A rapid, sensitive and rugged solid‐phase extraction ultra performance liquid chromatography tandem mass spectrometry (UPLC‐MS/MS) method was developed for determination of paroxetine in human plasma. The procedure for sample preparation includes simple SPE extraction procedure coupled with Hypersil Gold C18 column (100 mm ? 2.1 mm, i.d., 1.9 μm) with isocratic elution at a flow‐rate of 0.350 mL/min and fluoxetine was used as the internal standard. The analysis was performed on a triple‐quadrupole tandem mass spectrometer by multiple reactions monitoring mode via electrospray ionization. Using 500 μL plasma, the methods were validated over the concentration range 0.050–16.710 ng/mL for paroxetine, with a lower limit of quantification of 0.050 ng/mL. The intra‐ and inter‐day precision and accuracy of the quality control samples were within 10.0%. The recovery was 69.2 and 74.4% for paroxetine and fluoxetine respectively. A simple, sensitive and selective LC–MS–MS method has been developed for the simultaneous determination of acyclovir and valacyclovir in human plasma. Acyclovir and valacyclovir in plasma were concentrated by solid phase extraction and chromatographed on a C18 column using a mobile phase of 0.1% formic acid: methanol (30:70% v/v). The method was validated over a linear range of 47–10,255 and 5–1,075 ng mL?1 for acyclovir and valacyclovir respectively. The LOQs were 47.6 and 5.0 ng mL?1. The validated method was applied for the quantitation of acyclovir and valacyclovir from plasma samples in a pharmacokinetic study.  相似文献

As per the World Health Organization (WHO), cancer is the second most leading cause of death after cardiovascular diseases in worldwide with around 9.88 million total new cases and 1.08 million were observed due to skin cancer in 2018. Amongst two types of skin cancer, progression of melanoma cancer is increasing day by day due to the environmental changes than non-melanoma cancer. Most of B-Raf mutation, specifically B-RafV600E, is responsible for the progression of the melanoma cancer. Here, various 3D-QSAR techniques like comparative molecular field analysis (CoMFA), comparative molecular similarity indices analysis (CoMSIA), molecular hologram QSAR (HQSAR) and topomer CoMFA were used to design novel B-Raf inhibitors by using 28 synthetic B-Raf inhibitors. Except for topomer CoMFA model, remaining models were generated by three different alignment methods in which distil-based alignment method was found best and gave prominent statistical values. After performing N-fold statistical validation, in CoMFA, q2, r2 and r2pred values were found to be 0.638, 0.969 and 0.848, respectively. Similarly, q2, r2 and r2pred values were found to be 0.796, 0.978 and 0.891 in CoMSIA (SHD) and 0.761, 0.973 and 0.852 in CoMSIA (SH) by N-fold statistical validation. In HQSAR analysis, statistical values were found for q2 as 0.984, r2 as 0.999 and r2pred as 0.634 with 97 as best hologram length (BHL). The results of topomer CoMFA showed the q2 value of 0.663 and the r2 value of 0.967. Important features of purinylpyridine were identified by contour map analysis of all 3D-QSAR techniques, which could be useful to design the novel molecules as B-Raf inhibitors for the treatment of melanoma cancer.

A family of calix-crown hybrid molecules containing calix[4]arene and crown-5/6, either at lower rim or at both upper and lower rims, have been synthesised, characterised and their competitive complexation property towards alkali and alkaline earth metal ions in aqueous media have been investigated. The competitive metal ion extraction study, carried out with equimolar mixture of Li+, Na+, K+, Mg2+, Ca2+ and Sr2+ in aqueous media, revealed that the amount of K+ extracted is remarkably high compared to other metal ions. Complexation with K+ has been investigated by 1H NMR, association constants and thermodynamic parameters have been determined by isothermal calorimetric study. The molecular structures of one of the receptors and two of the K+ complexes have been established by single crystal X-ray study. One of the receptors formed bimetallic complex and it exhibited interesting polymeric network structure with bridged picrate anion. These receptors have been applied for extraction of metal ions from bittern.  相似文献

A sensitive LC–MS/MS method was developed and validated for quantitation of saroglitazar using turboion spray interface with positive ion mode. A liquid–liquid extraction, with a mixture of dichloromethane and diethyl ether, was employed for the extraction of saroglitazar and glimepiride (IS) from human plasma. The chromatographic separation was achieved using an ACE‐5, C18 (4.6 × 100 mm) column with a gradient mobile phase comprising acetonitrile and ammonium acetate buffer with trifluoracetic acid in purified water. Both analytes were separated within 10 min with retention times of 4.52 and 2.57 min for saroglitazar and IS, respectively. Saroglitazar quantitation was achieved by the summation of two MRM transition pairs (m/z 440.2 to m/z 366.0 and m/z 440.2 to m/z 183.1), while that of IS was achieved using transition pair m/z 491.3 to m/z 352.0. The calibration standards of saroglitazar showed linearity from 0.2 to 500 ng/mL, with a lower limit of quantitation of 0.2 ng/mL. The biases for inter‐ and intra‐batch assays were ?7.51–1.15% and ?11.21 to ?3.25%, respectively, while the corresponding precisions were 5.04–8.06% and 1.53–7.68%, respectively. The developed method was used to monitor the plasma concentrations of saroglitazar in clinical samples.  相似文献

Novel thermally stable mesoporous mixed metal Nb-M (M = V, Mo and Sb) oxides were synthesized in the presence of a nonionic Pluronic P123 surfactant. These oxides displayed promising pore structures and chemical compositions for selective oxidative functionalization of propane: high surface areas (up to 200 m2/g), large pore sizes (5-14 nm), and high pore volumes (up to 0.46 cm3/g). The oxidative dehydrogenation of propane to propylene over mesoporous mixed metal Nb-M oxides employed as a probe reaction suggested that the M component was dispersed as the molecular surface species and also formed a solid solution with NbOx in the inorganic walls of these mesoporous mixed metal oxides.  相似文献

The network formation and viscoelastic behavior of a liquid crystalline monomer, whose structure includes both acrylate and acetylene reactive groups, have been studied. By combining both photo and thermal polymerization, the networks can be formed in two separate steps, with the initial photopolymerization dominated by acrylate crosslinking and subsequent thermal polymerization dominated by acetylene crosslinking. In addition, the monomer exhibits a liquid crystalline phase. Photopolymerization while in the liquid crystal phase locks in the molecular ordering. Dynamic mechanical analysis shows that networks formed from the liquid crystalline phase have lower crosslink densities and narrower distributions of molecular weights between crosslinks when compared to networks formed from the isotropic phase (and at higher polymerization temperatures). After thermal postcure at 250°C, the networks formed from the isotropic monomer have a 23% higher dynamic mechanical storage modulus (in the glassy state) than the networks formed from the liquid crystalline monomer. A series of novel compounds 1-((1-(4-(2H-benzo[e][1,3]oxazin-3(4H)-yl)phenyl)ethylidene)amino)-6-((benzylidene)amino)-2-oxo-4-phenyl-1,2-dihydropyridine-3,5-dicarbonitriles (7a–o) were synthesized by a sequence of multistep reactions. IR, 1H NMR, 13C NMR, and mass spectral techniques were used to confirm the structures newly synthesized compounds. Antimicrobial activity of the title compounds (7a–o) was studied against strains of bacteria (Staphylococcus aureus and Streptococcus pyogenes, Escherichia coli and Pseudomonas aeruginosa) and fungi (Candida albicans, Aspergillus niger and Aspergillus clavatus) by serial dilution method. Compounds 7f and 7k exhibited significant antimicrobial activity.  相似文献

Optical absorption measurements were made on tin diselenide thin films grown on glass substrates by thermal evaporation of stoichiometric bulk compound. In the as-deposited films, optical absorption was dominated by an indirect transition at lower photon energies with a band gap of 1.4 eV. The direct allowed transition with a band gap of 1.62 eV was predominant at higher energies. Annealing the film at elevated temperatures resulted in the decrease of both the direct and indirect transition band gaps.  相似文献
