The growth of calcite in an aqueous gel of was studied and the appearance of the crystals was found to change over time. Crystals removed from the gel at progressively longer times showed severely affected surfaces resulting from dissolution. If crystals were removed from the gel after 3.5 hours, at which point there were no etch pits, and then placed in either buffer or pure water, etch pits, similar to those observed on crystals that are left in the gel, were observed. Control calcite crystals exposed to similar conditions (water or buffer) show no significant dissolution after equivalent times. A probable cause of the altered dissolution is the non-specific occlusion of gelator aggregates at sites of imperfection. The gel appears to provide a microenvironment in which the molecules that form the matrix also participate in the crystallization. This system allows the study of the unique properties of a gel for influencing the nucleation and growth of inorganic crystals, some of which may be important for better understanding biomineralization. 相似文献

We have compared the photodynamic activities of hematoporphyrin (HP) and protoporphyrin (PP) on isolated rat liver mitochondria by measuring the decline of the respiratory control ratio (RCR) after irradiation at 365 nm. Before addition to the respiratory mcdium, the dyes were dissolved in phosphate-buffered saline (PBS) or incorporated into unilamellar liposomes of dipalmitoyl-phosphatidylcholine (DPPC), sometimes enriched with cholesterol (Chol) or cardiolipin (Card), which are naturally present in mitochondrial membranes. Chol and especially Card strongly increase the porphyrin uptake by mitochondria. In all experimental conditions, PP is taken up by mitochondria to a higher extent than HP. Nevertheless, under conditions giving the same amount of mitochondriabound dye, HP is a morc efficient photosensitizer than PP. As the efficiency of singlet oxygen production has been shown to be equivalent for the two porphyrins in monomeric state, the resulting photobiological effects are explained in terms of different localization of HP and PP in the mitochondrial membrancs. In particular, HP preferentially localizes in the protein-rich polar domains of the inner mitochondrial membrane, whereas PP dissolvcs in the lipid regions of the mcmbrancs. 相似文献

Abstract The 83 μM hematoporphyrin (HP)-sensitized photooxidation of 0.1 mM tryptophan in aqueous solution buffered at pH 7.4 or in binary mixtures of phosphate buffer and organic solvents of higher (formamide) or lower (N,N-dimethylformamide, methanol, ethanol, tetrahydrofuran) polarity proceeds by a pure singlet oxygen (1O2) mechanism as suggested by azide quenching experiments, the rate-enhancing action of deuterated solvents, and the lack of any significant reaction between triplet HP and tryptophan. Both the first-order rate constant of the photoprocess and the photooxidation quantum yield (φ= 0.011 in phosphate buffer at pH 7.4) increase when the medium polarity is increased (e.g. φ= 0.024 in 90% formamide); this results mainly from the greater quantum yield of 1O2 generation and the longer lifetime of 1O2. The intrinsic reactivity of 1O2 with tryptophan is independent of formamide concentration. A moderate decrease in the medium polarity (e.g. in the range 0-30% methanol) enhances the efficiency of tryptophan photooxidation (φ= 0.014 in 30% methanol) as a result of the enhanced quantum yields of triplet HP and 1O2 formation. In contrast, the overall photooxidation rate is depressed at high concentrations of low-polarity organic solvents (e.g. φ= 0.0039 in 90% methanol) due to a 5.5-fold drop of the rate constant for the 1O2-tryptophan reaction which counteracts the enhancement of the lifetime and quantum yield of triplet HP and 1O2. The solvent composition also affects the equilibria between monomeric and multimeric forms of HP. However, under our experimental conditions, the aggregation state of HP appears to exert only a minor influence on the efficiency of tryptophan photooxidation. 相似文献

Abstract— Studies on the synthesis, preliminary in vivo biological activity, singlet oxygen and fluorescence yields of a di-meric porphyrin (Dl) are described. The pharmacokinetic behavior and photodynamic properties of the dimer Dl were examined in Balb/c mice bearing an MS-2 fibrosarcoma. Compound Dl shows a high selectivity for tumor localization (tumor/peritumoral tissue ratios of dye concentration ranging between ca 100 and 140 at 24 h after drug administration of 5.0-1.0 mg kg-1 into DL-AL-dipalmitoylphosphatidylcholine liposomes). The photo-therapeutic efficiency of dimer Dl was evaluated by following the growth curves of fibrosarcoma irradiated with red light (600-700 nm) with a total dose of 400 J cm-2 , at 24 h after intravenous injection. Photodynamic therapy-treated tumors showed a significant delay in growth as compared to untreated control mice. A Si(IV)-phthalocyanine bearing two methoxyethyleneglycol axial ligands bound to the central metal ion (SiPc) has been prepared by chemical synthesis and analyzed for its phototherapeutic activity after administration in a Cremophor or liposome formulation to C57B1/6 mice bearing a subcutaneously transplanted Lewis lung carcinoma (LLC). The maximum drug accumulation in the tumor is found at 24 h after intraperitoneal injection, independent of the delivery system. However, the tumor concentration of SiPc in the Cremophor formulation is about two-fold higher, while the drug concentration in liver and skin shows similar trends with the two delivery systems. The drug accumulation and retention in the brain is much larger when using Cremophor emulsion. Photodynamic therapy (672 nm, 370 mW m−2, 360 J cm−2) at 24 h after the injection of Cremophor emulsion- or DPPC liposome-formulated SiPc causes a very efficient and similar response for the LLC (8 versus 22 mm mean tumor diameter for the control groups at 21 days after phototreatment). These very promising effects, obtained both at higher and lower tumor drug concentrations, clearly demonstrate the potential phototherapeutical activity of the newly synthesized SiPc. 相似文献
