The detection of cancer biomarkers is of great significance for the early screening of cancer. Detecting the content of sarcosine in blood or urine has been considered to provide a basis for the diagnosis of prostate cancer. However, it still lacks simple, high-precision and wide-ranging sarcosine detection methods. In this work, a Ti3C2TX/Pt–Pd nanocomposite with high stability and excellent electrochemical performance has been synthesized by a facile one-step alcohol reduction and then used on a glassy carbon electrode (GCE) with sarcosine oxidase (SOx) to form a sarcosine biosensor (GCE/Ti3C2TX/Pt–Pd/SOx). The prominent electrocatalytic activity and biocompatibility of Ti3C2TX/Pt–Pd enable the SOx to be highly active and sensitive to sarcosine. Under the optimized conditions, the prepared biosensor has a wide linear detection range to sarcosine from 1 to 1000 µM with a low limit of detection of 0.16 µM (S/N = 3) and a sensitivity of 84.1 µA/mM cm2. Besides, the reliable response in serum samples shows its potential in the early diagnosis of prostate cancer. More importantly, the successful construction and application of the amperometric biosensor based on Ti3C2TX/Pt–Pd will provide a meaningful reference for detecting other cancer biomarkers.  相似文献

A Cu(II) coordination polymer (CP) (that is, [Cu2(OH)(H2O)2(cbca)·2H2O]n (1)) has been obtained with the reaction between Cu(II) salt and 4′-(1-carboxyethoxy)-[1,1′-biphenyl]-3,5-dicarboxylic acid (H3cbca), a semi-rigid tricarboxylic acid ligand under the hydrothermal reaction condition. Prevention activity of the new compound on hypotension after anesthesia surgery was evaluated and the related mechanism was researched at the same time. The noninvasive blood pressure monitor was firstly conducted to measure the blood pressure of the animal after compound treatment. Besides, the real time RT-PCR assay was also used to determine theα receptor relative expression on vascular endothelial cells after compound treatment.  相似文献

该文介绍了一种通过将K4Fe(CN)6配位到PbTiO3表面获得原位纳米酶的新策略。利用脱氧核糖核苷5''-单磷酸(dNMP)在PbTiO3表面的结合,阻止K4Fe(CN)6在PbTiO3表面原位形成纳米酶,构建了基于纳米酶的生物传感平台。当NF-κB p50存在时,dNMP难以产生,K4Fe(CN)6得以顺利结合到PbTiO3表面,并在其表面自发形成纳米酶,催化TMB氧化以定量检测NF-κB p50。实验表明,该方法对NF-κB p50的检测线性范围为3.0 pmol/L ~ 10 nmol/L,检出限(S/N = 3)为1.2 pmol/L,对实际样品的加标回收率为99.1% ~ 102%,相对标准偏差不大于5.3%。该方法为NF-κB p50的检测提供了一种无标记、无固定且具有信号放大作用的新策略,不仅灵敏度高、选择性好、操作简便,且在实际样品检测中具有潜在的应用价值。  相似文献

Image-guided photodynamic therapy (PDT) has received growing attention due to its non-invasiveness and precise controllability. However, the PDT efficiency of most photosensitizers are decreased in living system due to the aggregation-caused singlet oxygen (1O2) generation decreasing. Herein, we present an Iridium (III) pyridylpyrrole complex (Ir-1) featuring of aggregation-induced emission (AIE) and 1O2 generation characteristics for image-guided PDT of cancer. Ir-1 aqueous solution exhibits bright red phosphorescence peaked at 630 nm, large Stokes shift of 227 nm, and good 1O2 generation ability. The 1O2 generating rate of Ir-1 in EtOH/water mixture solution is 2.3 times higher than that of Rose Bengal. In vitro experimental results revealed that Ir-1 has better biocompatibility and higher phototoxicity compared with clinically used photosensitizers (Rose Bengal and Ce6), suggesting that Ir-1 can serve as a photosensitizer for image-guided PDT of cancer.  相似文献

Indoleamine 2,3-dioxygenase (IDO), an immune checkpoint protein, can cause the depletion of tryptophan (Trp) and accumulation of its metabolite of kynurenine (Kyn) in cancer cells, and generates the immunosuppressive microenvironment that supports tumor cell growth. A novel immunoregulatory prodrug micelle based on polyethylene glycol-derivatized an IDO-selective inhibitor of 1-methyltryptophan (1-MT), PEG-Fmoc-1-MT, was developed for inhibiting the IDO activity of the conversion of Trp to Kyn in tumor microenvironments. To investigate the 1-MT distribution and Trp/Kyn ratios in mice tumors with PEG-Fmoc-1-MT prodrug micelles treatment, a HPLC–MS/MS method for simultaneous determination of 1-MT and IDO biomakers of Trp and Kyn in mouse tumors was developed and validated. Triple-quadrupole mass spectrometry with positive electrospray ionization as source ionization in multiple reaction monitoring at m/z 219.0?→?160.1, 205.0?→?118.2, 209.0?→?146.1 and 249.3?→?148.3 was used for determination of 1-MT, Trp, Kyn and matrine (internal standard). The method demonstrated good linearity at the concentrations ranging from 10 to 10,000 ng/mL and lower limits of quantitation of 1 ng/mL for 1-MT, Trp and Kyn, respectively. The validated method was successfully applied to 1-MT tumor biodistribution and Trp/Kyn ratio studies in 4T1 tumor bearing mice i.v. with PEG-Fmoc-1-MT prodrug micelles. The mice tumors with PEG-Fmoc-1-MT prodrug micelles treatment exhibited higher 1-MT accumulation and lower Trp/Kyn ratio, in comparison with those of mice with 1-MT solution treatment. The developed PEG-Fmoc-1-MT prodrug micelles could be a promising IDO immunoregulatory prodrug micelles for cancer immunotherapy.

Because of its unsaturated bonds, C60 is susceptible to polymerize into dimers. The implications of nitrogen doping on the geometrical and electronic structure of C60 dimers have been ambiguous for years. A quarter‐century after the discovery of azafullerene dimer (C59N)2, we reported its single crystallographic structure in 2019. Herein, the unambiguous crystal structure information of (C59N)2 is elucidated specifically, revealing that the inter‐cage C—C single bond length of (C59N)2 is comparable with that of an ordinary C(sp3)‐C(sp3) single bond, and that the most stable conformer of (C59N)2 is gauche‐conformer with a dihedral angle of 66°. To amend the structural deviations, geometrical structure of (C59N)2 is optimized by a B3LYP‐D3BJ function, which is proved to be more consistent with its single crystal structure than those by the commonly used B3LYP function. Moreover, the calculation method is also suitable for other representative fullerene dimers, such as (C60)2 and its divalent anion. Additionally, the dissociation of (C59N)2 at 473 K under mass spectrometric conditions suggests the inter‐cage C—C bond is relatively weaker than an ordinary C—C single bond, which can be explained by the interaction energies of inter‐cages.  相似文献

Polyaniline (PANI)/Au composite nanotubes were synthesized and developed as an electrode material for a nicotinamide adenine dinucleotide (NADH) sensor. A MnO2 self-degradable template method was used to prepare the tube-like PANI nanomaterial. By introducing PANI nanotubes into Au colloid, Au nanoparticles (NPs) were successfully decorated onto the surface of PANI nanotubes through electrostatic effects. The morphology, composition, and optical properties of the resulting products were characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy, ultraviolet-visible (UV-vis) absorption spectra, and thermogravimetric analysis (TGA). In addition, the obtained PANI/Au composites were used as catalysts for the electrochemical oxidation of NADH. Cyclic voltammogram (CV) experiments indicated that PANI/Au-modified glassy carbon electrode showed a higher electrocatalytic activity towards the oxidation of NADH in a neutral environment. Differential pulse voltammogram (DPV) results illustrated that the fabricated NADH sensor had excellent anti-interference ability and displayed a wide linear range from 4?×?10?4 to 8?×?10?3 M with a detection limit of 0.5?×?10?7 M.  相似文献

Nickel-substituted layered perovskite PrBaCo2 ? x Ni x O5 + δ (PBCN) powders with various proportions of nickel (x?=?0, 0.1, 0.2, and 0.3, abbreviated as PBCN-0, PBCN-1, PBCN-2, and PBCN-3, respectively) are investigated as potential cathode materials for intermediate temperature solid oxide fuel cells (IT-SOFCs) based on the yttria-stabilized zirconia (YSZ) electrolyte. It is found that PBCN-1 has the highest electrical conductivity of 1,397 S cm?1 at 400 °C. Substitution of Co by Ni decreases the thermal expansion coefficient (TEC) clearly. The average TEC at the temperature range of 35–900 °C decreases from 22.8?×?10?6 K?1 for PBCN-0 to 18.9?×?10?6 K?1 for PBCN-3. The polarization resistances of PBCN samples on YSZ electrolyte at 800 °C are 0.053, 0.048, 0.052, and 0.042 Ω cm2 for PBCN-0, PBCN-1, PBCN-2, and PBCN-3, respectively. The single fuel cell with the configuration of PBCN-3/YSZ/Pt delivers the highest power densities of 100, 185, 360, 495, and 660 mW cm?2 at 600, 650, 700, 750, and 800 °C, respectively.  相似文献

The chemical fixation of CO2 under mild reaction conditions is of significance from a sustainable chemistry viewpoint. Herein a CO2‐reactive protic ionic liquid (PIL), [HDBU+][TFE?], was designed by neutralization of the superbase 1,8‐diazabicyclo[5.4.0]undec‐7‐ene (DBU) with a weak proton donor trifluoroethanol (TFE). As a bifunctional catalyst for simultaneously activating CO2 and the substrate, this PIL displayed excellent performance in catalyzing the reactions of CO2 with 2‐aminobenzonitriles at atmospheric pressure and room temperature, thus producing a series of quinazoline‐2,4(1H,3H)‐diones in excellent yields.  相似文献
