The nitrogenase MoFe protein contains two different FeS centers, the P-cluster and the iron–molybdenum cofactor (FeMo-co). The former is a [Fe8S7] center responsible for conveying electrons to the latter, a [MoFe7S9C-(R)-homocitrate] species, where N2 reduction takes place. NifB is arguably the key enzyme in FeMo-co assembly as it catalyzes the fusion of two [Fe4S4] clusters and the insertion of carbide and sulfide ions to build NifB-co, a [Fe8S9C] precursor to FeMo-co. Recently, two crystal structures of NifB proteins were reported, one containing two out of three [Fe4S4] clusters coordinated by the protein which is likely to correspond to an early stage of the reaction mechanism. The other one was fully complemented with the three [Fe4S4] clusters (RS, K1 and K2), but was obtained at lower resolution and a satisfactory model was not obtained. Here we report improved processing of this crystallographic data. At odds with what was previously reported, this structure contains a unique [Fe8S8] cluster, likely to be a NifB-co precursor resulting from the fusion of K1- and K2-clusters. Strikingly, this new [Fe8S8] cluster has both a structure and coordination sphere geometry reminiscent of the fully reduced P-cluster (PN-state) with an additional μ2-bridging sulfide ion pointing toward the RS cluster. Comparison of available NifB structures further unveils the plasticity of this protein and suggests how ligand reorganization would accommodate cluster loading and fusion in the time-course of NifB-co synthesis.

The K-cluster of NifB as a key intermediate in the synthesis of the nitrogenase active site supports [Fe4S4] cluster fusion occurs before carbide and sulfide insertion and displays ligand spatial arrangement reminiscent to that of the P-cluster.  相似文献

Effects of high-pH environments on a stationary phase prepared by gamma-radiation immobilization of poly(methyloctylsiloxane) on titanium-grafted silica were investigated by HPLC testing with standard sample mixtures. The HPLC parameters indicate good stationary phase stability to 10000 column volumes each of mobile phases with pH of 7, 9 and 12. At pH 13, the efficiency decreases slowly, although reasonably good separations are still possible until increasing flow resistance no longer allows easy passage of the mobile phase.  相似文献

In this work, a flow injection system with spectrophotometric detection was developed for the determination of lithium in pharmaceutical formulations used in the treatment of bipolar disorder. Reaction between Quinizarine (1,4-dihydroxyanthraquinone) and Li(I) ion in alkaline medium containing dimethylsulfoxide (DMSO) was explored for this purpose. The flow system was optimized regarding to its chemical (DMSO, Quinizarine and NaOH concentrations and sample pH) and physical parameters (sample loop volume, carrier flow rate and reactor length) in order to establish better conditions in terms of sensitivity and sampling frequency. The results obtained showed that the concentration of DMSO in the reagent solution presents remarkable influence on the magnitude of analytical signal. Chemical species that could be found in the formulations such as Na(I), K(I), Mg(II), Ca(II), Ti(IV), Cl, CO32− e sodium dodecylsulfate were tested as possible interfering ions. Among them, only non-monovalent cations presented noticeable interference on lithium signal. However, they were not found in concentrations high enough to cause interference in the determination of lithium in the samples. Sample preparation was performed by sonicating a slurry prepared by dispersing 100 mg of powdered sample in 15 mL of 0.10 mol L−1 HCl solution. Results obtained by developed methodology were not statistically different from those obtained by flame emission spectrometry. In the optimized conditions the method presented a linear range of 5-40 mg L−1 and a relative standard deviation of 3.6% at 5 mg L−1 Li concentration. Detection and quantification limits were 0.54 and 1.8 mg L−1, respectively. Sampling frequency, calculated as the time interval passed between two consecutive injections, was 60 samples per hour. The methodology was successfully applied in the determination of lithium in three commercial samples.  相似文献

In this work we report the experimental studies of Fe?Mn?Al alloys in the FCC disordered phase at room temperature by Mössbauer spectroscopy and X-ray diffraction. In this phase the alloys are antiferromagnetic with a constant mean hyperfine field ( \(\bar H\) ) near 26 kOe in the composition range from 0 to 7.5 at.% Al and 50 to 65 at.% Fe. When the Al or Fe concentration increases, the \(\bar H\) value gradually decreases to zero and the alloy becomes paramagnetic. In the same way when the Al concentration increases the lattice parameter increases linearly but when the Fe concentration increases the lattice parameter remains nearly constant for alloys with 5 at.% Al and decreases for alloys with 10 at.% Al.  相似文献

Consideration is given to the effect on gel-permeation chromatographic (GPC) data of the extent of substitution in nitrated cellulose. GPC parameters for samples containing 13.55–13.81% nitrogen (14.14% corresponds to complete substitution, DS = 3) were hardly affected by this variation in substitution. Variations that were observed are considered to arise within the samples themselves. Experiments with low molecular weight organic iodides, nitrates, and hydroxyl compounds indicate longer chain lengths than actual; this is attributed to extensive solvation of the substituent groups. The very long chain lengths obtained for cellulose nitrate by the present GPC procedure may arise from such an affect.  相似文献

Summary The Figueira coal-fired power plant (CFPP) is among the Brazilian CFPP which presents higher uranium concentration. Gamma-ray spectrometry was used to determine 238U, 226Ra, 210Pb, 232 Th and 40K contents in pulverized coal, furnace bottom ash and fly ash samples. The natural radionuclide concentrations in pulverized coal ranged from 813 to 2609 Bq . kg-1 for U series and from 22 to 40 Bq . kg-1 for 232 Th. The fly ash fraction gave concentrations ranging from 1442 to 14641 Bq . kg-1, for uranium series. The same enrichment factor was observed for 238U, 226Ra and 232 Th. Only 210Pb and stable Pb presented a high enrichment factor for the last stage filter fly ash. The concentration of the uranium series found in the ashes is close to the limit adopted by the Brazilian guideline (CNEN-NN-4.01).22 Therefore, it is advisable to evaluate the environmental impact of the installation.  相似文献

A kinetic method is presented for the determination of 0.5–5 μg ml?1 gallium based on its activating effect on the copper(II)-catalyzed oxidation of 4,4′-dihydroxybenzophenone thiosemicarbazone by hydrogen peroxide. The reaction is monitored spectrophotometrically at 415 nm. Two sets of reaction conditions are established; one for the direct determination of gallium, and another, in which indium affects the gallium response, for determination of indium. Mixtures of these cations can be determined at μg ml?1 levels and in gallium/indium ratios from 7.5:1 to 1:1.6, with an accuracy and precision of ca. 4.5%.  相似文献

Biomineral formation is a common trait and prominent for soil Actinobacteria, including the genus Streptomyces. We investigated the formation of nickel-containing biominerals in the presence of a heavy-metal-resistant Streptomyces mirabilis P16B-1. Biomineralization was found to occur both in solid and liquid media. Minerals were identified with Raman spectroscopy and TEM-EDX to be either Mg-containing struvite produced in media containing no nickel, or Ni-struvite where Ni replaces the Mg when nickel was present in sufficient concentrations in the media. The precipitation of Ni-struvite reduced the concentration of nickel available in the medium. Therefore, Ni-struvite precipitation is an efficient mechanism for tolerance to nickel. We discuss the contribution of a plasmid-encoded nickel efflux transporter in aiding biomineralization. In the elevated local concentrations of Ni surrounding the cells carrying this plasmid, more biominerals occurred supporting this point of view. The biominerals formed have been quantified, showing that the conditions of growth do influence mineralization. This control is also visible in differences observed to biosynthetically synthesized Ni-struvites, including the use of sterile-filtered culture supernatant. The use of the wildtype S. mirabilis P16B-1 and its plasmid-free derivative, as well as a metal-sensitive recipient, S. lividans, and the same transformed with the plasmid, allowed us to access genetic factors involved in this partial control of biomineral formation.  相似文献

The aim of this study is to evaluate the phytochemical profile, oral acute toxicity, and the effect of ylang-ylang (Cananga odorata Hook. F. & Thomson) essential oil (YEO) on acute inflammation. YEO was analyzed by gas chromatography/mass spectrometry. For in vitro tests, YEO was assessed using cytotoxicity, neutrophil chemotaxis induced by N-formyl methionyl leucyl phenylalanine (fMLP), and phagocytic activity tests. YEO was orally administered in zymosan-induced peritonitis, carrageenan-induced leukocyte rolling, and adhesion events in the in situ microcirculation model and in carrageenan-induced paw edema models. YEO (2000 mg/kg) was also tested using an acute toxicity test in Swiss mice. YEO showed a predominance of benzyl acetate, linalool, benzyl benzoate, and methyl benzoate. YEO did not present in vitro cytotoxicity. YEO reduced the in vitro neutrophil chemotaxis induced by fMLP and reduced the phagocytic activity. The oral treatment with YEO reduced the leukocyte recruitment and nitric oxide production in the zymosan-induced peritonitis model, reduced rolling and adherent leukocyte number induced by carrageenan in the in situ microcirculation model, and reduced carrageenan-induced edema and mechanical hyperalgesia. YEO did not present signs of toxicity in the acute toxicity test. In conclusion, YEO affected the leukocyte activation, and presented antiedematogenic, anti-hyperalgesic, and anti-inflammatory properties.  相似文献
