Synthesis of 6-oxopyrimidin-1(6H)-yl benzamide derivatives and evaluation of their antibacterial and cytotoxic activity

A series of novel 2-alkylamino and 2, 4-dialkyl amino 6-oxopyrimidin-1(6H)-yl) benzamide derivatives were prepared in good yields from a base-catalyzed ring opening of oxadiazolo[3,2-a]pyrimidin-5-one and evaluated for their antibacterial and cytotoxicity. Most of the compounds exhibited antibacterial activity. In particular, compounds 5b and 5k exhibited considerable antibiotic activity against Klebsiella pneumonia and Bacillus cereus. In addition, compounds 5g and 5i also inhibited the growth of two human tumor cell lines (A549 and H460) at micromolar concentrations.     

Synthesis of Novel Drug‐Like Small Molecules Based on Quinoxaline Containing Amino Substitution at C‐2

A series of novel “drug‐like” small molecules based on quinoxaline containing amino substitution at C‐2 were synthesized. All these molecules were prepared either via the reaction of 2‐phenyl‐3‐(piperazin‐1‐yl)quinoxaline with acyl bromides or benzyl bromides or various carboxylic acids or via the reaction of 2‐chloro‐3‐phenylquinoxaline with various amines. The structures of these novel compounds were confirmed by spectral analysis. The strategy used is simple and efficient and afforded good yields of quinoxaline derivatives.     

Simultaneous determination of sitagliptin and simvastatin in human plasma by LC‐MS/MS and its application to a human pharmacokinetic study

A simple, rapid and sensitive liquid chromatography–tandem mass spectrometric (LC‐MS/MS) assay method has been developed and validated for simultaneous quantification of sitagliptin and simvastatin in human plasma. Carbamazepine was used as an internal standard (IS). The analytes and IS were extracted from the human plasma by liquid–liquid extraction technique. The reconstituted samples were chromatographed on an Alltima HP C₁₈ column using an isocratic solvent mixture [acetonitrile–5 mm ammonium acetate (pH 4.5), 85:15 (v/v)] at a flow rate of 1.0 mL/min. Method validation was performed as per Food and Drug Administration guidelines and the results met the acceptance criteria. The calibration curves obtained were linear (r² ≥ 0.99) over the concentration range of 0.10–501 and 0.05–105 ng/mL for sitagliptin and simvastatin, respectively. The results of the intra‐ and inter‐day precision and accuracy studies were well within the acceptable limits. Both the analytes were found to be stable in a battery of stability studies. The method is precise and sensitive enough for its intended purpose. A run time of 3.0 min for each sample made it possible to analyze more than 300 plasma samples per day. Synthesis of indolo[2,3-a]quinolizine and hexahydro-1H-indolizino[8,7-b]indole derivatives by cascade condensation, cyclization, and Pictet-Spengler reaction: an application to the synthesis of (±)-harmicine

Synthesis of indole alkaloid related compounds using Schiff base formation, intramolecular cyclization (or N-alkylation), and Pictet-Spengler reaction as a cascade one pot condensation has been reported. The cascade chemistry has been applied to the synthesis of (±)-harmicine as a key step.     

A novel LC-MS/MS method for simultaneous quantification of tenofovir and lamivudine in human plasma and its application to a pharmacokinetic study

A new, rapid, sensitive and specific LC‐MS/MS method has been developed and validated for the simultaneous quantification of tenofovir and lamivudine in human plasma using abacavir as an internal standard. An API‐4000 LC‐MS/MS with electrospray ionization was operated in multiple‐reaction monitoring mode for the analysis. The analytes were extracted from plasma by solid‐phase extraction technique using an Oasis HLB cartridge. The reconstituted samples were chromatographed on a Chromolith ROD speed C₁₈ column using a mixture of 0.1% formic acid in water and acetonitrile (90:10 v/v) at a flow‐rate of 1 mL/min. The method was validated as per the FDA guidelines. The calibration curves were found to be linear in the range of 5–600 ng/mL for tenofovir and 25– 4000 ng/mL for lamivudine. The intra‐ and inter‐day precision and accuracy results were well within the acceptable limits. A run time of 2.8 min consumed for each sample made it possible to analyze more samples per day. Multidione, a novel diterpenoid from Jatropha multifida

A novel diterpenoid, multidione, has been isolated from the stems of Jatropha multifida and its structure has been settled from 1D and 2D NMR spectra. The compound possesses a phenolic moiety and a long side chain, structurally similar to the B ring of other lathyrane-diterpenoids in seco-form.The compound has possibly been derived biogenetically from a related lathyrane-diterpenoid.     

Efficient synthesis of homoallylic alcohols and amines using 2,4,6-trichloro-1,3,5-triazine

The allylation of aldehydes and imines has efficiently been carried out by treatment with allyltributylstannane in the presence of a catalytic amount of 2,4,6-trichloro-1,3,5-triazine at room temperature to form the corresponding homoallylic alcohols and amines, respectively, in high yields.     

A Stereoselective Total Synthesis of Verbalactone

A stereoselective total synthesis of verbalactone has been achieved starting from commercially available hexanal. The sequence involves Maruoka allylation, diastereoselective iodine‐induced electrophillic cyclization, ring opening of epoxide, and Yamaguchi macrolactonization as the key steps.
