The ligand class 2,2'-pyridylpyrrolide is surveyed, both for its structural features and its electronic structure, when attached to monovalent K, Cu, Ag, Au, and Rh. The influence of pyrrolide ring substituents is studied, as well as the question of push/pull interaction between the pyridyl and pyrrolide halves. The π donor ability of the pyrrolide is found to be less than that of an analogous phenyl. However, in contrast to the phenyl analog, the HOMO is pyrrolide π in character for pyridylpyrrolide complexes of copper and rhodium, while it is conventionally metal localized for planar, d(8) rhodium pyridylphenyl. Monovalent three-coordinate copper complexes show great deviations from Y-shaped toward T-shaped structures, including cases where the pyridyl ligand bonds only weakly. 相似文献

The aim of this study was to develop a novel, sensitive, precise, simple, and rapid capillary zone electrophoresis method for the quality control of spironolactone in three different formulation types and a rapid simultaneous determination of the content of spironolactone and canrenone in urine samples using fluocinonide as an internal standard. After optimization of separation conditions, the electrolyte solution was the pH 5.5, 20 mM phosphate buffer containing 4.5 g/L sulfated‐β‐cyclodextrin, 15 kV of electric filed across the capillary applied at 25°C. A diode array detector was used, and the detection wavelength was 260 nm. Under optimum conditions, good linearity was achieved with correlation coefficients from 0.9976 to 0.9997. Detection limits were 0.56 and 0.20 μg/mL, and the quantitation limits were 1.87 and 0.67 μg/mL, respectively. Excellent accuracy and precision were obtained. Recoveries of the analytes varied from 100.8 to 103.1%. The results indicated that baseline separation of analytes was obtained and this method was suitable for quantitative determination of spironolactone in pharmaceutical preparations and rapid simultaneous determination of the content of spironolactone and its major metabolite canrenone in urine samples. 相似文献

Off-target effects remain a significant challenge in the therapeutic use of gapmer antisense oligonucleotides (AONs). Over the years various modifications have been synthesized and incorporated into AONs, however, precise control of RNase H-induced cleavage and target sequence selectivity has yet to be realized. Herein, the synthesis of the uracil and cytosine derivatives of a novel class of 2′-deoxy-2′-fluoro-3′-C-hydroxymethyl-β-d -lyxo-configured nucleotides has been accomplished and the target molecules have been incorporated into AONs. Experiments on exonuclease degradation showed improved nucleolytic stability relative to the unmodified control. Upon the introduction of one or two of the novel 2′-fluoro-3′-C-hydroxymethyl nucleotides as modifications in the gap region of a gapmer AON was associated with efficient RNase H-mediated cleavage of the RNA strand of the corresponding AON:RNA duplex. Notably, a tailored single cleavage event could be engineered depending on the positioning of a single modification. The effect of single mismatched base pairs was scanned along the full gap region demonstrating that the modification enables a remarkable specificity of RNase H cleavage. A cell-based model system was used to demonstrate the potential of gapmer AONs containing the novel modification to mediate gene silencing. 相似文献

A series of 3,3′-(4-arylpyridine-2,6-diyl)bis(2H-chromen-2-one) derivatives have been synthesized by the one-pot, three-component reaction of 3-acetylcoumarin, an aromatic aldehyde, and ammonium acetate in acetic acid under microwave irradiation. This procedure has the major advantages of short reaction time, good yields, low energy consumption, easy operation, and environmental friendliness. Graphene-nanosheet-based highly porous magnetite nanocomposites (GN-HPMNs) have been prepared using a simple solvothermal method and used as an immobilization matrix for the fabrication of a solid-state electrochemiluminescence (ECL) sensor on paper-based chips. Highly porous Fe3O4 nanocrystal clusters were coated with acrylate and wrapped tightly on the skeleton of graphene nanosheets. The structures and sizes of the GN-HPMNs could be tuned by varying the proportions of the solvents ethylene glycol and diethylene glycol. Then, the relatively highly porous ones with an average diameter of about 65 nm were combined with Nafion to form composite films on an electrode surface for immobilization of Ru(bpy)3 2+ (bpy is 2,2′-bipyridine). Because of their porosity, negatively charged surface, and cooperative characteristics of magnetic nanomaterials and graphene, under an external magnetic field, the GN-HPMNs ensured effective immobilization, excellent electron transfer, and long-term stability of Ru(bpy)3 2+ in the composite film. The sensor developed exhibited excellent reproducibility with a relative standard deviation of 0.65 % for 30 continuous cycles. It was found to be much more favorable for detecting compounds containing tertiary amino groups and DNAs with guanine and adenine. A detection limit (signal-to-noise ratio of 3) of 5.0 nM was obtained for tripropylamine. As an application example, 0.5 nM single-nucleotide mismatch could be detected. This was the first attempt to introduce magnetic nanomaterials and an external magnetic field into paper-based chips. A simple, rapid, and efficient oxidation of oximes to the corresponding ketones and aldehydes with manganese dioxide catalyzed by kieselguhr under solvent-free conditions at room temperature in the yields between 82 and 98 is described. It seems that kieselguhr in the present procedure can highly expedite the reaction rate. By comparing to other methods described previously, the main advantages of this oxidation are that the oxidations are more efficient, the reaction conditions are milder, the reaction times are shorter, and the work-up is easier. Depending on their relative orientation, coupled oscillating carbonyl groups provide a VCD spectrum with a characteristic CO bond stretching region showing a strong bisignate VCD feature, which can be readily predicted adopting long available semiempirical methods. The extended coupled oscillator (ECO) formalism has been used to assign the absolute configuration of a recently synthesized chiral 3-substituted isoindolinone.
