In some applications, the purification and recovery of biomolecules is performed via a cascade of batch adsorption and desorption stages using agitated contactors and related filtration devices. Suspended bed chromatography is a recent process-scale innovation that is applicable to these separations. This hybrid technique exploits the benefits of combining batch adsorption in an agitated contactor with elution in an enclosed column system. To some extent, the process is similar to batch contactor chromatography but can be fully contained and significantly quicker. The process has two steps; first the fluid containing the sample is mixed with the adsorbent in a stirred tank. Second, the slurry suspension is transferred directly into a specialized column, such as an IsoPak column. The media with the adsorbed product is formed as a packed bed, whilst the suspension liquid is passed out of the column. The product is then eluted from the packed bed utilizing standard column-chromatography techniques. The performance of the suspended bed and the agitated contactor operations are demonstrated both by full-scale experimental results and process simulations. The purification of ovalbumin from a hen-egg white feedstock by anion-exchange chromatography was used as a case study in order to prove the concept. With the availability of both pump-packed systems and shear-resistant media, suspended bed chromatography is a better alternative for a range of applications than the traditional batch separations using agitated contactors.  相似文献

The S.C.M.O. methods has been employed to study the valence electronic structures of some isomers of the methylboron and methylaluminium hydrides. Basis sets including 3s, 3p and diffuse and contracted 3d orbitals were used for the aluminium atoms. For both boron and aluminium compounds hydrogen is energetically favoured over methyl as a bridging group. This stems mainly from the different nuclear repulsion energies of the isomers. Participation of the 3d orbitals is quite marked, particularly in the metal-metal cross-ring interactions.
Zusammenfassung Mittels der SCMO Methode wurde die Valenzelektronenstruktur einiger Isomere der Methylborund Methylaluminiumhydride untersucht. Die Basis enthielt für Aluminiumatome 3s, 3p und kontrahierte und nicht kontrahierte 3d Zustände. Sowohl bei den Bor- als auch den Aluminiumverbindungen sind Wasserstoffatome als Brücke gegenüber Methylgruppen energetisch begünstigt. Das rührt hauptsächlich von den unterschiedlichen Kernabstoßungsenergien her. Die Beteiligung der 3d Orbitale ist deutlich, besonders in der (transannularen) Metall-Wechselwirkung.

Résumé La méthode S.C.M.O. a été utilisée pour étudier les structures électroniques de valence de certains isomères des hydrures de méthylbore et de methylaluminium. Pour les atomes d'aluminium on a utilisé des bases contenant des orbitales 3s, 3p et des orbitales 3d diffuses et contractées. Tant pour les composés du bore que ceux de l'aluminium l'hydrogène est favorisé énergétiquement par rapport au méthyl en tant que groupement de pont. Ceci provient essentiellement des différences d'énergie de répulsion nucléaire. La participation des orbitales 3d est nettement marquée, particulièrement dans les interactions métal-métal à travers le cycle.

A rapid electrochemical method based on using a clean hydrogen‐bubble template to form a bimetallic porous honeycomb Cu/Pd structure has been investigated. The addition of palladium salt to a copper‐plating bath under conditions of vigorous hydrogen evolution was found to influence the pore size and bulk concentration of copper and palladium in the honeycomb bimetallic structure. The surface was characterised by X‐ray photoelectron spectroscopy, which revealed that the surface of honeycomb Cu/Pd was found to be rich with a Cu/Pd alloy. The inclusion of palladium in the bimetallic structure not only influenced the pore size, but also modified the dendritic nature of the internal wall structure of the parent copper material into small nanometre‐sized crystallites. The chemical composition of the bimetallic structure and substantial morphology changes were found to significantly influence the surface‐enhanced Raman spectroscopic response for immobilised rhodamine B and the hydrogen‐evolution reaction. The ability to create free‐standing films of this honeycomb material may also have many advantages in the areas of gas‐ and liquid‐phase heterogeneous catalysis.  相似文献

A synthesis of methyl 2-hydroxy-4-oxo-4-(substituted phenyl)butanoates has been accomplished using Friedel-Crafts acylation of substituted benzenes with 2-acetoxybutanoyl chloride.The key feature of this strategy is that various 4-arylbutanoates can be obtained conveniently,which are important intermediates for the synthesis of biologically active compounds including ACE inhibitors.  相似文献
