Persistent deficits in social communication and interaction, and restricted, repetitive patterns of behavior, interests or activities, are the core items characterizing autism spectrum disorder (ASD). Strong inflammation states have been reported to be associated with ASD. The endocannabinoid system (ECS) may be involved in ASD pathophysiology. This complex network of lipid signaling pathways comprises arachidonic acid and 2-arachidonoyl glycerol-derived compounds, their G-protein-coupled receptors (cannabinoid receptors CB1 and CB2) and the associated enzymes. Alterations of the ECS have been reported in both the brain and the immune system of ASD subjects. ASD children show low EC tone as indicated by low blood levels of endocannabinoids. Acetaminophen use has been reported to be associated with an increased risk of ASD. This drug can act through the ECS to produce analgesia. It may be that acetaminophen use in children increases the risk for ASD by interfering with the ECS.This mini-review article summarizes the current knowledge on this topic.  相似文献

Electrochemical processes, which underlie the use of conductive diamond electrodes for the simultaneous detection of two or more metal ions in solution by anodic stripping voltammetry (ASV), have been investigated. The model analyte system studied contains the two metal species, Ag+(aq) and Pb2+(aq), and the experimental techniques employed include cyclic and square wave voltammetries, along with X-ray photoelectron spectroscopy and secondary electron microscopy. Although the bulk metallic forms of Ag and Pb are immiscible, several interactions in the system between the two metal species present are observed, which significantly influence the electrodeposition and electrodissolution processes which underlie ASV. The subsequent nucleation and growth of a given metal on the electrode surface is enhanced by the presence of the second metal on the surface. The encapsulation of one metal by the other, within the metal particulates that form on the electrode surface, significantly reduces the stripping yield at the potentials characteristic of the individual metals. The stripping potentials are also influenced by bonding interactions between deposited Ag and Pb, which broaden the characteristic stripping peaks in cyclic voltammetry, as well as producing underpotential deposition and stripping. Given these interactions, the extent to which ASV at diamond electrodes can be used to determine the solution concentrations of Ag+(aq) and Pb2+(aq) is considered.  相似文献

The miniemulsion polymerization of styrene has been carried out using two pH-responsive cationic diblock macromonomers as reactive stabilizers. As a comparison, the analogous nonpolymerizable cationic diblock copolymer was also investigated. Each of these three stabilizers based on 2-(diethylaminoethyl)methacrylate and quaternized 2-(dimethylaminoethyl)methacrylate residues were prepared via oxyanionic polymerization and had relatively low polydispersities. It was found that all three copolymers were grafted to the polystyrene latex particles, as judged by X-ray photoelectron spectroscopy, aqueous electrophoresis and FTIR spectroscopy studies. Kinetics studies and colloidal characteristics indicated poorer stabilization properties of the partially quaternized diblock macromonomer and electron microscopy confirmed that the latexes invariably had relatively broad particle size distributions.  相似文献

Degeneration of the hippocampus is associated with Alzheimer's disease and occurs very early in the progression of the disease. Current options for treating the cognitive symptoms associated with Alzheimer's are inadequate, giving urgency to the search for novel therapeutic strategies. Pharmacologic agents that safely enhance hippocampal neurogenesis may provide new therapeutic approaches. We discovered the first synthetic molecule, named P7C3, which protects newborn neurons from apoptotic cell death, and thus promotes neurogenesis in mice and rats in the subgranular zone of the hippocampal dentate gyrus, the site of normal neurogenesis in adult mammals. We describe the results of a medicinal chemistry campaign to optimize the potency, toxicity profile, and stability of P7C3. Systematic variation of nearly every position of the lead compound revealed elements conducive toward increases in activity and regions subject to modification. We have discovered compounds that are orally available, nontoxic, stable in mice, rats, and cell culture, and capable of penetrating the blood-brain barrier. The most potent compounds are active at nanomolar concentrations. Finally, we have identified derivatives that may facilitate mode-of-action studies through affinity chromatography or photo-cross-linking.  相似文献

Nanoemulsions were formed spontaneously by diluting water-in-oil (W/O) or brine-in-oil (B/O) microemulsions of a hydrocarbon (octane), anionic surfactant (Aerosol-OT or AOT) and water or NaCl brine in varying levels of excess brine. The water-continuous nanoemulsions were characterized by interfacial tension, dynamic light scattering, electrophoresis, optical microscopy and phase-behavior studies. The mechanism of emulsification was local supersaturation and resulting nucleation of oil during inversion. For nanoemulsions formed at low salinities with Winsor I phase behavior, octane drops grew from initial diameters of 150-250nm to 480-1000nm over 24h, depending on salinity. Growth was caused by mass transfer but seemed to approach the asymptotic stage of Ostwald ripening described by the Lifshitz-Slyozov-Wagner (LSW) theory only for dilution with salt-free water. Near the higher cross-over salinity (Winsor III), the nanoemulsions showed much slower growth with droplet size consistently remaining below 200nm over 24h and reaching 250nm after 1week. Birefringence indicated the presence of liquid crystal for these conditions, which could have contributed to the slow growth rate. At even higher salinity levels in the Winsor II domain, W/O/W multiple emulsions having drops greater than 1μm in diameter were consistently recorded for the first 5-7h, after which size decreased to values below 1μm. The number and size of internal water droplets in multiple emulsion drops was found to decrease over time, suggesting coalescence of internal droplets with the continuous water phase and mass transfer of water from internal droplets to continuous phase as possible mechanisms of the observed drop shrinkage. Electrophoresis studies showed the nanoemulsions to be highly negatively charged (zeta potentials of -60mV to -120mV). The high charge on octane droplets helped assure stability to flocculation and coalescence, thereby allowing mass transfer to control growth in the Winsor I and III regions.  相似文献

Copper-catalyzed conjugate addition of diorgano zinc reagents to nitroacrylate 1 followed by a subsequent nitro-Mannich reaction and in situ lactamization leads to an efficient one-pot synthesis of 1,3,5-trisubstituted 4-nitropyrrolidin-2-ones (5). The versatility of the reaction is shown for a wide range of N-p-(methoxy)phenyl protected aldimines 3 derived from alkyl, aryl, and heteroaryl aldehydes. The densely functionalized pyrrolidin-2-ones 5 are isolated as single diastereoisomers (40 examples, 33-84% yield). An enantioselective copper-catalyzed conjugate addition of diethylzinc led to highly crystalline products that could be recrystallized to enantiopurity in high yield. A range of successful chemoselective transformations were investigated, which widens the applicability of the pyrrolidn-2-ones as stereochemically pure building blocks for further organic synthesis.  相似文献

The proficiency testing (PT) scheme ??AQUA?? for food microbiology was organised by the Istituto Zooprofilattico Sperimentale delle Venezie (IZSVe) according to ISO/IEC 17043 and ISO/TS 22117. This paper describes the IZSVe experience on the application of the above-mentioned standards for the PTs, with focus on the Enterobacteriaceae enumeration one. Freeze-dried food matrices contaminated with American Type Culture Collection bacterial strains were used as test samples for each microbiological PT organised by IZSVe. The sample homogeneity and stability were verified prior to distribution to participants and throughout the PT, respectively. The participating laboratories analysed samples using their routine methods, and results were transmitted to IZSVe. Data and methods used by each participating laboratory were analysed in order to evaluate the laboratory performance. With reference to the Enterobacteriaceae PT, the test samples were homogeneous and stable. In addition, most laboratory results were obtained using equivalent test methods. Statistical approaches applied to analyse data generated from all participating laboratories revealed similar outcomes as no significant outlying count and only 5?% of unacceptable results were observed. Finally, the z-score, with the standard deviation that does not vary from round toround, was applied to compare and to evaluate the performance of each laboratory over time highlighting possible persistent trends over several rounds.  相似文献

The effects of sample preparation and chromatographic method differences on the classification and recovery of metabolic biomarkers from UPLC‐MS measurements on urine samples of humans exposed to different dietary interventions have been investigated. Eight volunteers consumed three high‐fat meals (rich in saturated, monounsaturated and polyunsaturated fatty acids, respectively) in randomized order with a washout period in between. For each participant, urine samples were obtained prior to and at three timed intervals after each meal. Samples were processed either by dilution (1 : 4) or by liquid–liquid extraction and then run under two different gradient conditions. For each analysis method, a total of 96 observations (eight participants, four time points, three diets) were measured. The total ion count chromatograms were analyzed using partial‐least‐squares discriminant analysis. All three dietary classes could be discriminated irrespective of sample preparation and chromatographic method. However, the main discriminating metabolites varied according to sample preparation, indicating that sample treatment and chromatographic conditions influence the ability to extract biomolecular information. Diluted samples showed higher m/z compounds (ca 400 u) while liquid–liquid extraction samples showed low m/z at the same retention time span. Optimized methods for metabolite identification (e.g. organic acids) were statistically inferior to global screening for mixed compound identification, confirming that multiple compound class‐based metabolic profiles are likely to give superior metabonomic (diagnostic) classification, although great care has to be taken in the interpretation in relation to matrix effects. Conversion of macrocyclic imine entities into helical strands was achieved through three‐ and four‐component exchange reactions within constitutionally dynamic libraries. The generation of sequences of the intrinsic helicity codon, based on the hydrazone–pyrimidine fragment obtained by condensation of pyrimidine dialdehyde A with pyrimidine bis‐hydrazine B , shifted the equilibrium between all the possible macrocycles and strands towards the full expression (>98%) of helical product [ A / B ]. Furthermore, it was shown that chain folding accelerated the dynamic exchange reactions among the library members. Lastly, in four‐component experiments (involving A , B , E and either C or D ), even though the macrocyclic entities ([ A / C ], [ B / E ]; [ A / D ], [ B / E ]) were the kinetically preferred products, over time dialdehyde A relinquished its initial diamine partners C or D to opt for bis‐hydrazine B , which allowed the preferential formation of the helically folded strand. The present results indicate that self‐organisation pressure was able to drive the dynamic system towards the selective generation of the strand undergoing helical folding.  相似文献
