Coarse grained Langevin-type effective field equations may provide some guidance for the analysis of mesoscopic or microscopic molecular systems exhibiting fluctuations, or for systems of hundreds to thousands of atomic or subatomic particles produced in atomic or high-energy nuclear collisions. Trace element concentrations in honey collected from a wide area can be used as an environmental indicator, but sample preparation methods prior to analysis can have large effects on the results. The efficiency and reproducibility of sample preparation methods (dilution, ashing and digestion) for simultaneous multi-element analysis have been compared for 13 elements. Digestion in a PTFE bomb was found to be the most reliable method.  相似文献

Human kinesin spindle protein (KSP)/hsEg5, a member of the kinesin-5 family, is essential for mitotic spindle assembly in dividing human cells and is required for cell cycle progression through mitosis. Inhibition of the ATPase activity of KSP leads to cell cycle arrest during mitosis and subsequent cell death. Ispinesib (SB-715992), a potent and selective inhibitor of KSP, is currently in phase II clinical trials for the treatment of multiple tumor types. Mutations that attenuate Ispinesib binding to KSP in vitro have been identified, highlighting the need for inhibitors that target different binding sites and inhibit KSP activity by novel mechanisms. We report here a small-molecule modulator, KSPA-1, that activates KSP-catalyzed ATP hydrolysis in the absence of microtubules yet inhibits microtubule-stimulated ATP hydrolysis by KSP. KSPA-1 inhibits cell proliferation and induces monopolar-spindle formation in tumor cells. Results from kinetic analyses, microtubule (MT) binding competition assays, and hydrogen/deuterium-exchange studies show that KSPA-1 does not compete directly for microtubule binding. Rather, this compound acts by driving a conformational change in the KSP motor domain and disrupts productive ATP turnover stimulated by MT. These findings provide a novel mechanism for targeting KSP and perhaps other mitotic kinesins.  相似文献

The study of the metastable states, obtained by thermal quenching or by light irradiation in the [{Co(prm)(2)}(2){Co(H(2)O)(2)}{W(CN)(8)}(2)]·4H(2)O complex, is reported using powder X-ray diffraction, Raman spectroscopy, optical reflectivity, and magnetic measurements. This compound is characterized by a electron-transfer (ET) phase transition occurring between a high-temperature phase (HT phase) formed by paramagnetic Co(II)-W(V) units and a low-temperature phase (LT phase) formed by diamagnetic Co(III)-W(IV) units. Metastable phases can be induced at low temperature either by thermal quenching rapidly cooling phase named RC or by irradiation photo-induced phase named PI similar to the well-known Light-Induced Excited Spin State Trapping effect. The relaxation dynamics of the metastable phases have been studied and revealed some differences between the RC and PI phases. The sigmoidal shape of the relaxation curves in the RC phase is in agreement with the cooperative nature of the process. Thermodynamic parameters that govern the relaxation have been determined and used to reproduce the experimental Thermal-Induced Excited Spin State Trapping curve.  相似文献

The condensation of 4-hydroxy-5,6-diaminopyrimidine with 1,3-diaryl-2,3-dihalopropanones and with dibenzoylacetylenes was used to obtain derivatives of 1H-pyrimido[4,5-b]-1,5-diazepine and pyrimido[4,5-b]pyrazine, respectively. Their spectral characteristics were studied, and the directions in the formation of 7- and 6-membered heterocycles were determined. The integrated molecular transform (FTm) is a unitary numerical index of structure that is capable of uniquely representing different molecular structure conformations with the exception of enantiomers. Other molecular indices have been derived from FTm as well as from the normalized molecular moment (Mn), for example, the analogous electronic and charge transforms (FTe and FTc) and moments (Me and Mc). In this study, each of these indices was calculated for up to 10 sampled conformations of each of the C1–C10 normal alkanes as they were subjected to a standard annealing process. Statistical analyses of the resulting data in the individual series and subsequent box plots, permitting facile examination of those results, indicated that the respective transform indices (FTm, FTe, FTc) are unique, that is, with no statistically significantly overlap across the series. For the Mn and Me indices, the numerical values for methane overlapped those of ethane in the first instance and both ethane and propane in the second. The Mc index values overlapped in several instances in the series. Inasmuch as the noted molecular indices are based only on parameters of structural origin, these results have profound implications for the correlation and estimation of properties derived not only from a general structure representation, but also for those properties which may be dependent on specific molecular conformations. 4-Acetyl- and 4-succinyl-3-(2-aryl-2-oxoethyl)-3,4-dihydroquinoxalin-2(1H)-ones undergo the rearrangement into (Z)-2-(3-arylquinoxalin-2-ylidene)acetic acids accompanied by the elimination of the acyl groups. The nitration of 3-(2-oxo-2-phenylethyl)-3,4-dihydro-quinoxalin-2(1H)-one affords 5-nitro- and 7-nitro-2-carboxymethylidenequinoxalines. The bromination of quinoxalin-2-ones in AcOH gives 3-aryl-2-carboxymethylidenequinoxalines and the corresponding 7-bromo derivatives, with the former products predominating.  相似文献

The present investigation describes the synthesis and characterization of nanoparticles based on poly(acrylic acid) (PAA) intramolecularly cross-linked with diamine, 2,2′-(ethylenedioxy)bis(ethylamine), using water-soluble carbodiimide. The aqueous colloid dispersions of nanoparticles were clear or mildly opalescent depending on the ratio of cross-linking, pH of the solution, and the molecular weight of PAA, finding consistent with values of transmittance between 3% and 99%. The structure was determined by nuclear magnetic resonance spectroscopy, and the particle size was identified by dynamic light scattering (DLS) and transmission electron microscopy (TEM) measurements. It was found that particle size depends on the pH, and at a given pH, it was caused by the ratio of cross-linking and the molecular weight of PAA. Particle size measured by TEM varied in the range of 20 and 80 nm. In the swollen state, the average size of the particles measured by DLS was in the range of 35–160 nm.  相似文献
