We have analyzed the abundance of bacterial sulfonosphingolipids, including rosette-inducing factors (RIFs), in seven bacterial prey strains by using high-resolution tandem mass spectrometry (HRMS2) and molecular networking (MN) within the Global Natural Product Social Molecular Networking (GNPS) web platform. Six sulfonosphingolipids resembling RIFs were isolated and their structures were elucidated based on comparative MS and NMR studies. Here, we also report the first total synthesis of two RIF-2 diastereomers and one congener in 15 and eight synthetic steps, respectively. For the total synthesis of RIF-2 congeners, we employed a decarboxylative cross-coupling reaction to synthesize the necessary branched α-hydroxy fatty acids, and the Garner-aldehyde approach to generate the capnine base carrying three stereogenic centers. Bioactivity studies in the choanoflagellate Salpingoeca rosetta revealed that the rosette inducing activity of RIFs is inhibited dose dependently by the co-occurring sulfonosphingolipid sulfobacins D and F and that activity of RIFs is specific for isolates obtained from Algoriphagus.  相似文献

Combining nanotechnology and bioorthogonal chemistry for theranostic strategies offers the possibility to develop next generation nanomedicines. These materials are thought to increase therapeutic outcome and improve current cancer management. Due to their size, nanomedicines target tumors passively. Thus, they can be used for drug delivery purposes. Bioorthogonal chemistry allows for a pretargeting approach. Higher target-to-background drug accumulation ratios can be achieved. Pretargeting can also be used to induce internalization processes or trigger controlled drug release. Colloidal gold nanoparticles (AuNPs) have attracted widespread interest as drug delivery vectors within the last decades. Here, we demonstrate for the first time the possibility to successfully ligate AuNPs in vivo to pretargeted monoclonal antibodies. We believe that this possibility will facilitate the development of AuNPs for clinical use and ultimately, improve state-of-the-art patient care.  相似文献

A number of hydrogen-bond related quantities—geometries, interaction energies, dipole moments, dipole moment derivatives, and harmonic vibrational frequencies—were calculated at the Hartree—Fock, MP2, and different DFT levels for the HCN dimer and the periodic HCN crystal. The crystal calculations were performed with the Hartree—Fock program CRYSTAL92, which routinely allows an a posteriori electron-correlation correction of the Hartree—Fock obtained lattice energy using different correlation-only functionals. Here, we have gone beyond this procedure by also calculating the electron-correlation energy correction during the structure optimization, i.e., after each CRYSTAL92 Hartree—Fock energy evaluation, the a posteriori density functional scheme was applied. In a similar manner, we optimized the crystal structure at the MP2 level, i.e., for each Hartree—Fock CRYSTAL92 energy evaluation, an MP2 correction was performed by summing the MP2 pair contributions from all HCN molecules within a specified cutoff distance. The crystal cell parameters are best reproduced at the Hartree—Fock and the nongradient-corrected HF + LDA and HF + VWN levels. The BSSE-corrected MP2 method and the HF + P91, HF + LDA, and HF + VWN methods give lattice energies in close agreement with the ZPE-corrected experimental lattice energy. The (HCN)2 dimer properties are best reproduced at the MP2 level, at the gradient-corrected DFT levels, and with the B3LYP and BHHLYP methods. © 1996 John Wiley & Sons, Inc.  相似文献

This article deals with excited-state properties and spectroscopies which probe them. We argue that the orbitals obtained in local density theory form an excellent basis for discussing excited states and that the self-energy operator (or optical potential) associated with one-electron-type excited states is closely diagonal in such a basis. This diagonality often leads to considerable simplifications. We also discuss the reasons for the successes of the “GW -approximation” for the optical potential in predicting excited states. We further discuss a recent approach to calculate spectroscopic data. In particular, we present explicit results of this approach to photoelectron emission. The approach treats correlations within the target exactly but neglects some terms in the coupling between target and probe electron (= the photoemitted electron). This leads to closed expressions which exhibit a number of qualitative features. One is the cancellation of intrinsic and extrinsic losses at threshold excitation. Another is the broadening of the quasi-particles; we have energy-broadening for the occupied states but broadening through decaying wave functions for unoccupied states. © 1995 John Wiley & Sons, Inc.  相似文献

Reactivity and swelling of nanometer films of alternating maleic anhydride copolymers were investigated in dependence on the kind of comonomer and molar mass of copolymer in aqueous solution at pH 7.4 and pH 3.0 in order to reveal their characteristics under physiological conditions. Fully hydrolyzed (maleic acid) chains of the copolymers with styrene, propene, and ethylene comonomers covalently bound to SiO2 substrates showed a "mushroom" swelling behavior at pH 7.4 with a layer thickness scaling of N3/5. Decreasing the environmental pH was found to induce a comonomer-dependent shrinking or collapse of the immobilized polymers due to the change in ionization. From the swelling kinetics of non-hydrolyzed chains, the time constants and characteristics of swelling and anhydride hydrolysis were determined and found to depend on the type of comonomer. The short- and long-term swelling kinetics [l approximately t and approximately ln(t)1/2] were found to be in agreement with theoretical models of polymer swelling, while at intermediate time scales enhanced swelling was observed due to hydrolysis reaction of maleic anhydride groups. The findings elucidate the variety of properties of maleic anhydride copolymer films under physiological conditions, which can advantageously be applied for biofunctionalization of different templates.  相似文献

[structure: see text] An ester dienolate [2,3]-Wittig rearrangement was utilized to access the alkylated citric acid skeleton 6 that is characteristic for the viridiofungins and other members of the alkyl citrate family of secondary natural products. The [2,3]-sigmatropic rearrangement of (Z,Z)-15 provided the rearrangement product (+/-)-syn-16 in moderate yield and with very good diastereoselectivity. A Julia-Kocienski olefination efficiently served to connect the polar head (+/-)-syn-26 with the lipophilic tail (32a-c) of the viridiofungins. Amide formation between the racemic viridiofungin precursors 35a-c and the enantiomerically pure amino acid L-tyrosine methyl ester followed by preparative reversed-phase HPLC provided the isopropyl dimethyl ester of viridiofungin A ((+)-39a), A2 ((+)-39b), and A4 ((+)-39c) as well as the nonnatural diastereomers (-)-38a-c.  相似文献

We have investigated the use of isoelectric focusing and immunodetection for the separation of low molecular weight species of amyloid-beta (Aβ) peptides from their aggregates. From solutions of Aβ1–40 or Aβ1–42 monomeric peptides, low molecular weight material appeared at a pI value of ca. 5, while the presence of aggregates was detected as bands, observed at a pI of 6–6.5. The formation of Aβ aggregates (protofibrils) was verified by a sandwich ELISA, employing the protofibril conformation-selective antibody mAb158. In order to study the aggregation behavior when using a mixture of the monomers, we utilized the IEF separation combined with Western blot using two polyclonal antisera, selective for Aβ1–40 and Aβ1–42, respectively. We conclude that both monomers were incorporated in the aggregates. In a further study of the mixed aggregates, we used the protofibril conformation-selective antibody mAb158 for immunoprecipitation, followed by nanoelectrospray mass spectrometry (IP-MS). This showed that the Aβ1–42 peptide is incorporated in the aggregate in a significantly larger proportion than its relative presence in the original monomer composition. IP-MS with mAb158 was also performed, and compared to IP-MS with the Aβ-selective antibody mAb1C3, where a monomeric Aβ1–16 peptide was added to the protofibril preparation. Aβ1–16 is known for its poor aggregation propensity, and acted therefore as a selectivity marker. The results obtained confirmed the protofibril conformation selectivity of mAb158.  相似文献

An analytical method for the determination of inorganic arsenic in fish samples using HPLC-ICP-MS has been developed. The fresh homogenised sample was subjected to microwave-assisted dissolution by sodium hydroxide in ethanol, which dissolved the sample and quantitatively oxidised arsenite (As(III)) to arsenate (As(V)). This allowed for the determination of inorganic arsenic as a single species, i.e. As(V), by anion-exchange HPLC-ICP-MS. The completeness of the oxidation was verified by recovery of As(V) which was added to the samples as As(III) prior to the dissolution procedure. The full recovery of As(V) at 104±7% (n=5) indicated good analytical accuracy. The uncertified inorganic arsenic content in the certified reference material TORT-2 was 0.186±0.014 ng g–1 (n=6). The method was employed for the determination of total arsenic and inorganic arsenic in 60 fish samples including salmon from fresh and saline waters and in plaice. The majority of the results for inorganic arsenic were lower than the LOD of 3 ng g–1, which corresponded to less than one per thousand of the total arsenic content in the fish samples. For mackerel, however, the recovery of As(III) was incomplete and the method was not suited for this fat-rich fish.  相似文献
