In the course of our screening program for regulators of a molecular chaperone GRP78 expression, we isolated a novel inhibitor of GRP78 expression, designated as prunustatin A, from Streptomyces violaceoniger 4521-SVS3. The planar structure of prunustatin A was determined to be an oxidized type of the neoantimycin family. Its absolute stereochemistry was established to be 2R, 4S, 6S, 7R, 9S, and 29S by analyzing chemically degraded components obtained from the derivative of prunustatin A. 相似文献

The photochemistry of fac‐[Re(bpy)(CO)3Cl] ( 1 a ; bpy=2,2′‐bipyridine) initiated by irradiation using <330 nm light has been investigated. Isomerization proceeded in THF to give the corresponding mer‐isomer 1 b . However, in the presence of a small amount of MeCN, the main product was the CO‐ligand‐substituted complex (OC‐6‐24)‐[Re(bpy)(CO)2Cl(MeCN)] ( 2 c ; bpy=2,2′‐bipyridine). In MeCN, two isomers, 2 c and its (OC‐6‐34) form ( 2 a ), were produced. Only 2 c thermally isomerized to produce the (OC‐6‐44) form 2 b . A detailed investigation led to the conclusion that both 1 b and 2 c are produced by a dissociative mechanism, whereas 2 a forms by an associative mechanism. A comparison of the ultrafast transient UV‐visible absorption, emission, and IR spectra of 1 a acquired by excitation using higher‐energy light (e.g., 270 nm) and lower‐energy light (e.g., 400 nm) gave detailed information about the excited states, intermediates, and kinetics of the photochemical reactions and photophysical processes of 1 a . Irradiation of 1 a using the higher‐energy light resulted in the generation of the higher singlet excited state with τ≤25 fs, from which intersystem crossing proceeded to give the higher triplet state (3HES( 1 )). In THF, 3HES( 1 ) was competitively converted to both the triplet ligand field (3LF) and metal‐to‐ligand charge transfer (3M LCT) with lifetimes of 200 fs, in which the former is a reactive state that converts to [Re(bpy)(CO)2Cl(thf)]+ ( 1 c ) within 10 ps by means of a dissociative mechanism. Re‐coordination of CO to 1 c gives both 1 a and 1 b . In MeCN, irradiation of 1 a by using high‐energy light gives the coordinatively unsaturated complex, which rapidly converted to 2 c . A seven‐coordinate complex is also produced within several hundred femtoseconds, which is converted to 2 a within several hundred picoseconds. 相似文献

The highly selective mono-C-allylation of oligoglycinates such as a diethylenetriaminepentaacetate, an iminodiacetate, and an ethylenediaminetetraacetate via insertion of a vacuum operation between the N-allylation and C-migration steps is reported. It is contrastive that one-pot N-allylation-C-allylation procedure gave a mixture including multiallylated products. In the reaction with N-ylides, gem-C-diallylation and α,α'-C-diallylation of oligoglycinates are strongly inhibited even with the use of an excess of allyl bromide and base. A mechanism to explain this control of the frequency of C-allylation on oligoglycinates via N-ylides is also proposed. 相似文献

This work reports the design of a highly sensitive solid-state sensor device based on a water-gated organic thin-film transistor (WG-OTFT) for the selective detection of herbicide glyphosate (GlyP) in water. A competitive assay among carboxylate-functionalized polythiophene, Cu2+, and GlyP was employed as a sensing mechanism. Molecular recognition phenomena and electrical double layer (EDL) (at the polymer/water interface) originated from the field-effect worked cooperatively to amplify the sensitivity for GlyP. The limit of detection of WG-OTFT (0.26 ppm) was lower than that of a fluorescence sensor chip (0.95 ppm) which is the conventional sensing method. In contrast to the previously reported insulated molecular wires to block interchain interactions, molecular aggregates under the field-effect has shown to be effective for amplification of sensitivity through “intra”- and “inter”-molecular wire effects. The opposite strategy in this study could pave the way for fully utilizing the sensing properties of polymer-based solid-state sensor devices. 相似文献

We examined the intradermal delivery of a hydrophilic polyphenol chlorogenic acid by in vitro study using excised guinea pig dorsal skin and Yucatan micropig skin. Skin accumulation as well as the solubility of chlorogenic acid in aqueous vehicles was much greater than for other polyphenols such as quercetin and genistein. However, since enhancement of skin delivery seemed to be necessary to exhibit its protective effects against oxidative damage of skin, we examined the effects of microemulsions as vehicles. Using microemulsions consisting of 150 mM NaCl solution, isopropyl myristate, polyoxyethylene sorbitan monooleate (Tween 80) and ethanol, skin accumulation as well as solubility of chlorogenic acid further increased. Enhancement effect of an oil-in-water (o/w-type) microemulsion was greater than that of a water-in-oil (w/o-type) microemulsion possibly due to the greater increase in solubility. This finding was quite different from previous findings on relatively hydrophobic polyphenols such as quercetin and genistein. Pretreatment of guinea pig dorsal skin with chlorogenic acid containing microemulsion gel prevented erythema formation induced by UV irradiation. These findings indicate the potential use of hydrophilic chlorogenic acid with o/w-type microemulsion as a vehicle to protect skin against UV-induced oxidative damage. 相似文献

Combinations of electron-withdrawing and -donating substituents on the 8-hydroxyquinoline ligand of the tris(8-hydroxyquinoline)aluminum (Alq(3)) complexes allow for control of the HOMO and LUMO energies and the HOMO-LUMO gap responsible for emission from the complexes. Here, we present a systematic study on tuning the emission and electroluminescence (EL) from Alq(3) complexes from the green to blue region. In this study, we explored the combination of electron-donating substituents on C4 and C6. Compounds 1-6 displayed the emission tuning between 478 and 526 nm, and fluorescence quantum yield between 0.15 and 0.57. The compounds 2-6 were used as emitters and hosts in organic light-emitting diodes (OLEDs). The highest OLED external quantum efficiency (EQE) observed was 4.6%, which is among the highest observed for Alq(3) complexes. Also, the compounds 3-5 were used as hosts for red phosphorescent dopants to obtain white light-emitting diodes (WOLED). The WOLEDs displayed high efficiency (EQE up to 19%) and high white color purity (color rendering index (CRI≈85). 相似文献

A new imaging method is proposed for ultrathin films with a thickness of a few nanometers, based on the anomalous reflection (AR) of gold. In the AR effect, the reflectivity fairly decreases for blue or purple light (380 nm < λ < 480 nm) with the existence of a transparent dielectric layer at a gold surface. Thus, a thin gold film can be used as an imaging platform. Clear AR images are obtained for a microarray of protein (avidin) spots of diameter 120 μm with gaps of size 50 μm between the spots (36 spots/mm2). The resolution of the AR imaging is governed solely by the illumination spot size. AR imaging is a promising technique for high throughput analysis of biomolecular detection in a microarray format. 相似文献
