)]−, and K2 is the pre-equilibrium formation constant for the precursor complex [CrIIInta(OH)(NBS)]−. The thermodynamic activation parameters were calculated and it is proposed that electron transfer proceeds via an inner-sphere mechanism. The Myrteacae family is known as a rich source of phloroglucinols, a group of secondary metabolites with notable biological activities. Leaves of Psidium cattleianum were extracted with chloroform: methanol 8:2 to target the isolation of phloroglucinol derivatives. Isolated compounds were characterized using different spectroscopic methods: nuclear magnetic resonance (NMR), ultra-violet (UV) and mass spectrometry (MS). Two new phloroglucinols were evaluated for cytotoxicity against a panel of six human cancer cell lines, namely colorectal adenocarcinoma cells (HT-29 and HCT-116); hepatocellular carcinoma cells (HepG-2); laryngeal carcinoma (Hep-2); breast adenocarcinoma cells (MCF7 and MDA-MB231), in addition to normal human melanocytes HFB-4. Additionally, cell cycle analysis and annexin-V/FITC-staining were used to gain insights into the mechanism of action of the isolated compounds. The new phloroglucinol meroterpenoids, designated cattleianal and cattleianone, showed selective antiproliferative action against HT-29 cells with IC50’s of 35.2 and 32.1 μM, respectively. Results obtained using cell cycle analysis and annexin-V/FITC-staining implicated both necrosis and apoptosis pathways in the selective cytotoxicity of cattleianal and cattleianone. Our findings suggest that both compounds are selective antiproliferative agents and support further mechanistic studies for phloroglucinol meroterpenoids as scaffolds for developing new selective chemotherapeutic agents.  相似文献

The amlyoid-β peptide (Aβ) is closely linked to the development of Alzheimer''s disease. Molecular dynamics (MD) simulations have become an indispensable tool for studying the behavior of this peptide at the atomistic level. General key aspects of MD simulations are the force field used for modeling the peptide and its environment, which is important for accurate modeling of the system of interest, and the length of the simulations, which determines whether or not equilibrium is reached. In this study we address these points by analyzing 30-μs MD simulations acquired for Aβ40 using seven different force fields. We assess the convergence of these simulations based on the convergence of various structural properties and of NMR and fluorescence spectroscopic observables. Moreover, we calculate Markov state models for the different MD simulations, which provide an unprecedented view of the thermodynamics and kinetics of the amyloid-β peptide. This further allows us to provide answers for pertinent questions, like: which force fields are suitable for modeling Aβ? (a99SB-UCB and a99SB-ILDN/TIP4P-D); what does Aβ peptide really look like? (mostly extended and disordered) and; how long does it take MD simulations of Aβ to attain equilibrium? (at least 20–30 μs). We believe the analyses presented in this study will provide a useful reference guide for important questions relating to the structure and dynamics of Aβ in particular, and by extension other similar disordered proteins.

The convergence of MD simulations is tested using varying measures for the intrinsically disordered amyloid-β peptide (Aβ). Markov state models show that 20–30 μs of MD is needed to reliably reproduce the thermodynamics and kinetics of Aβ.  相似文献

The integrity, function, and performance of biomedical devices having thin polymeric coatings are critically dependent on the mechanical properties of the film, including the elastic modulus. In this report, the elastic moduli of several tyrosine-derived polycarbonate thin films, specifically desaminotyrosyl ethyl tyrosine polycarbonates p(DTE carbonate), an iodinated derivative p(I(2)-DTE carbonate), and several discrete blends are measured using a method based on surface wrinkling. The data shows that the elastic modulus does not vary significantly with the blend composition as the weight percentage of p(I(2)-DTE carbonate) increases for films of uniform thickness in the range of 67 to 200 nm. As a function of film thickness, the observed elastic moduli of p(DTE carbonate), p(I(2)-DTE carbonate) and their 50:50 by mass blend show little variation over the range 30 to 200 nm.  相似文献

The nucleophilic aromatic substitution under ultrasound irradiation of a dichlorobenzene iron η6-complex with various secondary amines is reported. The reaction time at moderate temperatures is considerably shortened (15 min) compared to non sonicated reaction conditions at room temperature (several days) or at solvent refluxing temperature (12-48 h). Controlled mono- or di-substitution was achieved by the tuning of the amine nucleophilicity and the solvent polarity. The method was successfully applied to the synthesis of differently substituted phenylenediamines.  相似文献
