A novel, rapid and sensitive LC‐MS/MS method for the determination of 1‐O‐Acetylbritannilactone (ABL), a sesquiterpene lactone abundant in Inula britannica, was developed and validated using heteroclitin D as internal standard. Separation was achieved on a reversed phase Hypersil Gold C18 column (50 × 4.6 mm, i.d., 3.0 µm) using isocratic elution with methanol–5 mM ammonium acetate buffer aqueous solution (80:20, v/v) at a flow rate of 0.4 mL/min. Calibration curve was linear (r > 0.99) in a concentration range of 1.60–800 ng/mL with the lower limit of quantification of 1.60 ng/mL. Intra‐ and inter‐day accuracy and precision were validated by relative error (RE) and relative standard deviation (RSD) values, respectively, which were both less than ±15%. The validated method has been successfully applied to a pharmacokinetic study of ABL in rats. The elimination half‐lives were 0.412 ± 0.068, 0.415 ± 0.092 and 0.453 ± 0.071 h after a single intravenous administration of 0.14, 0.42, and 1.26 mg/kg ABL, respectively. The area under the plasma concentration–time curve from time zero to the last quantifiable time point and from time zero to infinity and the plasma concentrations at 2 min were linearly related to the doses tested. Abstract

Polyammonium macrocycles containing sulfur and furan units in the macrocyclic ring have been synthesized and studied for ATPase activity. The synthetic methodology involved using tosyl protection for the amines and the formation of macrocyclic Lactams, followed by reduction using borane in THF. Deprotection of the tosylated forms of the macrocycle was accomplished using sodium in butanol for the furan macrocycles, and HBr in HOAc for the sulfur containing macrocycle. The macrocycles were found to be poor catalysts for ATP hydrolysis compared to other similar polyammonium macrocycles.  相似文献

本文以环状单萜醇α-萜品醇、L-薄荷醇及链状单萜醇香叶醇、香茅醇为先导化合物,采用酰氯酯化法合成肉豆蔻酸α-萜品醇酯(TER-C14)、肉豆蔻酸-L-薄荷醇酯(MEN-C14)、肉豆蔻酸香叶醇酯(GER-C14)和肉豆蔻酸香茅醇酯(CIT-C14),并考察单萜醇及其肉豆蔻酯作为促透剂对布南色林(Blo)的促透效果。通过体外经皮渗透实验、体外释放实验和分子模拟技术初步探究单萜醇及其肉豆蔻酯的促透机制。结果显示,当GER-C14或CIT-C14为促透剂时,均有显著的促透效果(P<0.05),并且24h经皮累积透过量是空白组的4.84倍和4.45倍。促透机制为肉豆蔻酸单萜醇酯破坏药物与神经酰胺之间的氢键相互作用,增加脂质迁移率和药物的自由能,从而促进药物的渗透。肉豆蔻酸单萜醇酯有望作为新型促透剂在经皮给药系统中广泛应用。  相似文献

土壤基质复杂,土壤中残留的抗生素种类繁多,浓度多为痕量水平,高灵敏度的仪器方法、有效的净化和富集方法、多种类抗生素的同时检测是土壤中抗生素检测的重点和难点。该研究建立了固相萃取-超高效液相色谱-串联质谱法同时测定土壤中7类(磺胺类、氟喹诺酮类、四环素类、大环内酯类、β-内酰胺类、酰胺醇类和林可酰胺类)30种抗生素的方法。首先,通过参数优化确定最佳质谱条件,选择BEH-C18色谱柱,以0.1%(v/v)甲酸甲醇溶液-0.1%(v/v)甲酸水溶液为流动相,10%(v/v)甲醇水溶液为进样溶剂。然后,通过提取条件(萃取剂种类及体积)和固相萃取条件(上样液pH、淋洗液有机溶剂比例、洗脱液种类及体积)的优化,确定使用10 mL乙腈和Na2EDTA-McIlvaine缓冲液的混合溶液(1∶1, v/v)为萃取剂,萃取液pH调节至8.0后,采用HLB小柱进行固相萃取,并以10 mL超纯水淋洗净化,最后用10 mL甲醇-乙腈(1∶1, v/v)洗脱目标分析物。在优化的分析条件下,该方法的定量限为0.043~4.04 μg/kg,目标化合物的标准曲线线性关系良好,相关系数在0.992~1.00的范围内,在20、100、200 μg/kg的添加浓度下,大多数目标化合物的加标回收率范围为44.8%~164%,相对标准偏差为0.700%~14.8%。将该方法用于6个实际土壤样品的分析,结果显示在30种抗生素中,17种抗生素有检出,其中12种抗生素的检出率为100%。环丙沙星和诺氟沙星是土壤样品中含量最高的两种抗生素,它们的含量范围分别是13.7~32.1和15.6~43.6 μg/kg。本研究建立的方法简单、快速、溶剂使用量少,能用于土壤样品中痕量水平的7类30种抗生素的同时测定。  相似文献

A facile microwave-assisted ethylene glycol method is developed to synthesize the SnO2 nanoparticles dispersed on or encapsulated in reduced graphene oxide (SnO2-rGO) hybrids. The morphology, structure, and composition of SnO2-rGO are investigated by scanning electron microscopy, transmission electron microscope, thermo-gravimetric analyzer, X-ray diffraction, Raman spectroscopy, and X-ray photoelectron spectroscopy. The electrochemical performance of SnO2-rGO as anode materials for lithium-ion batteries was tested by cyclic voltammetry, galvanostatic charge–discharge cycling, and rate capability test. It is found that the SnO2 nanoparticles with a uniform distribution have p-type doping effect with rGO nanosheets. The as-prepared SnO2-rGO hybrids exhibit remarkable lithium storage capacity and cycling stability, and the possible mechanism involved is also discussed. Their capacity is 1222 mAhg?1 in the first cycle and maintains at 700 mAhg?1 after 100 cycles. This good performance can be mainly attributed to the unique nanostructure, good structure stability, more space for volume expansion of SnO2, and mass transfer of Li+ during cycling.  相似文献

The Fourier transform infrared spectroscopy and Fourier transform Raman spectra of phenylacetyl chloride were recorded and analyzed in the range 3500–400 and 3500–200 cm?1 at room temperature, respectively. In order to obtain the structural information and conformational stabilities, a potential energy surface scan for internal rotation was carried out at the B3LYP/6‐31G(d) level. The potential energy surface reveals that the title compound has two minimal conformers (A and B). The optimized geometries, structural parameters, stabilities, energies, thermodynamic parameters, vibrational wavenumbers, infrared intensities, and Raman activities for the two conformers (A and B) have been obtained by employing B3LYP and MP2 calculations with 6‐311++G (d, p) basis sets. The conformational energy difference between A and B is very small, indicating that the B conformer coexists with the A conformer. The detailed vibrational assignments of vibrational spectra of each conformer have been made on the basis of the potential energy distributions analysis. The highest occupied molecular orbital –lowest unoccupied molecular orbital energy gap and molecular electrostatic potential of the two conformers have been also calculated for comparison of their chemical activities. Large amount of work has been published on the isotacticity–properties relationship of isotactic polypropylene (iPP). However, the stereo‐defect distribution dependence of morphology and mechanical properties of iPP injection molding samples is still not clear. In this study, two different isotactic polypropylene (iPP) resins (PP‐A and PP‐B) with similar average isotacticity but different stereo‐defect distribution were selected to investigate the morphology evolution and mechanical properties (tensile and notching) of their injection molding samples using differential scanning calorimetry (DSC), dynamic mechanical analysis (DMA), 2D wide angle X‐ray diffraction (2D‐WAXD), and scanning electron microscope (SEM). The results of DMA showed that the molecular movement ability of PP‐A (with less uniform distribution of stereo‐defect) was stronger than that of PP‐B, meanwhile the analysis of DSC and SEM suggested that after injection molding, smaller spherullites, and crystals with higher perfection had formed in the specimens of PP‐A. The resulting of tensile properties of PP‐A were found to be better than that of PP‐B. The results of morphology evolution by SEM observation and 2D‐WAXD showed that PP‐A is more likely to occur interspherulite deformation and can disperse the tensile stress more efficiently, and therefore, its crystal structure can withstand a greater force when tensile stress is applied. On the other hand, PP‐B has larger spherulites and boundaries, and low perfection of lamellaes, and the intraspherulte deformation tend to take place. 针对种晶的表面粗糙度和边部形态对MPCVD法生长金刚石单晶的影响进行了研究。结果表明,当样品表面粗糙度Ra值达到0.0066μm时,单晶金刚石沉积层已经可以呈现出较高的结晶质量。当表面粗糙度Ra值达到0.0162μm后,种晶的中心区域受到的影响较小,但种晶边缘区域的沉积却受到了较明显的影响。研究边部形态的实验中,在同一种晶的不同区域抛磨出45°边棱和90°边棱,生长后分别对这两个区域进行了拉曼光谱测试,测试结果表明,90°边棱处1332 cm-1金刚石本征峰的半高宽较小,沉积层质量较好,初步推测90°是更适合的种晶边棱角度。  相似文献

The direct formic acid fuel cell is an emerging energy conversion device for which palladium is considered as the state‐of‐the‐art anode catalyst. In this communication, we show that the activity and stability of palladium for formic acid oxidation can be significantly enhanced using nickel phosphide (Ni2P) nanoparticles as a cocatalyst. X‐ray photoelectron spectroscopy (XPS) reveals a strong electronic interaction between Ni2P and Pd. A direct formic acid fuel cell incorporating the best Pd–Ni2P anode catalyst exhibits a power density of 550 mW cm?2, which is 3.5 times of that of an analogous device using a commercial Pd anode catalyst.  相似文献
