The recently published crystal structure of the D3 dopamine receptor shows a tightly packed region of aromatic residues on helices 5 and 6 in the space bridging the binding site and what is thought to be the origin of intracellular helical motion. This highly conserved region also makes contacts with residues on helix 3, and here we use double mutant cycle analysis and unnatural amino acid mutagenesis to probe the functional role of several residues in this region of the closely related D2 dopamine receptor. Of the eight mutant pairs examined, all show significant functional coupling (Ω > 2), with the largest coupling coefficients observed between residues on different helices, C3.36/W6.48, T3.37/S5.46, and F5.47/F6.52. Additionally, three aromatic residues examined, F5.47, Y5.48, and F5.51, show consistent trends upon progressive fluorination of the aromatic side chain. These trends are indicative of a functionally important electrostatic interaction with the face of the aromatic residue examined, which is likely attributed to aromatic-aromatic interactions between residues in this microdomain. We also propose that the previously determined fluorination trend at W6.48 is likely due to a sulfur-π interaction with the side chain of C3.36. We conclude that these residues form a tightly packed structural microdomain that connects helices 3, 5, and 6, thus forming a barrier that prevents dopamine from binding further toward the intracellular surface. Upon activation, these residues likely do not change their relative conformation, but rather act to translate agonist binding at the extracellular surface into the large intracellular movements that characterize receptor activation. 相似文献

Selenium Polycations Stabilized by Polymeric Chlorobismuthate Anions: Syntheses and Crystal Structures of Se4[Bi4Cl14] and Se10[Bi5Cl17] Reactions of selenium with selenium(IV) chloride and bismuth(III) chloride in sealed evacuated glass ampoules at temperatures between 110 and 155 °C yield a series of compounds which are composed of discrete selenium polycations and polymeric chlorobismutate anions. Besides the already known Se8[Bi4Cl14] two new compounds have been identified by crystal structure analyses as Se4[Bi4Cl14] (tetragonal, P4/n, a = 1089.1(2) pm, c = 993.7(2) pm, Z = 2) and Se10[Bi5Cl17] (monoclinic, P21/c, a = 1079.24(8) pm, b = 2062.9(2) pm, c = 1676.1(2) pm, β = 90.87(1)°, Z = 4). Se4[Bi4Cl14] was obtained as red transparent platelike crystals and is the first example of a compound with (chalcogen4)2+ ions of exact square‐planar symmetry and molecular point group D4h in the solid state. The cations are surrounded by layers of two‐dimensional polymeric anions [Bi4Cl14]2–. Se10[Bi5Cl17] forms dark grey crystals with a reddish luster. The structure contains the known bicyclic polycation Se102+ which is disordered over two positions and the first three‐dimensional polymeric chlorobismutate anion [Bi5Cl17]2–. The rotation of the carboxylate groups in DOTA (DOTA = 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetate) complexes of several lanthanide ions and Sc(3+) was investigated with density functional theory (DFT) calculations and with variable temperature (17)O NMR studies at 4.7-18.8 T. The data obtained show that the rotation is much slower than the other dynamic processes taking place in these complexes. The exchange between the bound and unbound carboxylate oxygen atoms for the largest Ln(3+) ions (La(3+)→Sm(3+)) follows a pathway via a transition state in which both oxygens of the carboxylate group are bound to the Ln(3+) ion, whereas for the smaller metal ions (Tm(3+), Lu(3+), Sc(3+)) the transition state has a fully decoordinated carboxylate group. The activation free energies show a steady increase from about 75 to 125-135 kJ·mol(-1) going from La(3+) to Lu(3+). This computed trend is consistent with the results of the (17)O NMR measurements. Fast exchange between bound and unbound carboxylate oxygen atoms was observed for the diamagnetic La-DOTA, whereas for Pr-, Sm-, Lu-, and Sc-DOTA the exchange was slow on the NMR time scale. The trends in the linewidths for the various metal ions as a function of the temperature agree with trends in the rates as predicted by the DFT calculations. 相似文献

Understanding the crystallization of calcium carbonate is relevant in numerous fields like biomineralization, geology and industrial applications where calcium carbonate forms. In order to enhance the knowledge about the formation of calcium carbonate with focus on the vaterite polymorph, in this work calcium carbonate has been crystallized in aqueous solutions at temperatures from 5 °C to 90 °C. Special attention has been directed to higher temperatures for which the effect of the initial supersaturation on the resulting crystal morphologies and the onset of dendritic growth have been studied. In analogy to snow crystal formation, it has been found that in a certain temperature range hexagonal plate‐like crystals form at low supersaturation whereas dendritic patterns start to appear at higher supersaturation. The symmetrical branches characteristic for dendritic growth get less pronounced as the temperature is decreased. The results reported here related to the interdependence between supersaturation, crystal morphology and growth mechanisms, can be used in future works to predict particle formation and to design crystal architectures. A quantitative analysis of long‐range order in the self‐organized porous structure of anodic alumina films has been performed on the basis of a microradian X‐ray diffraction study. The structure is shown to possess orientational order over macroscopic distances larger than 1 mm. At the same time, the interpore positional order is only short‐range and does not extend over more than ∼10 interpore distances. These positional correlations are mostly lost gradually rather than at the domain boundaries, as suggested by the divergence of the peak width for the higher‐order reflections. In the direction of the film growth the pores have a very long longitudinal self‐correlation length of the order of tens of micrometres. 相似文献

The 16 O+ 15 N reaction products have been studied by the γ-ray detection method in the CM energy range 15.5 to 36.1 MeV and by the kinematical coincidence method at energies ranging from E CM =20.6 to 33.5MeV. The γ-ray yield excitation function of the 16O 3? inelastic channel shows the existence of resonant structures. Two structures with ~ 1.6 MeV width are observed in the large angle elastic scattering excitation function, they are correlated with the resonances seen in the inelastic channel. An airborne laser system for remote sensing has been developed and tested at flight altitudes from 500 to 1200 m. The system
is based on two continuous-wave CO2 lasers (output 3 W), tunable to different wavelengths, and detects the laser radiation scattered back from earth by heterodyne
reception. The feasibility of the heterodyne reception technique under aircraft environment conditions was demonstrated, and
man-made SF6 clouds and different types of uncovered minerals were traced. The data obtained indicate that the measuring range of the
system can be increased to satellite distance by using a larger telescope and a more powerful laser and by reducing the electrical
bandwidth.
