The Mitochondria-Independent Cytotoxic Effect of Leflunomide on RPMI-8226 Multiple Myeloma Cell Line

Leflunomide, an anti-inflammatory agent, has been shown to be effective in multiple myeloma (MM) treatment; however, the mechanism of this phenomenon has not been fully elucidated. The aim of the study was to assess the role of mitochondria and dihydroorotate dehydrogenase (DHODH) inhibition in the cytotoxicity of leflunomide in relation to the MM cell line RPMI 8226. The cytotoxic effect of teriflunomide—an active metabolite of leflunomide—was determined using MTT assay, apoptosis detection, and cell cycle analysis. To evaluate DHODH-dependent toxicity, the cultures treated with teriflunomide were supplemented with uridine. Additionally, the level of cellular thiols as oxidative stress symptom was measured as well as mitochondrial membrane potential and protein tyrosine kinases (PTK) activity. The localization of the compound in cell compartments was examined using HPLC method. Teriflunomide cytotoxicity was not abolished in uridine presence. Observed apoptosis occurred in a mitochondria-independent manner, there was also no decrease in cellular thiols level. Teriflunomide arrested cell cycle in the G2/M phase which is not typical for DHODH deficiency. PTK activity was decreased only at the highest drug concentration. Interestingly, teriflunomide was not detected in the mitochondria. The aforementioned results indicate DHODH- and mitochondria-independent mechanism of leflunomide toxicity against RPMI 8226 cell line. 相似文献

Salt concentrations in brine and temperature are the major environmental factors that affect activity of microorganisms and, thus may affect formation of biogenic amines (BAs) during the fermentation process. A model system to ferment cucumbers with low salt (0.5%, 1.5% or 5.0% NaCl) at two temperatures (11 or 23 °C) was used to study the ability of indigenous microbiota to produce biogenic amines and metabolize amino acid precursors. Colony counts for presumptive Enterococcus and Enterobacteriaceae increased by 4 and up to 2 log of CFU∙mL−1, respectively, and remained viable for more than 10 days. 16S rRNA sequencing showed that Lactobacillus and Enterobacter were dominant in fermented cucumbers with 0.5% and 1.5% salt concentrations after storage. The initial content of BAs in raw material of 25.44 ± 4.03 mg∙kg−1 fluctuated throughout experiment, but after 6 months there were no significant differences between tested variants. The most abundant BA was putrescine, that reached a maximum concentration of 158.02 ± 25.11 mg∙kg−1. The Biogenic Amines Index (BAI) calculated for all samples was significantly below that needed to induce undesirable effects upon consumption. The highest value was calculated for the 23 °C/5.0% NaCl brine variant after 192 h of fermentation (223.93 ± 54.40). Results presented in this work indicate that possibilities to control spontaneous fermentation by changing salt concentration and temperature to inhibit the formation of BAs are very limited. 相似文献

Mammalian reovirus is an enteric virus that contains a double-stranded RNA genome. The genome consists of ten RNA segments that encode eight structural and three non-structural proteins. The structural proteins form a double-layered structure. The innermost layer, called the core, consists of five proteins (lambda1, lambda2, lambda3, micro 2, and sigma2). Protein lambda3 is the RNA-dependent RNA polymerase (RdRp) and micro 2 is thought to be an RdRp cofactor. Translation of most reovirus proteins is known to commence at the first start codon. However, the translation initiation site of the viral core protein micro 2, encoded by the M1 RNA segment, has been in dispute. Although the theoretical molecular weight of micro 2 is 83 267 Da the actual molecular weight is unknown because micro 2 runs aberrantly in SDS-PAGE and has resisted characterization by Edman degradation, indicating that the amino terminus is post-translationally modified. In this study, we used proteolysis coupled with MALDI-Qq-TOFMS to determine that translation of micro 2 initiates at the first AUG codon, that its actual molecular weight approximates the theoretical value of 83 kDa, that the amino terminal methionine residue is removed, and that the next amino acid (alanine) is post-translationally acetylated. 相似文献

Liquid secondary ion (LSI) mass spectra of ion-pair precipitates obtained for Triton X-100 with strontium, lead, cadmium and mercury tetraphenylborates and for selected butoxylene-ethoxylene monoalkyl ethers with barium tetraiodobismuthate(III) are discussed. A series of novel aza-BODIPY dyes substituted with p-(dimethylamino)phenyl groups were synthesized and their spectral and electrochemical properties were compared. In particular, the impact of p-(Me2N)Ph- groups on these characteristics was of consideration. For two aza-BODIPYs studied, a near-IR absorption band was observed at circa λabs=796 nm. Due to the pronounced intramolecular charge transfer (ICT) exerted by the presence of strongly electron-donating p-(Me2N)Ph- substituents, the compounds studied were weakly emissive with the singlet lifetimes (τS) in the picosecond range. Nanosecond laser photolysis experiments of the brominated aza-BODIPYs revealed T1→Tn absorption spanning from ca. 350 nm to ca. 550 nm with the triplet lifetimes (τT) ranged between 6.0 μs and 8.5 μs. The optical properties of the aza-BODIPYs studied were pH-sensitive. Upon protonation of the dimethylamino groups with trifluoroacetic acid in toluene, a stepwise disappearance of the NIR absorption band at λabs=790 nm was observed with the concomitant appearance of a blue-shifted absorption band at λabs=652 nm, which was accompanied by a prominent emission band at λfl=680 nm. The transformation from a non-emissive to an emissive compound is associated with the inhibition of the ICT. As estimated by CV/DPV measurements, all aza-BODIPYs studied exhibited two irreversible oxidation and two quasi-reversible reduction processes. All compounds studied exhibit extremely high photostability and thermal stability. 相似文献

The properties of the carbonate neutral carrier 4-( n-hexadecyl)-3-nitro-1-trifluoroacetylbenzene were compared with those of a similar carrier, without a nitro group, studied previously. In spite of differences in the Hammett constant of the carbonyl group responsible for interaction with the analyte, the analytical characteristics of both carriers, measured under the same conditions, were comparable. Special care was taken to avoid the presence of an excessive carbon dioxide level in the diffusion layer at the membrane-solution interface. The internal reference solution was prepared without carbonate components; the external solution was protected from contact with atmospheric carbon dioxide. Under such conditions the detection limit of both electrodes was extended to 10(-11 )mol L(-1), and the selectivity towards salicylate, chloride, and acetate was significantly improved. 相似文献

The LC-MS/MS was applied for the determination of flavonoids' stability under four types of solvent extraction methods (reflux heating, sonication, maceration and microwave) from maize samples. The 11 flavonoids belong to different groups: flavonols (kaempferol, myricetin, rhamnetin, quercetin, rutin), flavanones (naringenin, naringin, hesperedin), flavones (apigenin, luteolin), isoflavones (genistein) were studied. The effect of the degradation of flavonoids depended on extraction mode and chemical structure. The smallest decomposition was observed by heated reflux extraction procedure within 30 min in water bath and by microwave assisted extraction under 160 W during 1 min. The decomposition for flavonoids depends on number of substituents in flavonoid molecule. The most unstable compound (recovery below 50%) in tested condition was myricetin. The higher number of hydroxyl groups promote degradation of flavonoids, whereas sugar moiety and methoxyl groups protect flavonoids of degradation during microwave and ultrasonic assisted extraction. 相似文献
