Alzheimer’s disease (AD) is a complex neurodegenerative disorder with a multifaceted pathogenesis. This fact has long halted the development of effective anti-AD drugs. Recently, a therapeutic strategy based on the exploitation of Brazilian biodiversity was set with the aim of discovering new disease-modifying and safe drugs for AD. In this review, we will illustrate our efforts in developing new molecules derived from Brazilian cashew nut shell liquid (CNSL), a natural oil and a byproduct of cashew nut food processing, with a high content of phenolic lipids. The rational modification of their structures has emerged as a successful medicinal chemistry approach to the development of novel anti-AD lead candidates. The biological profile of the newly developed CNSL derivatives towards validated AD targets will be discussed together with the role of these molecular targets in the context of AD pathogenesis.  相似文献

Poly(methyl methacrylate) (PMMA)-based polymers have been extensively used for manufacturing of artificial bone cements for treatment of osteoporosis. A typical bone cement recipe contains methyl methacrylate, which polymerizes in situ during cement application. An inherent problem of this reaction is the high amount of heat released during the cement preparation, which may lead to irreparable damage of living tissues. Optimization of PMMA-based bone cement (PMMABC) recipes is thus an important step towards safe and reliable clinical usage of these materials. A theoretical and experimental investigation is performed here to unveil the influence of some preparation variables on the production of PMMABC and to allow for future optimization of the PMMABC recipe. It is shown that the degree of mixing of the components of the recipe plays a fundamental role on the development of the temperature profile. For this reason, the PMMABC obtained with the in-situ blending of PMMA and barium sulfate during the suspension polymerization leads to much better homogeneity of the final test pieces and improved control of the temperature profile.  相似文献

Novel electrochemical sensors for epinephrine (EP) based on a glassy carbon electrode (GCE) modified with a redox polymer film and iron (III) oxide nanoparticles (Fe2O3NP) have been developed. Two redox polymers‐poly(brilliant cresyl blue) (PBCB) and poly(Nile blue) (PNB), and two different architectures‐polymer/Fe2O3/GCE and Fe2O3/polymer/GCE were investigated. The electrochemical oxidation of epinephrine at the modified electrodes was performed by differential pulse voltammetry (DPV), in pH 7 electrolyte, and the analytical parameters were determined. The results show enhanced performance, more sensitive responses and lower detection limits at the modified electrodes, compared to other electrochemical epinephrine sensors reported in the literature. The best voltammetric response with the lowest detection limit was obtained for the determination of epinephrine at PBCB/Fe2O3/GCE. The novel sensors are reusable, with good reproducibility and stability, and were successfully applied to the determination of epinephrine in commercial injectable adrenaline samples.  相似文献

Essential oils (EOs) are a mixture of chemical compounds with a long history of use in food, cosmetics, perfumes, agricultural and pharmaceuticals industries. The main object of this study was to find chemical patterns between 45 EOs and antiprotozoal activity (antiplasmodial, antileishmanial and antitrypanosomal), using different machine learning algorithms. In the analyses, 45 samples of EOs were included, using unsupervised Self-Organizing Maps (SOM) and supervised Random Forest (RF) methodologies. In the generated map, the hit rate was higher than 70% and the results demonstrate that it is possible find chemical patterns using a supervised and unsupervised machine learning approach. A total of 20 compounds were identified (19 are terpenes and one sulfur-containing compound), which was compared with literature reports. These models can be used to investigate and screen for bioactivity of EOs that have antiprotozoal activity more effectively and with less time and financial cost.  相似文献

Molecule interacting with CasL 2 (MICAL2), a cytoskeleton dynamics regulator, are strongly expressed in several human cancer types, especially at the invasive front, in metastasizing cancer cells and in the neo-angiogenic vasculature. Although a plethora of data exist and stress a growing relevance of MICAL2 to human cancer, it is worth noting that only one small-molecule inhibitor, named CCG-1423 (1), is known to date. Herein, with the aim to develop novel MICAL2 inhibitors, starting from CCG-1423 (1), a small library of new compounds was synthetized and biologically evaluated on human dermal microvascular endothelial cells (HMEC-1) and on renal cell adenocarcinoma (786-O) cells. Among the novel compounds, 10 and 7 gave interesting results in terms of reduction in cell proliferation and/or motility, whereas no effects were observed in MICAL2-knocked down cells. Aside from the interesting biological activities, this work provides the first structure–activity relationships (SARs) of CCG-1423 (1), thus providing precious information for the discovery of new MICAL2 inhibitors.  相似文献

This paper presents the development of a capillary electrophoresis method with diode array detector coupled to multivariate curve resolution–alternating least squares (MCR-ALS) to conduct the resolution and quantitation of a mixture of six quinolones in the presence of several unexpected components. Overlapping of time profiles between analytes and water matrix interferences were mathematically solved by data modeling with the well-known MCR-ALS algorithm. With the aim of overcoming the drawback originated by two compounds with similar spectra, a special strategy was implemented to model the complete electropherogram instead of dividing the data in the region as usually performed in previous works. The method was first applied to quantitate analytes in standard mixtures which were randomly prepared in ultrapure water. Then, tap water samples spiked with several interferences were analyzed. Recoveries between 76.7 and 125 % and limits of detection between 5 and 18 μg L?1 were achieved.  相似文献

Abstract

The aim of this review is to combine all the information related to the characterization of skin tissues and cells, focusing on the identification of the specific biochemical characteristics of skin cancer. We have characterized two types of melanoma by FTIR spectroscopy: a murine melanoma (B16F10 cell line) and a human melanoma (C8161 cell line). The cells were deposited on IR transparent CaF2 windows, the spectral range used lay between 900-4000 cm? 1, transmission mode with 2 cm? 1 resolution, and 32 scans. A biochemical association is presented for all the absorption bands identified in this study. Besides the characterization of both cell lines above, a collection of works done in the area of skin cancer was also carried out, in this review; some interesting results obtained by different authors with respect to the characterization of different samples (melanomas, follicle sheath, basal cell carcinoma, epidermis tissues, dermis tissue, and human stratum corneum) are presented and compared to the chemical and biological associations performed in each case. The identification of biochemical injuries provides important information that, associated with clinical examination, can assist the diagnosis of diseases. Several FTIR techniques can be used in the diagnosis of biochemical changes in biological tissues, by identifying molecular markers associated with malignant and benign changes or variations in the composition of amino acids in tissues and cells. In the near future, a further study to compare histopatological analysis and biochemical characterization by FTIR spectroscopy would be interesting, in order to verify the accuracy and validate the applicability of this technique in the diagnosis of skin lesions from a statistical viewpoint.  相似文献

An HPLC method with an evaporative light scattering detector was optimized and validated for quantification of carbohydrates in beer. The chromatographic separation was achieved using a Spherisorb NH2, 5 microm chromatographic column and gradient elution with acetonitrile/water. The determinations were performed in the linear range of 0.05-5.0 g/L for fructose, 0.05-5.0 g/L for glucose, 0.05-15.0 g/L for maltose, 0.05-10.0 g/L for maltotriose, and 0.05-5.0 g/L for maltotetraose. The detection limits were 0.005 g/L for fructose, 0.008 g/L for glucose, and 0.01 g/L for maltose, maltotriose, and maltotetraose. The reliability of the method in terms of precision and accuracy was evaluated in three beer matrices, low alcohol beer, 6% alcohol beer, and beer made with part of adjuncts (4.5% alcohol). Relative standard deviations (RSDs) ranged between 1.59 and 5.95% (n = 10), and recoveries ranged between 94 and 98.4%.  相似文献

Blooms of Ostreopsis spp. have been recently reported along the Mediterranean coasts of Spain, France, Italy, and Greece posing serious risks to human health. Occurrence of Ostreopsis spp. may result in palytoxin contamination of seafood and, in order to prevent sanitary risks, the need exists to develop efficient extraction procedures to be coupled to rapid and sensitive monitoring methods of palytoxin-like compounds in seafood. In the present study, the best conditions for both extraction of palytoxin from seafood and palytoxin quantification by using liquid chromatography tandem mass spectrometry (LC-MS/MS) were investigated. Three seafood matrices (mussels, sea-urchins, and anchovies) were selected and five different extraction systems were tested, namely: the official protocol for extraction of lipophilic toxins and various aqueous methanol or acetonitrile solutions (MeOH/H2O 1:1, MeOH/H2O 8:2, MeCN/H2O 8:2 and MeOH 100%). Extraction with MeOH/H2O 8:2 provided the best results in terms of accuracy and matrix interference on LC-MS/MS detection of palytoxin. Accuracy and intra-day reproducibility (n = 3) were evaluated for all the selected matrices but only for mussels at three spiking concentration levels, including the provisional limit proposed by the Community Reference Laboratory for marine biotoxins (250 μg kg−1). Limits of quantitation of palytoxin in mussels, sea-urchins and anchovies tissues were calculated using matrix-matched standards; taking into account extraction efficiency of MeOH/H2O 8:2, they resulted to be 228, 343, and 500 μg kg−1, respectively.  相似文献
