The synthesis of a poly(azo)urethane by fixing CO(2) in bis-epoxide followed by a polymerization reaction with an azodiamine is presented. Since isocyanate is not used in the process, it is termed "clean method" and the polymers obtained are named "NIPUs" (non-isocyanate polyurethanes). Langmuir films were formed at the air-water interface and were characterized by surface pressure vs mean molecular area per mer unit (Pi-A) isotherms. The Langmuir monolayers were further studied by running stability tests and cycles of compression/expansion (possible hysteresis) and by varying the compression speed of the monolayer formation, the subphase temperature, and the solvents used to prepare the spreading polymer solutions. The Langmuir-Blodgett (LB) technique was used to fabricate ultrathin films of a particular polymer (PAzoU). It is possible to grow homogeneous LB films of up to 15 layers as monitored using UV-vis absorption spectroscopy. Higher number of layers can be deposited when PAzoU is mixed with stearic acid, producing mixed LB films. Fourier transform infrared (FTIR) absorption spectroscopy and Raman scattering showed that the materials do not interact chemically in the mixed LB films. The atomic force microscopy (AFM) and micro-Raman technique (optical microscopy coupled to Raman spectrograph) revealed that mixed LB films present a phase separation distinguishable at micrometer or nanometer scale. Finally, mixed and neat LB films were successfully characterized using impedance spectroscopy at different temperatures, a property that may lead to future application as temperature sensors. A method for separating proteins with a molecular mass difference of 2 kDa using SDS-PAGE under nonreducing conditions is presented. A sample mixture containing several human growth hormone (hGH) isoforms was initially separated on a weak anion-exchange column. Fractions rich in 24 kDa hGH as determined by analytical SDS-PAGE were pooled and further separated by cation-exchange chromatography. The fractions pooled from the cation-exchange chromatography contained two hGH isoforms with a 2 kDa molecular mass difference according to SDS-PAGE analysis, 22 and 24 kDa hGH. The 22 and 24 kDa hGH were separated using continuous-elution preparative double-inverted gradient PAGE (PDG-PAGE) under nonreducing conditions. The preparative electrophoresis gel was composed of three stacked tubular polyacrylamide matrices, a 4% stacking gel, a 13-18% linear gradient gel, and a 15-10% linear inverted gradient gel. Fractions containing purified 24 kDa hGH were pooled and Western blot analysis displayed immunoreactivity to antihGH antibodies. PDG-PAGE provides researchers with an electrophoretic technique to preparatively purify proteins under nonreducing conditions with molecular mass differences of 2 kDa. 相似文献

The bimolecular rate constants for the reactions of sulfate radicals with epicatechin (EC), epicatechingallate (ECG), and epigallocatechingallate (EGCG) were found to be (1.46 ± 0.06) × 109, (1.20 ± 0.08) × 109, and (1.04 ± 0.07) × 109, respectively. The activation energy [EA = 9 ± 3 kJ mol?1] and preexponential factor [A = (4.8 ± 0.6) × 1010] for the reaction of EC with the sulfate radical were measured in the temperature range 288–303 K. The phenoxyl radicals of EC (λmax = 310 nm) were obtained both by the reaction of this flavonoid with the sulfate radicals and by photoionization. In this paper, we use random walk theory to describe the length dynamics of microtubules, one of the principal components of the cytoskeleton. We present a simple two-state model (growing and shrinking) of microtubule length evolution that incorporates a variable rate of switching between the states. Using the generating function technique, we calculate the mean length of microtubule, its variance and diffusion coefficient. We also report analytical and computer simulation results for the mean number of positive monomers in microtubule, and find good qualitative agreement with experimental data. 相似文献

In this communication, we revise some aspects of the [ideal gas/Van der Waals fluid] partition, derived in an earlier publication. The general character of the conclusions concerning the dependence of the partial molar free energy of solution, deltaGs(n), on the chain length, n, of linear solute molecules is shown through the relationship with more general partition formulations. Simultaneously, the correction of an error in the expression of the retention time dependence on the phase ratio of the chromatographic column, beta, is carried out. A highly sensitive gas chromatographic assay is described for the simultaneous determination of gallopamil, a calcium channel blocking agent, and its major metabolite, norgallopamil. A multi-step extraction procedure is employed followed by on-column capillary gas chromatographic analysis using nitrogen-selective detection. Acetylation of norgallopamil is performed to enable accurate quantification of the metabolite. Linearity was achieved over the range 1-50 ng/ml for both analytes.
