Detachment of heme prosthetic groups from gaseous myoglobin ions has been studied by collision-induced dissociation and infrared multiphoton dissociation in combination with Fourier-transform ion cyclotron resonance mass spectrometry. Multiply charged holomyoglobin ions (hMbn+) were generated by electrospray ionization and transferred to an ion cyclotron resonance cell, where the ions of interest were isolated and fragmented by either collision with Ar atoms or irradiation with 3 mum photons, producing apomyoglobin ions (aMbn+). Both charged heme loss (with [Fe(III)-heme]+ and aMb(n-1)+ as the products) and neutral heme loss (with [Fe(II)-heme] and aMbn+ as the products) were detected concurrently for hMbn+ produced from a myoglobin solution pretreated with reducing reagents. By reference to Ea = 0.9 eV determined by blackbody infrared radiative dissociation for charged heme loss of ferric hMbn+, an activation energy of 1.1 eV was deduced for neutral heme loss of ferrous hMbn+ with n = 9 and 10. 相似文献

Several syn and anti atropisomers of 2-(5-benzo[b]fluorenyl)-2′-hydroxy-1,1′-binaphthyl and related compounds were synthesized from 1,1′-binaphthyl-2,2′-diol (BINOL). It was possible to separate the syn and anti atropisomers by silica gel column chromatography. The syn atropisomers are potential hetero-bidentate ligands for complex formation with metals. By starting from enantiomerically pure (R)-(+)-BINOL and (S)-(−)-BINOL, four optically active syn atropisomers and two anti atropisomers with high enantiomeric purity were obtained. In this study, heparin was immobilized onto cellulose acetate hollow fibers to improve the anticoagulation performance during hemodialysis. In vitro evaluation was carried out using mini‐hemodialyzer circulating with fresh porcine whole blood to simulate kidney therapy. The dialysis performance and hemocompatibility were estimated. The results showed that heparinized hemodialyzer could be used through out the whole dialysis time (4 hr) without injecting additional heparin to prevent coagulation in the dialysis system. In addition, the hemocompatibility was evaluated by measuring activated partial thromboplastin time (APTT), prothrombin time (PT), and fibrinogen time (FT). The complete blood count (CBC) including red blood cell (RBC), hemoglobin (Hgb), hematocrit (Hct), white blood cell (WBC), and platelet were determined. The results showed that heparinization could keep the CBC stable during dialysis, whereas unmodified cellulose acetate hemodialyzer would cause a decrease in RBC unless heparin was injected during dialysis. Heparinized hemodialyzer showed longer APTT, PT, and FT than unmodified hemodialyzer. Heparinized hemodialyzer also showed slightly higher clearance than unmodified hemodialyzer. These results indicated that the dialysis performance and hemocompatibility of cellulose acetate hemodialyzer could be improved by the immobilization of heparin. High‐resolution angular and velocity distributions for neutral analytes (tryptophan and poly‐tryptophan) and matrix (2,4,6‐trihydroxyacetophenon, THAP) are measured by using 355 nm laser desorption. The information suggests that two separate mechanisms dominate the angular and velocity distributions at the beginning and before the end of desorption. A molecular jet‐like isentropic expansion dominates the plume expansion at the beginning of desorption. This only occurs at high surface temperature, thus resulting in a large velocity normal to the surface and a very narrow angular distribution. Most of the analytes are produced under these conditions. Before the end of desorption, the surface temperature decreases and the mechanism of thermal desorption at low vapor pressure takes over. The velocities become small and the angular distribution is close to cosθ. Only a very small amount of analytes are generated under these conditions. Compared to tryptophan, poly‐tryptophan has a much narrower angular distribution, thereby suggesting that it is only produced at the higher surface temperatures. 相似文献

Biodiesel was synthesized from soybean oil by transesterification over Li2SiO3 catalyst.The Li2SiO3 can be used for biodiesel production directly without further drying or thermal pretreatment,no obvious difference in the FAME conversion(92.4-96.7%) between the air-exposed catalyst(24-72 h) and the fresh one(94.2%).This leads to important benefits when considering industrial applications of Li2SiO3 as a solid catalyst for storing and handling catalyst without taking special actions. 相似文献

This paper compares rates of charge transport by tunneling across junctions with the structures AgTSX(CH2)2nCH3 //Ga2O3 /EGaIn (n=1–8 and X= ? SCH2? and ? O2C? ); here AgTS is template‐stripped silver, and EGaIn is the eutectic alloy of gallium and indium. Its objective was to compare the tunneling decay coefficient (β, Å?1) and the injection current (J0, A cm?2) of the junctions comprising SAMs of n‐alkanethiolates and n‐alkanoates. Replacing AgTSSCH2‐R with AgTSO2C‐R (R=alkyl chains) had no significant influence on J0 (ca. 3×103 A cm?2) or β (0.75–0.79 Å?1)—an indication that such changes (both structural and electronic) in the AgTSXR interface do not influence the rate of charge transport. A comparison of junctions comprising oligo(phenylene)carboxylates and n‐alkanoates showed, as expected, that β for aliphatic (0.79 Å?1) and aromatic (0.60 Å?1) SAMs differed significantly. 相似文献

Transthyretin (TTR) is a homotetrameric plasma protein associated with human amyloid diseases. Although Tafamidis has been recently approved for the treatment of TTR familial amyloid polyneuropathy (FAP), there is still a need for more effective drugs in the treatment of TTR amyloidosis diseases. In this study, a computer‐aided approach combining molecular docking, virtual screening and molecular dynamics (MD) simulations was employed to identify potent TTR amyloidosis inhibitors from National Cancer Institute (NCI), Maybridge and Asdi databases. A receptor‐specific scoring function was also developed using comparative binding energy (COMBINE) method to accurately predict the inhibitory activities for the selected compounds during virtual screening. The developed receptor‐specific scoring function demonstrated good predictive ability by yielding strong correlation coefficients between experimental activities and estimated activities for 32 training set and 9 test set compounds, respectively. Moreover, it was successfully applied to rank the candidate compounds from structure‐based virtual screening. Finally, three compounds (NSC220163, MFCD00276817 and SPB06319) were identified as potential leads, which exhibited higher predicted inhibitory activities and higher binding affinities in comparison to the Tafamidis. Our results further suggest that halogen bonding interaction plays a crucial role in stabilizing the TTR‐inhibitor complex. These results indicate that our computational approach could effectively discover more potent TTR amyloidosis inhibitors, which can be further validate by in vitro and in vivo biological tests. 相似文献

New gelators for urea-containing triazine derivatives were synthesised, and their gelation potential was examined using different organic solvents. These compounds were found to form the organogels with a variety of organic solvents, such as hexane and other solvents. The elongated alkyl tails of the gelators displayed an obvious decrease in the critical gelation concentrations of apolar solvents and an increase in the compatibility of gelation in polar solvents. The resulting thermo-reversible gels were characterised by using the dropping ball method and a number of other instruments. The melting temperature (Tm) of the gels in decalin and CCl4 increased with the gelator concentrations. The intermolecular hydrogen bonding of gelation in different organic solvents was observed using an FT-IR spectrometer. Temperature-dependent UV–vis and fluorescence analysis showed that the organogels displayed diverse aggregations and various fluorescence effects in different organic solvents. Blue fluorescence and J-aggregation in decalin and the quenched effect and π–π stacking in CCl4 were observed. Further, the morphological self-assembled feature in different organic solvents was studied with a scanning electron microscope, and the morphological features demonstrated that there were different aggregations in different solvents. In conductivity electrolyte experiments, the organogel electrolytes exhibited high conductivity (σ) compared with the corresponding tetrabutylammonium perchlorate (TBAP)/THF solution. The conductivity of the gel electrolytes increased with the concentration of the electrolyte salts and temperature. When the sol–gel temperature was achieved, a high ion conductivity was observed compared with the corresponding TBAP/THF solution. When the ratio of the added electrolyte salts exceeded 5%, gelation was inhibited. Furthermore, the effect of the electrolyte salts on the Tm of the gel was confirmed. Safranine, a cationic dye, was removed from synthetic wastewater by ion flotation. Over 98% of safranine was removed from the solution in 10 min. A stoichiometric amount of surfactant (1 mol of surfactant to 1 mol of dye) was found to be most effective for safranine removal. The separation efficiency of safranine decreased with increasing concentration of NaNO3. Safranine was also removed by adsorbing colloid flotation technique using Fe(OH)3 as the coagulant. Sodium lauryl sulfate was used as the collector, and over 97% of safranine was removed in 5 min. The separation efficiency decreased with increasing ionic strength of the solution. The deleterious effect of neutral salt was compensated somewhat with the aid of Al^3 as the activator. Both ion flotation and adsorbing colloid flotation may be applicable in the removal of safranine from wastewater. 相似文献
