Pseudomonas aeruginosa biofilm-related infections are the major cause of premature death in cystic fibrosis patients. Strategies to induce biofilm dispersal are of interest, because of their potential in preventing biofilm-related infections. Our previous work demonstrated that n-butanolic Cyclamen coum extract with ciprofloxacin could eliminate 1- and 3-day-old P. aeruginosa PAO1 biofilms. To gain new insights into the role of C. coum extract and its synergistic effect with ciprofloxacin in eliminating P. aeruginosa PAO1 biofilms, two-dimensional gel electrophoresis (2-DE) in combination with mass spectrometry-based protein identification were used. Changes in the bacterial protein expression were analyzed when 3-day-old biofilm cells were exposed to the C. coum extract alone and in combination with ciprofloxacin. Proteins involved in alginate biosynthesis, quorum sensing, adaptation/protection, carbohydrate and amino acid metabolism showed a weaker expression in the C. coum extract-ciprofloxacin-treated biofilm cells compared to those in the untreated cells. Interestingly, the proteome of C. coum extract-ciprofloxacin-treated biofilm revealed more resemblance to the planktonic phenotype than to the biofilm phenotype. It appears that saponin extract in combination with ciprofloxacin causes biofilm disruption due to several mechanisms such as motility induction, cell envelope integrity perturbation, stress protein expression reduction, and more importantly, signal transduction perturbation. In conclusion, exposure to a combination of biofilm dispersal such as saponin extract and antimicrobial agents may offer a novel strategy to control preestablished, persistent P. aeruginosa biofilms and biofilm-related infections.  相似文献

Optical emission spectroscopy (OES) was used to study the plasma generated by the activation of the gas phase CH4 + H2 both by hot filaments and by a plasma discharge (DC HF CVD) during the nucleation of CVD diamond. The effects of nucleation parameters, such as methane concentration and extraction potential, on the plasma chemistry near the surface were investigated. The density of the diamond nucleation and the quality of the diamond films were studied by scanning electron microscopy (SEM) and Raman scattering, respectively. The OES results showed that the methane concentration influenced strongly the intensity ratio of Hβ-Hα implying an increase of electron mean energy, as well as CH, CH+, C2. A correlation between the relative increase of CH+ and the diamond nucleation density was found, conversely the increase of C2 contributed to the introduction of defects in the diamond nuclei.  相似文献

This review paper provides an overview of chromogenic polymers and their classifications, mechanisms, chemistry, synthesis procedures, and potential applications with a focus on packaging. Commonly and academically accepted classifications derived from chemical engineering, material science, and packaging science are used. Furthermore, recent progress and outputs aligned with chromogenic polymers for overcoming the common challenges are discussed. A variety of human tumors like prostate and breast cancer express bombesin receptors. Due to this a new bombesin analogue was labeled with 99mTc via HYNIC and tricine as a coligand and investigated further. Peptide was synthesized on a solid phase using Fmoc strategy. Labeling with 99mTc was performed at 100 °C for 10 min and radiochemical analysis involved ITLC and HPLC methods. The stability of radiopeptide was checked in the presence of human serum at 37 °C up to 24 h. Internalization was studied with the human GRP receptor cell line PC-3. The biodistribution was studied in mice. Labeling yield of >98% was obtained corresponding to a specific activity of ~80.9 GBq/μmol. Radiopeptide internalization into PC-3 cells was moderate and specific (10.7 ± 1.2% at 4 h). A high and specific GRP receptor expressing mouse tumor and pancreas uptake (1.12 ± 0.08 and 1.04 ± 0.11% ID/g after 1 h respectively) was also determined.  相似文献

Digoxin is widely used as a cardiac glycoside drug in the treatment of various heart conditions. Because it is a toxic drug, it should be regularly monitored in the serum of patients under treatment. In this study, colloidal nanogold is synthesized and the preparation of nanogold-labeled monoclonal antibody probe to digoxin is described under optimal conditions. In addition, an immunochromatographic (IC) method for digoxin analysis employing nanogold-labeled probe is developed. With this technique, it requires only 5 min to complete the quantitative detection of digoxin. The detection time is decreased 20–30 times in comparison to radioimmunoassay (RIA). The sensitivity to digoxin was about 2 ng/ml by naked eye, which is within the therapeutic and toxic ranges of digoxin. The results of serum samples obtained by IC strip were in agreement with those obtained by RIA. The IC strip was sufficiently sensitive and accurate to be used for the rapid detection of digoxin in serum samples.  相似文献

Benzoylquinazolinone derivatives 3a–n were synthesized via a simple one-step reaction, and evaluated for in vitro α-glucosidase inhibitory activity. Compounds 3d , 3f–g , 3i , and 3m–n showed more inhibitory activity than standard drug acarbose (IC50 = 750.0 ± 1.5 μM), and among them, compound 3d displayed the highest α-glucosidase inhibitory activity (IC50 = 261.6 ± 0.1 μM). The kinetic analysis of the compound 3d revealed that this compound inhibited α-glucosidase in a competitive manner (Ki = 255 μM). The docking studies were applied to predict binding modes of the synthesized compounds in active site of α-glucosidase.  相似文献

Nanocomposite of Silica-zirconia-molybdate designated as Si-Zr-Mo was prepared via the reaction of the in situ generated zirconium-tetra octanoxide [Zr(Oct)4] through condensation of zirconium-tetra-n-butoxide and 1-octanol in a sol–gel method with sulfuric acid and tetraethylorthosilicate (TEOS) followed by grafting of MoO42? on modified silico zirconia nanocomposite under reflux conditions. The prepared nanocomposite was characterized using inductively coupled plasma (ICP), N2 sorption isotherms, transmission electron microscopy (TEM), and FT-IR spectroscopy. The as prepared nanocomposite had a surface area and pore dimension of 140 m2/g and 1.48 nm, respectively. The morphology of sulphated silico zirconia nanocomposite after immobilization MoO42? has been changed from nanoparticles to nanaorods. It was found that the synthesized nanocomposite successfully catalyze the oxidative dehydrogenation of 1,4-dihydropyridines (1,4-DHPs) with 92–100% conversion and 80–100% selectivity toward the desired products.  相似文献

An efficient, one-pot and three-component synthesis of a new series of 2-acyl-3,4,6-triaryl-1,4-dihydropyrazines is described. For the fast morphine monitoring in flow injection systems a highly sensitive method is being introduced in this work. The fast Fourier transformation with continuous cyclic voltammetry (FFTCV) in a flowing solution as a detection system was applied for the prompt morphine monitoring. Here it should be stressed that this technique is simple, precise, accurate, time saving and economical. This research includes the observation of the effects of various parameters on the sensitivity of the detection system. Eventually, it was concluded that the best condition was obtained within the pH value of 2, scan rate value of 40 V s−1, accumulation potential of 400 mV and accumulation time of 0.6 s.In detail, the noteworthy advantages which this method illustrates in comparison with other reported methods are the following; no necessity for the oxygen removal from the test solution, a sub-nano molar detection limit and the fast determination of any such compound in a wide variety of chromatographic methods.The method proved to be linear over the concentration range of 285-305,300 pg mL−1 (r = 0.999) with a detection limit and a quantitation limit of 95.5 and 285 pg mL−1, respectively. Consequently, the method illustrates the requisite accuracy, sensitivity, precision and selectivity to assay morphine in its tablets and biological fluids.  相似文献
