The synthesis, structure, and physical properties of a Heisenberg exchange-coupled cluster containing naphthalene groups are described. [Fe2(O)(O2CCH2C10H7)2(TACN-Me3)2]2+ (3) crystallizes in space group P1 with unit cell parameters a = 12.94(2) A, b = 14.84(2) A, c = 15.23(2) A, alpha = 101.12(7) degrees, beta = 90.8(1) degrees, gamma = 114.14(7) degrees, V = 2605(6) A3, and Z = 2 with R = 0.0425 and wR2 = 0.1182. Variable-temperature magnetic susceptibility data indicate that the two high-spin FeIII centers are antiferromagnetically coupled with J = -105 cm-1 (H = -2 JS1.S2), which is typical for this class of compounds. The room-temperature static emission spectrum of the compound in deoxygenated CH3CN solution is centered near 335 nm and has features reminiscent of both methyl-2-naphthylacetate (1) and [Zn2(OH)(O2CCH2C10H7)2(TACN-Me3)2]+ (2) with the following two caveats: (1) the overall emission intensity is roughly a factor of 10 less than that of the free ester (1, phi = 0.13) or the ZnII analogue (2, phi = 0.14), and (2) there is significant broadening of the low-energy shoulder of the emission envelope. Time-correlated single photon counting data revealed biphasic emission for 3 with tau 1 = 4.6 +/- 1 ns and tau 2 = 47 +/- 1 ns. The latter compares favorably with that found for 2 (tau = 47 +/- 1 ns) and is assigned as the S0-S1 fluorescence of naphthalene. Emission anisotropy, time-gated emission spectra, and nanosecond time-resolved absorption measurements all support the assignment of the 4.6 ns component as being due to a singlet excimer that forms between the two naphthylacetate groups of 3, a process that is likely mediated by the structural constraints of the oxo-bis-carboxylato diiron core. No direct evidence for intramolecular electron and/or energy transfer from the photoexcited naphthyl group to the iron-oxo core was obtained, suggesting that the short-lived excimer may contribute to circumventing such pathways in this type of system.  相似文献

Synthesis and targeted delivery of dendrimer-based fluorescent biosensors in living HeLa cells are reported. Following electroporation dendrimers are shown to display specific subcellular localization depending on their size and surface charge and this property is preserved when they are functionalized with sensing moieties. We analyze the case of double dendrimer conjugation with pH-sensitive and pH-insensitive molecules leading to the realization of ratiometric pH sensors that are calibrated in vitro and in living cells. By tuning the physicochemical properties of the dendrimer scaffold sensors can be targeted to specific cellular compartments allowing selective pH measurements in different organelles in living cells. In order to demonstrate the modularity of this approach we present three different pH sensors with tuned H(+) affinity by appropriately choosing the pH-sensitive dye. We argue that the present methodology represents a general approach toward the realization of targetable ratiometric sensors suitable to monitor biologically relevant ions or molecules in living cells.  相似文献

Mass spectrometers equipped with atmospheric pressure ion sources (API-MS) have been designed to be interfaced with liquid chromatographs (LC) and have rarely been connected to gas chromatographs (GC). Recently, we introduced a heated nebulizer microchip and showed its potential to interface liquid microseparation techniques and GC with API-MS. This study demonstrates the feasibility of GC-microchip atmospheric pressure photoionization-tandem mass spectrometry (GC-μAPPI-MS/MS) in the analysis of underivatized anabolic steroids in urine. The APPI microchip provides high ionization efficiency and produces abundant protonated molecules or molecular ions with minimal fragmentation. The feasibility of GC-μAPPI-MS/MS in the analysis of six selected anabolic steroids in urine samples was studied with respect to intra-batch repeatability, linearity, linear range, and limit of detection (LOD). The method showed good sensitivity (LODs 0.2-1 ng/mL), repeatability (relative standard deviation<10%), and linearity (regression coefficient≥0.9995) and, therefore, high potential for the analysis of anabolic steroids. Quantitative performance of the method was tested with two authentic urine samples, and the results were in good agreement with those obtained with conventional GC-electron ionization-MS after derivatization.  相似文献

Three divalent copper coordination polymers containing aromatic dicarboxylate ligands and the long-spanning tethering imine bis(4-pyridylmethyl)piperazine (bpmp) have been prepared and structurally characterized. The length of the dicarboxylate pendant arms, carboxylate binding mode, and the inclusion of anionic components play a synergistic role in structure direction in this system. {[Cu(ip)(bpmp)(H2O)]·5H2O}n (ip = isophthalate, 1) displays neutral (4,4) rectangular coordination polymer grids that stack in an ABCD repeat pattern. Use of the longer pendant arm dicarboxylate 1,3-phenylenediacetate (phda) resulted in {[Cu2(phda)2(bpmp)]·H2O}n (2), a 3-D network coordination polymer with primitive cubic topology that features strongly antiferromagnetically coupled (J = −331(1) cm−1) {Cu2(CO2)4} paddlewheel units. In the presence of excess nitrate ions, {[Cu(phda)(Hbpmp)](NO3)·3H2O}n (3) was isolated instead of 2; 3 manifests cationic 2-D coordination polymer layers built from weakly antiferromagnetically coupled (J = −2.43(1) cm−1) {Cu2O2} dimers linked through phda and protonated bpmp ligands. The striking difference in magnetic properties is ascribed to the equatorial–equatorial versus axial–equatorial bridging of copper coordination spheres in 2 and 3, respectively.  相似文献

Protein phosphorylation is one of the most common and extensively studied posttranslational modifications (PTMs). Compared to the O-phosphorylation of Ser, Thr, and Tyr residues, our understanding of histidine phosphorylation is relatively limited, particularly in higher eukaryotes, due to technical difficulties stemming from the intrinsic instability and isomerism of phosphohistidine (pHis). We report the design and synthesis of stable and nonisomerizable pHis analogues. These pHis analogues were successfully utilized in solid-phase peptide synthesis and semi-synthesis of histone H4. Significantly, the first antibody that specifically recognizes pHis was obtained using the synthetic peptide as the immunogen.  相似文献

Several samples of polymeric micelles, formed by amphiphilic derivatives of PHEA, obtained by grafting into polymeric backbone of PEGs and/or hexadecylamine groups (PHEA-PEG-C(16) and PHEA-C(16)) and containing different amount of Tamoxifen, were prepared. All Tamoxifen-loaded polymeric micelles showed to increase drug water solubility. TEM studies provided evidence of the formation of supramolecular core/shell architectures containing drug, in the nanoscopic range and with spherical shape. Samples with different amount of encapsulated Tamoxifen were subjected to in vitro cytotoxic studies in order to evaluate the effect of Tamoxifen micellization on cell growth inhibition. All samples of Tamoxifen-loaded polymeric micelles showed a significantly higher antiproliferative activity in comparison with free drug, probably attributable to fluidification of cellular membranes, caused by amphiphilic copolymers, that allows a higher penetration of the drug into tumoral cells. To gain preliminary information about the potential use of prepared micelles as Tamoxifen drug delivery systems, studies evaluating drug release ability of micelle systems in media mimicking biological fluids (buffer solutions at pH 7.4 and 5.5) and in human plasma were carried out. These studies, performed evaluating the amount of Tamoxifen that remains in solution as a function of time, showed that at pH 7.4, as well as in plasma, PHEA-C(16) polymeric micelles were able to release lower drug amounts than PHEA-PEG(5000)-C(16) ones, while at pH 5.5, the behavior difference between two kind of micelles was less pronounced.  相似文献

The extent of conformational change that calcium binding induces in EF-hand proteins is a key biochemical property specifying Ca(2+) sensor versus signal modulator function. To understand how differences in amino acid sequence lead to differences in the response to Ca(2+) binding, comparative analyses of sequence and structures, combined with model building, were used to develop hypotheses about which amino acid residues control Ca(2+)-induced conformational changes. These results were used to generate a first design of calbindomodulin (CBM-1), a calbindin D(9k) re-engineered with 15 mutations to respond to Ca(2+) binding with a conformational change similar to that of calmodulin. The gene for CBM-1 was synthesized, and the protein was expressed and purified. Remarkably, this protein did not exhibit any non-native-like molten globule properties despite the large number of mutations and the nonconservative nature of some of them. Ca(2+)-induced changes in CD intensity and in the binding of the hydrophobic probe, ANS, implied that CBM-1 does undergo Ca(2+) sensorlike conformational changes. The X-ray crystal structure of Ca(2+)-CBM-1 determined at 1.44 A resolution reveals the anticipated increase in hydrophobic surface area relative to the wild-type protein. A nascent calmodulin-like hydrophobic docking surface was also found, though it is occluded by the inter-EF-hand loop. The results from this first calbindomodulin design are discussed in terms of progress toward understanding the relationships between amino acid sequence, protein structure, and protein function for EF-hand CaBPs, as well as the additional mutations for the next CBM design.  相似文献

Hydrogels have been prepared using two partially deacetylated hyaluronan (deHA) samples having a molar ratio of free amino groups to total initial N-acetamido groups equal to 23 and 33%, respectively. Taking advantage of such primary amino functionalities, polymeric networks could be easily obtained in aqueous media by means of two different crosslinking procedures, namely: a) Ugi multicomponent reactions and, more simply, b) glutaraldehyde reticulation. In this paper, we wish to report on the synthesis of deHA based hydrogels and, in particular, on their structural elucidation as obtained by NMR studies.
