X-ray structures for six Fe(II) and Fe(III) complexes from two closely heptadentate N-tripodal ligands, L1H(3) = tris[(imidazol-4-yl)-3-aza-3-butenyl]amine and L2H(3) = tris[(imidazol-2-yl)-3-aza-3-butenyl]amine, are described: three complexes in the L1 series (namely, [Fe(II)(L1H(3))](2+) and [Fe(III)(L1H(3))](3+) at low pH and [Fe(III)(L1)](0) at high pH) and three complexes in the L2 series (namely, [Fe(II)(L2H(3))](2+) at low pH and [Fe(II)(L2H)](0) and [Fe(III)(L2)](0) at high pH). Most of these complexes are stable in both Fe(II) and Fe(III) redox states and with the ligand in various protonation states. In the solid state, hydrogen bonds networks were obtained. Structural differences induced by 2- or 4-imidazole substitution are described and discussed. In solution, interconversions between different forms, with regard to oxidation and protonation states, were investigated by UV-visible spectroscopy, cyclic voltammetry, and potentiometry. The deprotonation pattern of these polyimidazole iron(II) and iron(III) complexes is described in detail. pK(a)s of the imidazolate/imidazole moieties in MeOH/H(2)O are reported. Two new species, namely, [Fe(II)(L1)](-) and [Fe(II)(L2)](-), were shown to be obtained in DMSO upon strong base addition and characterized by UV-vis spectroscopy and cyclic voltammetry. Half-wave potentials of Fe(III)/Fe(II) complexes with ligand moieties in several protonation states are reported, both in DMSO and in MeOH/H(2)O. Because of the presence of free imidazole groups coordinated to the iron, the potential of the iron(III)/iron(II) couples can be tuned by pH. A shift of DeltaE = E(deprot) - E(prot) ranging from -270 to -320 mV per exchanged proton in DMSO was measured. This study shows moreover that interconversions (with regard to both redox and protonation states) can be reversed several times. As the complexes have been isolated in order to be tested as superoxide dismutase mimics, preliminary reactions with dioxygen and with superoxide, considered as oxidant and reducer of biological importance, are reported. In these two series, O(2)(-) behaves either as a base or as a reducer and no adducts have been observed.  相似文献

Abstract— Experiments are described that enable the kinetic behavior of singlet oxygen, O2(IΔg), to be monitored in the time-resolved mode using a photomultiplier to detect deep orange light (γmax 660 nm). Reaction of lithiated methoxyallene, 1-ethoxyethoxyallene, 1-(methylthio)propyne and 2-butyne with methoxymethyl isothiocyanate, MeOCH2N=C=S followed by methylation affords the imidothioates H2C=C=C(R)C(SMe)=NCH2OMe [R=Me, OMe, OCH(Me)OEt, SMe]. Rearrangement to the fully conjugated systems H2C=CH---C(R)=C(SMe)---N=CHOMe and subsequent electrocyclisation of these compounds leads to the 5-substituted 6-(methylthio)-2-methoxy-2,3-dihydropyridines with good to excellent yields. In the presence of acidic catalysts or by heating at elevated temperatures these dihydropyridines eliminate methanol to afford 3-substituted 2-(methylthio)pyridines. The aroma compound 2-(methylthio)-3-pyridinol was obtained by acid-catalysed treatment of 3-(1-ethoxyethoxy)-2-(methylthio)pyridine.  相似文献

A procedure that generates random conformations of a protein chain, and then applies energy minimization to find the structure of lowest energy, is described. Single-residue conformations are represented in terms of four conformational states, α, ?, α*, and ?*. Each state corresponds to a rectangular region in the ?, ψ map. The conformation of an entire chain is then represented by a sequence of single-residue conformational states. The distinct “chain-states” in this representation correspond to multidimensional rectangular regions in the conformational space of the whole protein. A set of highly-probable chain-states can be predicted from the amino acid sequence using the pattern recognition procedure developed in the first two articles of this series. The importance-sampling minimization procedure of the present article is then used to explore the regions of conformational space corresponding to each of these chain-states. The importance-sampling procedure generates a number of random conformations within a particular multidimensional rectangular region, sampling most densely from the most probable, or “important,” sections of the ?, ψ map. All values of ? and ψ are allowed, but the less-probable values are sampled less often. To achieve this, the random values of ? and Φ are generated from bivariate gaussian distributions that are determined from known X-ray structures. Separate gaussian distributions are used for proline residues in the α and ? states, for glycine residues in the α, ?, α*, and ?* states, and for ordinary residues involved in 29 different tripeptide conformations. Energy minimization is then applied to the randomly-generated structures to optimize interactions and to improve packing. The final energy values are used to select the best structures. The importance-sampling minimization procedure is tested on the avian pancreatic polypeptide, using chain-states predicted from the amino acid sequence. The conformation having the lowest energy is very similar to the X-ray conformation.  相似文献

Summary A series of -cyclodextrin complexes containing various guest molecules was studied using computer-aided molecular modeling and conformation analysis techniques. The geometry of each complex was studied using crystallographic data. The positions of the glycosidic O4 atoms indicate that the -cyclodextrin molecules are elliptically distorted. This distortion can be related to the van der Waals volume of the guest molecules. This correlation is different for aromatic and non-aromatic guest compounds. Rigid body docking experiments demonstrated that in crystal structures the guest molecule occupies a position in the cavity of nearly minimum interaction energy when there are no other molecules having interactions with the guest molecule. From the crystallographic data several rules could be deduced which seem to determine the conformation of -cyclodextrin molecules in complexes. A procedure was developed to construct -cyclodextrin molecules that are able to encompass guest molecules having a given van der Waals volume.  相似文献

Arsenic compounds are ubiquitous and widespread in the environment as a result of natural or anthropogenic occurrence. Fish are the major source of protein for human consumption. They are also a source of contamination, because of the amounts of heavy elements they can contain, some of which are highly toxic. Fish bones are high in calcium, which is an essential mineral for normal body function. It consists of water, organic material, and mineral matter. Chelating agents have been used clinically as antidotes for acute and chronic metal intoxications. In the present study, an attempt is made to investigate the bio-accumulation of arsenic and its effect on the biochemical and mineral contents of Labeo rohita bones using, Fourier transform infrared (FT-IR) spectroscopy. The results of the present study indicate that arsenic exposure induces significant reduction on the biochemical and mineral contents of the L. rohita bones. Further, the DMSA treatment significantly improves these levels. This shows that DMSA is an effective chelator for arsenic toxicity. Quantitative curve-fitting analyses of amide I band have proved useful in studying the nature and the extent of protein conformational changes. A decrease in α-helical and random coil structures and an increase in β-sheet structures have been observed due to arsenic exposure. In conclusion, the present study shows that the FT-IR spectroscopy coupled with second derivative and curve-fitting analysis gives useful information about the biochemical and mineral contents of the L. rohita bones.  相似文献

Zinc is an essential metal for different physiological functions and becomes toxic when elevated concentrations are introduced into the environment. In the present study, an attempt is made to analyze zinc-induced biochemical changes in the liver tissues of freshwater fingerlings of Labeo rohita using Fourier Transformation Infrared Spectroscopy. Several important features have been observed in the FTIR spectra of zinc-intoxicated liver tissues, namely, altered membrane lipid, altered protein profile, and increased glycogen content, indicating an alteration in the lipid and protein profiles leading to modification in membrane composition. Further, it is observed that acute exposure to zinc causes some alteration in protein profile with a decrease in α-helix and an increase in random coil structure. Treatment with the chelating agent D-penicillamine reduces the biochemical contents in the liver tissues. This shows that D-penicillamine is a good antidote for zinc toxicity.
