Amaryllidaceae species are known as ornamental plants. Some contain galanthamine, an acetylcholinesterase inhibitor. The chemical composition of the alkaloid extract of bulbs of Nerine bowdenii Watson has been analyzed by means of GC/MS. Twenty-two compounds were detected and nineteen of them identified, one of which was belladine. The alkaloid extract showed promising cholinesterase inhibitory activities against human blood acetylcholinesterase (HuAChE; IC50 = 87.9 +/- 3.5 microg/mL) and human plasma butyrylcholinesterase (HuBuChE; IC50 = 14.8 +/- 1.1 microg/mL). Belladine inhibited HuAChE and HuBuChE in a dose-dependent manner with IC50 values of 781 +/- 12.5 microM and 284.8 +/-4.2 microM, respectively.  相似文献

Reduction potentials of several M(2+/3+) (M = Ru, Os) octahedral complexes, namely, [M(H2O)6](2+/3+), [MCl6](4-/3-), [M(NH3)6](2+/3+), [M(en)3](2+/3+) [M(bipy)3](2+/3+), and [M(CN)6](4-/3-), were calculated using the CASSCF/CASPT2/CASSI and MRCI methods including spin-orbit coupling (SOC) by means of first-order quasi-degenerate perturbation theory. It was shown that the effect of SOC accounts for a systematic shift of approximately -70 mV in the reduction potentials of the studied ruthenium (II/III) complexes and an approximately -300 mV shift for the osmium(II/III) complexes. SOC splits the sixfold-degenerate (2)T(2g) ground electronic state (in ideal octahedral symmetry) of the M(3+) ions into the E((5/2)g) Kramers doublet and G((3/2)g) quartet, which were calculated to split by 1354-1573 cm(-1) in the Ru(3+) complexes and 4155-5061 cm(-1) in the Os(3+) complexes. It was demonstrated that this splitting represents the main contribution to the stabilization of the M(3+) ground state with respect to the closed-shell (1)A(1g) ground state in M(2+) systems. Moreover, it was shown that the accuracy of the calculated reduction potentials depends on the calculated solvation energies of both the oxidized and reduced forms. For smaller ligands, it involves explicit inclusion of the second solvation sphere into the calculations, whereas implicit solvation models yield results of sufficient accuracy for complexes with larger ligands. In such cases (e.g., [M(bipy)3](2+/3+) and its derivatives), very good agreement between the calculated (SOC-corrected) values of the reduction potentials and the available experimental values was obtained. These results led us to the conclusion that especially for Os(2+/3+) complexes, inclusion of SOC is necessary to avoid systematic errors of approximately 300 mV in the calculated reduction potentials.  相似文献

The multiplication of terrorist actions in the recent events is alarming and the detection of chemical warfare agents (CWAs) has become one of the highest research priorities in the fields of security and public health. The biomimetic properties of molecularly imprinted polymers (MIPs) render them attractive for molecular recognition as well as sensing purposes. The degradation products of easily hydrolysable organophosphorus nerve agents such as pinacolyl methylphosphonate (PMP), a hydrolysis by-product of soman, are often used as templates in MIP synthesis. In this study, we describe the first example of PMP-imprinted polymer microspheres synthesized by precipitation polymerization. This one-step process involves methacrylic acid (MAA) as the monomer and divinylbenzene (DVB) as the cross-linker, in a toluene/acetonitrile mixture. Subsequent morphological characterizations of the PMP-imprinted particles show that they have diameters between 1 and 10 mum (as opposed to 4-5 mum for the non-imprinted microspheres), surface areas of up to 680 m(2) g(-1) and high porosities with pore sizes smaller than 2 nm. The present investigation also evidences the imprinting effect via batch binding experiments and reports on the use of a novel fluorescence-based methodology, where 4-methylumbelliferone (4MU) is utilised as a sensing agent to determine the PMP concentration in solution.  相似文献

We report an efficient and streamlined way to improve the analysis and identification of peptides and proteins in complex mixtures of soluble proteins, cell lysates, etc. By using the shotgun proteomics methodology combined with bioaffinity purification we can remove or minimize the interference contamination of a complex tryptic digest and so avoid the time-consuming separation steps before the final MS analysis. We have proved that by means of enzymatic fragmentation (endoproteinases with Arg-C or/and Lys-C specificity) connected with the isolation of specific peptides we can obtain a simplified peptide mixture for easier identification of the entire protein. A new bioaffinity sorbent was developed for this purpose. Anhydrotrypsin (AHT), an inactive form of trypsin with an affinity for peptides with arginine (Arg) or lysine (Lys) at the C-terminus, was immobilized onto micro/nanoparticles with superparamagnetic properties (silica magnetite particles (SiMAG)-Carboxyl, Chemicell, Germany). This AHT carrier with a determined binding capacity (26.8 nmol/mg of carrier) was tested with a model peptide, human neurotensin, and the resulting MS spectra confirmed the validity of this approach.  相似文献

The study includes findings about limitations of acetylcholinesterase (AChE) based assay. Eleven organophosphorus pesticides: chlorpyrifos ethyl, chlorpyrifos methyl, DFP, dichlorvos, dimethoate, fenthion, paraoxon ethyl, paraoxon methyl, phosalone, pirimiphos methyl and pirimiphos ethyl were photometrically assayed using AChE as a recognition element. The study was carried out in order to find approachability of AChE based assay. In the first round, common organic solvents were tested for interfering in assay, since samples collection and extraction is a necessary part in samples processing. Isopropanol was found as the most convenient due to minimal inhibition not exceeding 5%. Though all analysed pesticides inhibit AChE in vivo, some of them are toxic after metabolisation. We found AChE based assay approachable for assay of DFP, paraoxons, and dichlorvos. These are oxoforms of organophosphorus pesticides. From thioforms of assayed pesticides, only fenthion was able significantly inhibit AChE in vitro. Electrochemical biosensor with AChE attached on platinum electrode was used for confirmation of interaction pesticide – AChE and complex stability estimation. DFP, paraoxons and dichlorvos were allowed to interact with AChE in biosensor. These pesticides were settled firmly in AChE active site as no spontaneous recovery of AChE activity was observed.  相似文献

Organic photochromic molecules are important for the design of photoresponsive functional materials, as switches and memories. Over the past 10 years, research efforts have been directed towards the incorporation of photoresponsive molecules into metal systems, in order either to modulate the photochromic properties, or to photoregulate the redox, optical and magnetic properties of the organometallic moieties. This review article focuses on some of the recent work reported within the last few years in the area of organometallic and coordination complexes containing photochromic ligands for the photoregulation of optical and nonlinear optical properties. The first part is related to photochromic 1,2-diarylethene (DAE)-containing metal complexes, examples of mono- and multi-DAE metal-based will be discussed. The second part deals with metal complexes incorporating spiropyran and spirooxazine derivatives.  相似文献

The antimony film microelectrode (SbFME) is presented for measuring trace heavy metal ions in combination with anodic stripping voltammetry. The SbFME was tested in model solutions of 0.01 M HCl containing Cd(II) and Pb(II) in the presence of dissolved oxygen. The microsensor exhibited good linear behavior in the examined concentration range 20 μg L−1 to 100 μg L−1 and LoD of 1.9 and 3.1 μg L−1 for Cd(II) and Pb(II), respectively. The suitability of SbFME for measuring trace levels of Cu(II) was demonstrated using the standard reference material of natural water exploiting the sensor's specific characteristic of low re‐oxidation signal for antimony.  相似文献
