The exchange of subunits between oligomer protein particles depends upon a cycle of dissociations and associations. To examine the dynamics of these cycles we have employed two methods based on the transfer of excitation energy between fluorochromes attached to different subunits of protein oligomers, at various temperatures and pressures. In the heterotransfer method, identical solutions independently labeled with two different fluorophores, donor D and acceptor A, are mixed. The fluorescence spectrum permits the determination of the subunit exchange by the increase in A and decrease in D fluorescence as mixed AD oligomers are formed. In the homotransfer method the aggregates are labeled with fluorescein to the extent that, ideally, each subunit carries a fluorophore. The emission is strongly depolarized because sufficiently often it takes place after a transfer to a fluorophore oriented differently from the one originally excited. Both dissociation and subunit exchange with unlabeled material result in an increase in polarization and can be independently determined by the homotransfer method. Both homo- and heterotransfer have been employed in the study of the effect of temperature on the stability of the aggregates and the relation between the rate of dissociation and the rate of exchange when dissociation of oligomers is induced by hydrostatic pressure. 相似文献

Characterizing and controlling the interlayer orientations and stacking orders of two‐dimensional (2D) bilayer crystals and van der Waals (vdW) heterostructures is crucial to optimize their electrical and optoelectronic properties. The four polymorphs of layered gallium selenide (GaSe) crystals that result from different layer stackings provide an ideal platform to study the stacking configurations in 2D bilayer crystals. Through a controllable vapor‐phase deposition method, bilayer GaSe crystals were selectively grown and their two preferred 0° or 60° interlayer rotations were investigated. The commensurate stacking configurations (AA′ and AB stacking) in as‐grown bilayer GaSe crystals are clearly observed at the atomic scale, and the Ga‐terminated edge structure was identified using scanning transmission electron microscopy. Theoretical analysis reveals that the energies of the interlayer coupling are responsible for the preferred orientations among the bilayer GaSe crystals. 相似文献

An evaluation of the gas-phase ion chemistry of rotenone (1) by electrospray ionisation (ESI) mass spectrometry (MS) and tandem mass spectrometry (MS2) is presented, aiming at providing tools for its determination in natural and biological matrices. The behaviour of its cycloadducts with benzonitrile-N-oxide (2) and 2,4,6-trimethylbenzonitrile-N-oxide (3) was also evaluated and the MS data thus obtained have provided evidence into the mechanism of formation of the key product ion at m/z 192 which can be considered a marker in the MS and MS2 spectra of rotenone and its derivatives. 相似文献

Trace level vanadium determination is reported using a dual beam thermal lens spectrometer. The thermal lens was generated using an argon ion beam laser (pump beam) which was focused into a sample cuvette. The thermal lens signal (TLs) was monitored with a He-Ne laser beam and a photodiode detector. Multichannel averager software was developed for processing the transient TLs. The optimal set up, ensuring maximum sensitivity and linear calibration graphs was obtained using experimental design techniques. Under optimized conditions, the detection limits for aqueous and ethanol-water (2+3 v/v) and (4+1 v/v) vanadium complex solutions were, respectively, 0.0071 mg/l, 0.0065 and 0.0039 mg/l. 相似文献

These compounds perform a good tension resistance to the impact and the humidity absorption being used at the present time and also are in the automobile industry in parts underneath the hood, especially in the radiator frames. The aim of this work is to study the effect of ionizing radiation on the properties of polyamide 6 with fiberglass reinforcement and undergone to different irradiation doses. Samples were prepared and irradiated on JOB 188 accelerator with an electron beam energy of 1.5 MeV in air with different doses and a dose rate of 27.99 kGy/h. Photodegradation of organic compounds in water solutions by means of economically advantageous and environment-friendly processes is a topic of growing interest. In recent years a great attention has been devoted to TiO2-based photocatalysts for the oxidative degradation of various organic pollutants. In this context, we have prepared new photo-catalytic polycrystalline TiO2 systems impregnated with sensitizers, i.e., copper, iron or manganese porphyrins, and investigated their photoactivity for 4-nitrophenol oxidation compared with that of bare TiO2. A significant improvement of the photoreactivity was observed in the case of TiO2 impregnated with copper porphyrin, while only a slight beneficial effect was observed in the case of iron porphyrin. Modeling of extreme values in the presence of heterogeneity is still a relatively unexplored area. We consider losses pertaining to several related categories. For each category, we view exceedances over a given threshold as generated by a Poisson process whose intensity is regulated by a specific location, shape and scale parameter. Using a Bayesian approach, we develop a hierarchical mixture prior, with an unknown number of components, for each of the above parameters. Computations are performed using Reversible Jump MCMC. Our model accounts for possible grouping effects and takes advantage of the similarity across categories, both for estimation and prediction purposes. Some guidance on the specification of the prior distribution is provided, together with an assessment of inferential robustness. The method is illustrated throughout using a data set on large claims against a well-known insurance company over a 15-year period. 相似文献

In this work we study the period function T of solutions to the conservative equation x″(t)+f(x(t))=0. We present conditions on f that imply the monotonicity and convexity of T. As a consequence we obtain the criterium established by C. Chicone and find conditions easier to apply. We also get a condition obtained by Cima, Gasull and Mañosas about monotonicity and, following some of their calculations, present results on the period function of Hamiltonian systems where H(x,y)=F(x)+n-1|y|n.
