A fast and reliable analytical method using microwave assisted extraction has been developed. Several extraction solvents (methanol (MeOH) and ethanol (EtOH), 30-70% in water and water), temperatures (50-150 °C), extraction solvent volume, as well as the sample size (1.0-0.1 g) and extraction time (5-30 min) were studied for the optimization of the extraction protocol. The optimized extraction conditions for quantitative recoveries were: 0.5 g of sample, 50 °C, 20 min and 50% ethanol as extracting solvent. No degradation of the isoflavones was observed using the developed extraction protocol and a high reproducibility was achieved (>95%).  相似文献

Reaction of the tetranuclear complex [PtIMe3]4 with the ligand (S)- and (R)-Ph2P(C6H4)CHNC*H(Ph)Me in a 1:4 molar ratio yields the mononuclear neutral complexes in diastereoisomeric mixtures [PtIMe32-Ph2P(C6H4)CHNC*H(Ph)Me-P,N}]. Iodide abstraction from mixture with AgBF4 in the presence of pyridine (Py) induces a reductive elimination reaction with loss of ethane, leading to the cationic complex [PtMe(Py){κ2-Ph2P(C6H4)CHNC*H(Ph)Me-P,N}][BF4] [C* = (S)-, 3; (R)-, 4]. When this reaction was carried out in the presence of PPh3 a consecutive orthometallation reaction with loss of methane is produced, forming the cationic complex [Pt(PPh3){κ3-Ph2P(C6H4)CHNC*H(C6H4)Me-C,P,N}][BF4], [(S)-, 5; (R)-, 6]. All species were characterised in solution by 1H and 31P{1H} NMR spectroscopy, elemental analysis and mass spectrometry.The crystal structure of the diastereoisomer (OC-6-44-C)-[PtIMe32-(R)-Ph2P(C6H4)CHNC*H(Ph)Me-P,N}] has been determined by single-crystal X-ray diffraction.  相似文献

QS-21 is one of the most promising new adjuvants for immune response potentiation and dose-sparing in vaccine therapy given its exceedingly high level of potency and its favorable toxicity profile. Melanoma, breast cancer, small cell lung cancer, prostate cancer, HIV-1, and malaria are among the numerous maladies targeted in more than 80 recent and ongoing vaccine therapy clinical trials involving QS-21 as a critical adjuvant component for immune response augmentation. QS-21 is a natural product immunostimulatory adjuvant, eliciting both T-cell- and antibody-mediated immune responses with microgram doses. Herein is reported the synthesis of QS-21A(api) in a highly modular strategy, applying novel glycosylation methodologies to a convergent construction of the potent saponin immunostimulant. The chemical synthesis of QS-21 offers unique opportunities to probe its mode of biological action through the preparation of otherwise unattainable nonnatural saponin analogues.  相似文献

Using a simple model it is shown that the cost of constraining a hydrated potassium ion inside a narrow nanopore is smaller than the cost of constraining the smaller hydrated sodium ion. The former allows for a greater distortion of its hydration shell and can therefore maintain a better coordination. We propose that in this way the larger ion can go through narrow pores more easily. This is relevant to the molecular basis of ion selective nanopores and since this mechanism does not depend on the molecular details of the pore, it could also operate in all sorts of nanotubes, from biological to synthetic.  相似文献

We study the temporal dynamics of the generalized repressilator, a network of coupled repressing genes arranged in a directed ring topology, and give analytical conditions for the emergence of a finite sequence of unstable periodic orbits that lead to reachable long-lived oscillating transients. Such transients dominate the finite time horizon dynamics that is relevant in confined, noisy environments such as bacterial cells (see our previous work [Strelkowa and Barahona, J. R. Soc. Interface 7, 1071 (2010)]), and are therefore of interest for bioengineering and synthetic biology. We show that the family of unstable orbits possesses spatial symmetries and can also be understood in terms of traveling wave solutions of kink-like topological defects. The long-lived oscillatory transients correspond to the propagation of quasistable two-kink configurations that unravel over a long time. We also assess the similarities between the generalized repressilator model and other unidirectionally coupled electronic systems, such as magnetic flux gates, which have been implemented experimentally.  相似文献

A route for the degradation of the side chain of ent-labdane derivatives has been devised, giving the useful synthon 2beta,12-dihydroxy-13,14,15,16,17-pentanor-ent-labdane-8-one (8). The use of this compound in the preparation of terpenylquinone derivatives shall be reported elsewhere. In addition we have synthesized the compound 2beta,12-diacetoxy-8beta,17-epoxy-13,14,15,16-tetranor-ent-labdane (10), which upon catalytic epoxide ring opening in alkaline or acid media gave rise in all cases to the formation of tricyclicc ompounds.  相似文献

Huddling is a grouping behavior where animals maintain close bodily contact and save energy. We tested the hypothesis that this thermoregulatory behavior behaves as a system with continuous (second‐order phase) transition called critical when the environmental temperature (driving parameter) is near a critical value. To do so, we followed theoretical and experimental approaches, examining data from geometrical models, metabolic rate during huddling in small mammals, and also conducting an experiment on thermoregulatory huddling behavior with white mice. Our results support all predictions for systems under continuous‐phase transition triggered by low temperatures, a phenomenon reported for first time in a biological system. We suggest that huddling behavior in social animals, a recognized adaptive behavior, may be considered a self‐organized system coupled with an external driving parameter. © 2011 Wiley Periodicals, Inc.
