The proportionately low abundance of membrane proteins hampers their proteomic analysis, especially for a quantitative LC‐MS/MS approach. To overcome this limitation, a method was developed that consists of one cell disruption step in a hypotonic reagent using liquid nitrogen, one isolation step using a low speed centrifugation, and three wash steps using high speed centrifugation. Pellets contained plasma, nuclear, and mitochondrial membranes, including their integral, peripheral, and anchored membrane proteins. The reproducibility of this method was verified by protein assay of four separate experiments with a CV of 7.7%, and by comparative LC‐MS/MS label‐free quantification of individual proteins between two experiments with 99% of the quantified proteins having a CV ≤30%. Western blot and LC‐MS/MS results of markers for cytoplasm, nucleus, mitochondria, and their membranes indicated that the enriched membrane fraction was highly pure by the absence of, or presence of trace amounts of, nonmembrane marker proteins. The average yield of membrane proteins was 237 μg/10 million HT29‐MTX cells. LC‐MS/MS analysis of the membrane‐enriched sample resulted in the identification of 2597 protein groups. In summary, the developed method is reproducible, produces a highly pure membrane fraction, and generates a high yield of membrane proteins.  相似文献

Bare (unmodified) and crown ether (CE)-modified Fe3O4 magnetic nanoparticles (MNPs) were investigated for the rapid extraction of 226Ra from water samples. It involved synthesizing the MNPs, introducing them into the sample solutions, ultrasonicating and agitating the suspension, magnetically separating the nanoparticles from solution, and measuring the 226Ra content in the supernatant. Experimental parameters such as salt choice, salt concentration and pH were optimized to achieve maximum extraction of 226Ra onto the MNPs. 226Ra content was determined using a Hidex 300SL liquid scintillation counter with α/β separation capability, or a gamma spectrometric detection system. The bare Fe3O4 nanoparticles showed significant pH dependence for the extraction of 226Ra from an aqueous solution over a pH range of 2–10. They gave an extraction of 95 ± 1 and 98 ± 1 % at pH 9 in 0.1 M NaCl and 0.1 M NaClO4, respectively, whereas an extraction of 8–24 % was obtained, over the pH ranges from 2 to 5. The CE-modified MNPs yielded extraction efficiencies as high as 99 ± 1 % in the presence of 0.01 M picric acid at pH 4. This study demonstrates that the surface functionalization of Fe3O4 MNPs with suitable ligand modification can offer a selective mode of extraction for 226Ra in the presence of its daughter progenies.  相似文献

A series of transition metal (Ni, Cu, Pd) complexes derived from macrocyclic tetrabenzo[b,f,j,n] [1,5,9,13]tetraazacyclohexadecine (TAAB) was synthesized and their mesomorphic properties studied by differential scanning calorimetry, polarized optical microscopy and X-ray powder diffraction (XRD). These compounds have eight alkoxy side chains attached around the central molecular core and form disc-like molecules. All the derivatives exhibited columnar mesophases over a wide range of temperature. The mesomorphic behaviour was found to be dependent on the incorporated metal and the carbon length of the alkoxy side chains. The clearing temperatures decreased in the order M = Ni > Pd > Cu; this decrease was probably due to the size of the metal ions. Some derivatives with shorter side chains (n = 10, 12) were room temperature liquid crystals. All compounds were found to exhibit hexagonal columnar (Colh) phases which were confirmed by powder XRD.  相似文献

Supramolecular side chain liquid crystalline polymers were prepared from poly(3-carboxypropylmethylsiloxane) (PSI100) and azobenzene-based derivatives through intermolecular hydrogen bonding between the carboxylic acid groups of PSI100 and the imidazole rings in the azobenzene-based derivatives. The presence of H-bonding was confirmed using FTIR spectroscopy. The polymeric complexes behave as liquid crystalline polymers and exhibit nematic mesophases identified on the basis of the observation of Schlieren textures. The mesogenic behaviour of these complexes was studied by polarizing optical microscopy and X-ray diffraction. The thermal behaviour of the complexes was investigated by differential scanning calorimetry. On increasing the spacer length, the transition temperatures initially increase. A further increase in spacer length, however, leads to a decrease in the transition temperatures. The electron donor-acceptor interaction between unlike mesogenic units in supramolecular copolymeric complexes helps to stabilize the mesophase.  相似文献

The synthesis, characterization and mesomorphic properties of a new type of liquid crystalline compound, the 2,5-bis(3,4,5-trialkoxyphenyl)-1,3,4-oxadiazoles, 3a-3h, are reported. These heterocyclic compounds are derived from unsaturated 1,3,4-oxadiazole as the core group, and obtained by the condensation reaction of 3,4,5-trialkoxybenzoic acid N-(3,4,5-trialkoxybenzoyl)-hydrazides and phosphorus oxychloride in toluene under reflux. All compounds were characterized by 1H and 13C NMR spectroscopy, and elemental analysis. The mesomorphic properties of these and the related compounds 1, 2 were characterized and studied by differential scanning calorimetry and polarizing optical microscopy. The formation of columnar mesophases was found to be dependent on the numbers of alkoxy sidechains. The compounds 3 exhibited hexagonal columnar (Colh) phases, however compounds 1, 2 formed crystalline phases. Compounds 3b-3e with shorter carbon chains were room temperature liquid crystals. Polar induction by nitrogen and/or oxygen atoms on the heterocyclic core ring might be responsible for the formation and better observed mesomorphic properties in this type of compound.  相似文献

A new catalytic solid substrate-room temperature phosphorimetry (SS-RTP) for the determination of trace arsenic(V) has been established. It is based on the fact that fullerenol (F-ol) emitted strong and stable room temperature phosphorescence (RTP) on nitric acid cellulose membrane (NCM) substrate. H2O2 could oxidise F-ol to cause the quenching of RTP. As(V) could catalyse H2O2 to oxidise F-ol and decrease the RTP signal of F-ol sharply. After adding tween-80 in the system, its ΔI p enhanced 7.7 times compared with the without-tween-80 levels. Under the optimum conditions, the linear dynamic range of this method was 0.016?11.2 ag spot?1 with a detection limit (LD) of 9.3 zg spot?1 (corresponding concentration: 2.3 × 10?17 g mL?1). This sensitive, simple and selective method has been successfully applied to the determination of trace As(V) in human hair and tea samples. The reaction mechanism for SS-RTP is also discussed.  相似文献

以溴代正烷烃、乙二胺、1,3-丙磺内酯合成了一系列新型磺酸盐型Gemini表面活性剂(SGS-8、SGS-10、SGS-12、SGS-14);采用IR、1H NMR及元素分析等手段对合成产物进行结构表征,并研究了其表面活性,采用等温滴定微量热技术、表面张力法和稳态荧光光谱法研究了SGS-12与聚丙烯酰胺(PAM)的相互作用规律及热力学参数。结果表明,合成的产物纯度较高,具有较低的cmc值和较好的表面活性;SGS-12/PAM混合体系的表面张力曲线和芘探针在体系中微极性的变化表明,二者发生相互作用且形成混合胶束;SGS-12与PAM结合是一个由焓驱动的自发过程,△H和△S均小于0,表明相互作用力以氢键和范德华力为主,合成产物在聚丙烯酰胺分子上的平均结合数为287。  相似文献

Three unconventional dendrimers that contained rigid NH? triazine linkages and peripheral tert‐butyl moieties were prepared by using a convergent approach and characterized by 1H and 13C NMR spectroscopy, mass spectrometry, and elemental analysis. Based on a thermogravimetric analysis study, these dendrimers were observed to display thermal stability at about 300 °C. The NH? triazine moiety, which possessed protonated and proton‐free nitrogen sites (like the imidazole unit), displayed the capture of polarizable CO2 molecules through hydrogen‐bond and/or dipole–quadrupole interactions. In addition, the adsorption of various amounts of CO2 and N2 at different pressures suggests that the dendritic pores, which arise from the stacking of the middle co‐planar and rim protuberant dendrimers, G n ‐N~N‐G n (n=1–3), either swell or shrink at high pressure, thus indicating that these dendrimers may have a breathing ability.  相似文献

Herein, we report a “threading followed by shrinking” approach for the synthesis of rotaxanes by using an “oxygen‐deficient” macrocycle that contained two arylmethyl sulfone units and the dumbbell‐shaped salt bis(3,5‐dimethylbenzyl)ammonium tetrakis(3,5‐trifluoromethylphenyl)borate as the host and guest components, respectively. The extrusion of SO2 from both of the arylmethyl sulfone units of the macrocyclic component in the corresponding [2]pseudorotaxane resulted in a [2]rotaxane that was sufficiently stable to maintain its molecular integrity in CD3SOCD3 at 393 K for at least 5 h.  相似文献

We demonstrate that surface‐induced dissociation (SID) coupled with ion mobility mass spectrometry (IM‐MS) is a powerful tool for determining the stoichiometry of a multi‐subunit ribonucleoprotein (RNP) complex assembled in a solution containing Mg2+. We investigated Pyrococcus furiosus (Pfu) RNase P, an archaeal RNP that catalyzes tRNA 5′ maturation. Previous step‐wise, Mg2+‐dependent reconstitutions of Pfu RNase P with its catalytic RNA subunit and two interacting protein cofactor pairs (RPP21⋅RPP29 and POP5⋅RPP30) revealed functional RNP intermediates en route to the RNase P enzyme, but provided no information on subunit stoichiometry. Our native MS studies with the proteins showed RPP21⋅RPP29 and (POP5⋅RPP30)2 complexes, but indicated a 1:1 composition for all subunits when either one or both protein complexes bind the cognate RNA. These results highlight the utility of SID and IM‐MS in resolving conformational heterogeneity and yielding insights on RNP assembly.  相似文献
