A highly sensitive assay for protein with dibromochloro-arsenazo-Al(3+) using resonance light scattering technique and its application

A simple, sensitive and selective method has been developed for the determination of protein using resonance light scattering (RLS) technique. The method is based on the interaction of protein and arsenazo-DBC-Al(3+) in the pH range of 5.0-7.0, which causes a substantial enhancement of the resonance scattering signal of arsenazo-DBC-Al(3+) in the wavelength range of 300-550 nm with the maximum RLS platform at 405-420 nm. With this method, 2.50-50.00 mug ml(-1) of bovine serum albumin (BSA) and 2.50-60.00 mug ml(-1) of human serum albumin (HSA) can be determined, and the detection limits, calculated three times the standard deviation (S.D.) of six blank measurements, for BSA and HSA were 123.4 and 89.6 ng ml(-1), respectively. Moreover, the method is free from interference from many amino acids and metal ions. The method, with high sensitivity, selectivity and reproducibility, was satisfactorily applied to the determination of total protein in human serum samples. Mechanism studies indicated that arsenazo-DBC-Al(3+) could bind to BSA depending mainly on electrostatic forces, which results in enhanced RLS in the arsenazo-DBC-Al(3+)-protein system.     

An improved method for the synthesis of cellulose membrane-bound peptides with free C termini is useful for PDZ domain binding studies

SPOT synthesis permits parallel synthesis and screening of thousands of cellulose membrane-bound peptides to study protein-protein interactions in a proteomic context. Recognition of C-terminal residues is one of the most common binding features of PDZ domains. Unfortunately, most solid support-bound peptide libraries lack a free C terminus due to C-terminal fixation on the solid support. To overcome this restriction, we developed a robust methodology based on our previous strategy for generating peptides with authentic C termini. To validate this improved method, we screened a human peptide library of 6223 C termini with the syntrophin PDZ domain. Furthermore, using the same library, new peptide ligands derived from membrane proteins and receptors were found for the ERBIN PDZ domain. Finally, we identified the protein kinase breakpoint cluster region, which is known as a negative regulator of cell proliferation and oncogenic transformation, as an ERBIN ligand.

在非水溶剂（二甲基亚砜（ＤＭＳＯ）及Ｎ,Ｎ＿二甲基甲酰胺（ＤＭＦ））中的伏安曲线．呈现三对电流峰,表明存在三个可逆或准可逆酞菁配体的单电子转移过程,而在水溶液中则不呈现准可逆行为,且波形改变很大．此外,还比较了四磺酸基酞菁锌的水溶液在自然光和红光（６００～７００ｎｍ）照射下的循环伏安曲线,其氧化还原峰的数目和峰电位基本不变,但在红光照射下比自然光照射下的峰电流明显增大．     

Titanium production in a plasma reactor: A feasibility investigation

The feasibility of producing titanium metal from titanium tetrachloride in a thermal plasma under equilibrium and adiabatic expansion conditions has been theoretically investigated. Free energy minimization and adiabatic expansion calculations to simulate a nozzle expansion were used to study the practicality of production. The crucial requirements for the production of titanium powder from TiCl₄ and H₂ appear to be rapid quenching of the plasma gas at high temperature (e.g., 3700 K) and appropriate reactant concentrations. Quenching of tire plasma gas and production of titanium powder can be achieved by adiabatic expansion through a nozzle. Preliminary experimental data indicate that titanium powder of approximately 5 nm in size can be produced in an argon plasma rising a nozzle expansion approach.     

Synthesis of 5,6,8,9,14,14a-hexahydroisoquino[1,2-b][3]benzazepines

Syntheses of 5,6,8,9,14,14a-hexahydroisoquino[1,2-b][3]benzazepine-2,3,11,12-tetrol and related compounds are described. Key steps involve an initial construction of the isoquinoline ring under the Bischler-Napieralski conditions, which is followed by the building of the azepine ring via an intramolecular lactam formation on to the isoquinoline nucleus.     

5-Br-PADCAP分光光度法测定血清锌

为建立快速、简便、灵敏的分光光度法测定血清锌 ,在表面活性剂TritonX -1 0 0存在下 ,用 2 -(5 -溴 -2 -吡啶偶氮 ) -5 -[N ,N -二羧基甲基 ]苯酚 (5 -Br-PADCAP)作显色剂 ,不去蛋白分光光度法测定血清锌。结果表明 ,锌络合物最大吸收波长为 5 5 7nm ,线性范围 0～ 5 0 0μmol L ,摩尔吸光系数为 1 1 2× 1 0 5L·mol-1·cm-1。回收率为 99 9% ,批内变异系数 (CV)为2 1 % ,批间变异系数为 2 9% ,与原子吸收分光光度法 (x)比较具有良好的相关性 ,y =1 0 1 0x-0 1 5 4,r=0 991 8,P >0 0 5。可见本法测定血清锌不必去蛋白、用血量少、方法简便、灵敏可靠 ,适合临床应用     

Pt/HL和Pt/KL沸石在异丙醇分解反应中的不同催化行为

Ｐｔ／ＫＬ沸石在异丙醇分解反应中具有很高的脱氢活性,而Ｐｔ／ＨＬ沸石具有明显的缺电子性,在此反应中难以显示出脱氢活性,通过加氢作用,可使沸石的酸位免受积炭覆盖,从而可增强Ｐｔ／ＨＬ沸石的酸性催化作用。     

Elucidating dominant factors of PO43-, Cd2+ and nitrobenzene removal by biochar: A comparative investigation based on distinguishable biochars

Four distinct biochars were employed to remove three typical pollutants, meanwhile, path analysis, a multi-statistical regression method, was performed to elucidate the dominant factors of biochar adsorption. This work can provide a new insight to prepare a targeted biochar as adsorbents.     

A new method for inversion of hydroxyl group of carbocyclic nucleosides

The hydroxyl group of carbocyclic nucleosides was inversed when the compounds were treated with Me_3SiCl,KCN and a catalytic amount of NaI in DMF/CH_3CN.     

Synthesis and structure of Li4Sr3Ge2N6: a new quaternary nitride containing Ge2N6 anions

A new Li-containing quaternary nitride, Li₄Sr₃Ge₂N₆, was obtained as single crystals from constituent elements in molten Na. It crystallizes in space group C2/m (No. 12) with a=6.1398(7) Å, b=10.021(1) Å, c=6.3130(7) Å, β=91.279(2)°, and Z=2. It contains the first example of isolated nitridogermanate anions of Ge₂N₆¹⁰⁻, which is also the first example of edge-sharing tetrahedral [GeN₄].
