This study investigated the relaxation behaviors of octadecylamine (ODA), stearic acid (SA), and SA/ODA mixed monolayers at the air/water interface. Area relaxations of monolayers at constant surface pressure were studied by a nucleation and growth mechanism and by direct observation using a Brewster angle microscope (BAM). The results showed that ODA and SA monolayers exhibit different characteristics in the area loss and in the BAM morphology. In the initial relaxation stage, SA monolayer illustrates a more stable characteristic than ODA. But at the later stage, the area loss of SA monolayer increases more quickly than that for ODA due to significant nucleation and growth of 3D aggregates. The BAM results demonstrated that 3D aggregates of large scale domains are likely to form on a SA monolayer even when the area loss is insignificant. On the contrary, only dotlike aggregates of low density were found on the ODA monolayer when relaxation is carried out at higher surface pressure. The relaxation behavior of SA monolayer can be described by the Vollhardt model. However, the relaxation of ODA monolayer does not follow the nucleation model described by Vollhard but can reasonably be attributed to the effect of dissolution. For the SA/ODA mixed monolayers, the relaxation behaviors in the initial and final stages follow different mechanisms, which is attributed to the formation of distinct phases as observed from the BAM. This result also implied that SA and ODA are not completely miscible to be a homogeneous phase. Phases of various compositions were formed in the mixed monolayers, and thus, the relaxation mechanism was shifted during the relaxation process as dominated by different relaxation behaviors of various phases.  相似文献

Methyl 3,4‐di‐(2′‐hydroxyethoxy)benzylidenecyanoacetate ( 3 ) was prepared by hydrolysis of methyl 3,4‐di‐(2′‐vinyloxyethoxy)benzylidenecyanoacetate ( 2 ). Diol 3 was condensed with 2,4‐toluenediisocyanate, 3,3′‐dimethoxy‐4,4′‐biphenylenediisocyanate, and 1,6‐hexamethylenediisocyanate to yield polyurethanes 4 – 6 containing the nonlinear optical chromophore 3,4‐dioxybenzylidenecyanoacetate. The resulting polyurethanes 4 – 6 were soluble in common organic solvents such as acetone and dimethylformamide. Polymers 4 – 6 indicated thermal stability up to 300 °C in thermogravimetric thermograms with glass‐transition temperature values obtained from differential scanning calorimetric thermograms in the range of 78–102 °C. Copolymers of 2,2,6,6-tetramethylpiperidinyl methacrylate (TPMA) with styrene (S) and with methyl methacrylate (MMA) were synthesized using AIBN as initiator. S–TPMA copolymers from feed ranging from 0.10–0.80 mole fractions TPMA and MMA-TPMA copolymers from feed of 0.04–0.85 mole fractions TPMA were used in the determination of monomer reactivity ratios r1, r2. Four different methods were employed in the calculations of r1 and r2 and all calculated results were in good agreement with each other. The structure of S–TPMA copolymers was inferred to be of an alternating nature while that of MMA–TPMA copolymers was random. Both copolymers are potential hindered amine light stabilizers (HALS) and are expected to be less extractable from, and more compatible with, polystyrene and poly(methyl methacrylate) base polymers.  相似文献

We describe the application of capillary liquid chromatography (LC) time-of-flight (TOF) mass spectrometric instrumentation for the rapid characterization of microbial proteomes. Previously (Lipton et al., Proc. Natl. Acad. Sci. U.S.A. 2002, 99, 11049) the peptides from a series of growth conditions of Deinococcus radiodurans have been characterized using capillary LC MS/MS and accurate mass measurements which are captured as an accurate mass and time (AMT) tag database. Using this AMT tag database, detected peptides can be assigned using measurements obtained on a TOF due to the additional use of elution time data as a constraint. When peptide matches are obtained using AMT tags (i.e., using both constraints) unique matches of a mass spectral peak occurs 88% of the time. Not only are AMT tag matches unique in most cases, the coverage of the proteome is high; approximately 3500 unique peptide AMT tags are found on average per capillary LC run. From the results of the AMT tag database search, approximately 900 ORFs detected using LC-TOFMS, with approximately 500 ORFs covered by at least two AMT tags. These results indicate that AMT database searches with modest mass and elution time criteria can provide proteomic information for approximately one thousand proteins in a single run of <3 h. The advantage of this method over using MS/MS based techniques is the large number of identifications that occur in a single experiment as well as the basis for improved quantitation. For MS/MS experiments, the number of peptide identifications is severely restricted because of the time required to dissociate the peptides individually. These results demonstrate the utility of the AMT tag approach using capillary LC-TOF MS instruments, and also show that AMT tags developed using other instrumentation can be effectively utilized.  相似文献

Herpes virus entry mediator (HVEM) is a newly discovered member of the tumor necrosis factor receptor (TNFR) superfamily that has a role in herpes simplex virus entry, in T cell activation and in tumor immunity. We generated mAb against HVEM and detected soluble HVEM (SHVEM) in the sera of patients with various autoimmune diseases. HVEM was constitutively expressed on CD4(+) and CD8(+) T cells, CD19(+) B cells, CD14(+) monocytes, neutrophils and dendritic cells. In three-way MLR, mAb 122 and 139 were agonists and mAb 108 had blocking activity. An ELISA was developed to detect sHVEM in patient sera. sHVEM levels were elevated in sera of patients with allergic asthma, atopic dermatitis and rheumatoid arthritis. The mAbs discussed here may be useful for studies of the role of HVEM in immune responses. Detection of soluble HVEM might have diagnostic and prognostic value in certain immunological disorders.  相似文献

Aromatic ring amination reactions in the ruthenium complex of 2-(phenylazo)pyridine is described. The substitutionally inert cationic brown complex [Ru(pap)(3)](ClO(4))(2) (1) (pap = 2-(phenylazo)pyridine) reacts smoothly with aromatic amines neat and in the presence of air to produce cationic and intense blue complexes [Ru(HL(2))(3)](ClO(4))(2) (2) (HL(2) = 2-[(4-(arylamino)phenyl)azo]pyridine). These were purified on a preparative TLC plate. The X-ray structure of the new and representative complex 2c has been solved to characterize them. The results are compared with those of the starting complex, [Ru(pap)(3)](ClO(4))(2) (1). The transformation 1 --> 2 involves aromatic ring amination at the para carbon (with respect to the diazo function) of the pendant phenyl rings of all three coordinated pap ligands in 1. The transformation is stereoretentive, and the amination reaction is regioselective. The extended ligand HL(2) coordinates as a bidentate ligand and chelates to ruthenium(II) through the pyridine and one of the azo nitrogens. The amine nitrogen of this bears a hydrogen atom and remains uncoordinated. Similarly, the amination reaction on the mixed-ligand complex [Ru(pap)(bpy)(2)](ClO(4))(2) produces the blue complex [Ru(HL(2))(bpy)(2)](ClO(4))(2) (3) as anticipated. The reactions of [RuCl(2)(dmso)(4)] and [Ru(S)(2)(L)(2)](2+) (dmso = dimethyl sulfoxide, S = labile coordinated solvent, L = 2,2'-bipyridine (bpy) and pap) with the preformed HL(2) ligand have been explored. The structure of the representative complex [RuCl(2)(HL(2a))(2)] (5a) is reported. It has the chlorides in trans configuration while the pyridine as well as azo nitrogens are in cis geometry. Optical spectra and redox properties of the newly synthesized complexes are reported. All the ruthenium complexes of HL(2) are characterized by their intense blue solution colors. The lowest energy transitions in these complexes appear near 600 nm, which have been attributed to intraligand charge-transfer transitions. For example, the lowest energy visible range transition in [Ru(HL(2b))(3)](2+) appears at 602 nm and its intensity is 65 510 M(-1) cm(-1). All the tris chelates show multiple-step electron-transfer processes. In [Ru(HL(2))(3)](2+), six reductions waves constitute the complete electron-transfer series. The electrons are believed to be added successively to the three azo functions. In the mixed-ligand chelates [Ru(HL(2))(pap)(2)](2+) and [Ru(HL(2))(bpy)(2)](2+) the reductions due to HL(2), pap, and bpy are observed.  相似文献

Bioassay‐guided fractionation of an AcOEt extract of Casearia membranacea resulted in the isolation of six new clerodane diterpenes, caseamembrins G–L ( 1 – 6 ). The structures of the new compounds, including their relative configurations, were established by an extensive study of their spectral data, especially 2D NMR. The cytotoxic activities of the isolated diterpenes against human oral epidermoid (KB), cervical epitheloid (Hela), and liver (Hep59T/VGH) carcinoma cell lines were investigated.  相似文献

Earlier study showed that glucocorticoid induced tumor necrosis factor receptor (GITR), a new TNFR family, activated murine macrophages to express inducible nitric oxide synthase (iNOS) and to generate nitric oxide (NO). A possible involvement of pro-inflammatory cytokines on NO production by GITR was investigated in vitro systems and signaling molecules contributing to sGITR-induced iNOS production are determined in Raw 264.7 cells, a murine macrophage cell line. The result showed that the synergy was afforded by the combination of GITR with IFN-g in a dose-dependent manner but IFN-gamma alone was not able to induce NOS. No effects were observed with TNF-alpha, IL-1beta, or IL-6 co-treated with GITR. To determine signaling molecules contributing to sGITR-induced iNOS production, a specific inhibitor for signal pathway proteins tested showed that PDTC (NF-kappaB) and genistein (tyrosine kinase) inhibited NOS induction significantly, while sodium orthovanadate (tyrosine phosphatase) potentiated NOS expression. These results suggest that activations of NF-kappaB were involved in induction of iNOS by GITR and IFN-gamma priming caused earlier and stronger NF-kappaB activation.  相似文献

Paranemic crossover (PX) DNA is a four-stranded coaxial DNA complex containing a central dyad axis that relates two flanking parallel double helices. The strands are held together exclusively by Watson-Crick base pairing. The key feature of the structure is that the two adjacent parallel DNA double helices form crossovers at every point possible. Hence, reciprocal crossover points flank the central dyad axis at every major or minor groove separation. This motif has been modeled and characterized in an oligonucleotide system; a minor groove separation of five nucleotide pairs and major groove separations of six, seven, or eight nucleotide pairs produce stable PX DNA molecules; a major groove separation of 9 nucleotide pairs is possible at low concentrations. Every strand undergoes a crossover every helical repeat (11, 12, 13, or 14 nucleotides), but the structural period of each strand corresponds to two helical repeats (22, 24, 26, or 28 nucleotides). Nondenaturing gel electrophoresis shows that the molecules are stable, forming well-behaved complexes. PX DNA can be produced from closed dumbbells, demonstrating that the molecule is paranemic. Ferguson analysis indicates that the molecules are similar in shape to DNA double crossover molecules. Circular dichroism spectra are consistent with B-form DNA. Thermal transition profiles suggest a premelting transition in each of the molecules. Hydroxyl radical autofootprinting analysis confirms that there is a crossover point at each of the positions expected in the secondary structure. These molecules are generalized Holliday junctions.  相似文献
