The tridentate dianionic ligand 2-[2'-(hydroxyisopropoxyphosphoryl)phenylsulfanyl]benzoate (L(2-)) reacts with cis-[Pt(NH(3))(2)(H(2)O)(2)](2+) to form an S,O-chelate in which the O-coordinated group is either carboxylate or phosphonate, depending on the degree of protonation of the complex. Carboxylate appears to be the stronger ligand, and the stoichiometric reaction between cis-[Pt(NH(3))(2)(H(2)O)(2)](2+) and L(2-) yields the neutral species [Pt(L)(NH(3))(2)], with L bound by sulfanyl and carboxylate groups, both in solution and in the solid state. Upon protonation of [Pt(L)(NH(3))(2)], the stronger basicity of the carboxylate causes the Pt coordination to switch from carboxylate to phosphonate, and the uncoordinated carboxylate group becomes protonated. In methanolic solution, the first-order kinetics of this rearrangement could be observed by (31)P NMR spectroscopy. Both complexes-the carboxylate-bound neutral complex [Pt(L)(NH(3))(2)].H(2)O (triclinic, P1 (no. 2), a=9.529(6), b=9.766(6), c=12.299(7) angstroms, alpha=106.91(2), beta=101.71(2), gamma=102.05(2) degrees, Z=2) and the perchlorate salt of the phosphonate-bound complex [Pt(LH)(NH(3))(2)]ClO(4).H(2)O (monoclinic, P2(1)/c (no. 14), a=12.095(2), b=14.046(2), c=14.448(2) angstroms, beta=95.55(2) degrees, Z=4)-were characterized by X-ray crystallography.  相似文献

The use of microfluidic drops as microreactors hinges on the active control of certain fundamental operations such as droplet formation, transport, division and fusion. Recent work has demonstrated that local heating from a focused laser can apply a thermocapillary force on a liquid interface sufficient to block the advance of a droplet in a microchannel (C. N. Baroud, J.-P. Delville, F. Gallaire and R. Wunenburger, Phys. Rev. E: Stat., Nonlinear, Soft Matter Phys., 2007, 75(4), 046302). Here, we demonstrate the generality of this optical approach by implementing the operations mentioned above, without the need for any special microfabrication or moving parts. We concentrate on the applications to droplet manipulation by implementing a wide range of building blocks, such as a droplet valve, sorter, fuser, or divider. We also show how the building blocks may be combined by implementing a valve and fuser using a single laser spot. The underlying fundamentals, namely regarding the fluid mechanical, physico-chemical and thermal aspects, will be discussed in future publications.  相似文献

Drug discovery research often relies on the use of virtual screening via molecular docking to identify active hits in compound libraries. An area for improvement among many state-of-the-art docking methods is the accuracy of the scoring functions used to differentiate active from nonactive ligands. Many contemporary scoring functions are influenced by the physical properties of the docked molecule. This bias can cause molecules with certain physical properties to incorrectly score better than others. Since variation in physical properties is inevitable in large screening libraries, it is desirable to account for this bias. In this paper, we present a method of normalizing docking scores using virtually generated decoy sets with matched physical properties. First, our method generates a set of property-matched decoys for every molecule in the screening library. Each library molecule and its decoy set are docked using a state-of-the-art method, producing a set of raw docking scores. Next, the raw docking score of each library molecule is normalized against the scores of its decoys. The normalized score represents the probability that the raw docking score was drawn from the background distribution of nonactive property-matched decoys. Assuming that the distribution of scores of active molecules differs from the nonactive score distribution, we expect that the score of an active compound will have a low probability of having been drawn from the nonactive score distribution. In addition to the use of decoys in normalizing docking scores, we suggest that decoy sets may be a useful tool to evaluate, improve, or develop scoring functions. We show that by analyzing docking scores of library molecules with respect to the docking scores of their virtually generated property-matched decoys, one can gain insight into the advantages, limitations, and reliability of scoring functions.  相似文献

The design of bioproducts implies the use of renewable carbon but also the conversion of this carbon through clean processes. This step is often a limiting one if we consider the whole life cycle “from the raw materials to the fate of the products”. We proposed, in this work, to adapt conventional methods to the conversion of a natural raw material, the fusel oil, a co-product generated by ethanol industry to prepare acetates, carbonates and isovalerates. Selected conditions are compared to conventional routes to quantify their ecoefficiency and to check their potential development for the preparation of new biosolvents. In another step, we have calculated the volatile organic compound amount emitted during the production of a new cosmetic formulation using the fusel oil derivatives. This complete but simple example shows how to identify a real competitive alternative to the usual production chains.  相似文献

In tissues, cell microenvironment geometry and mechanics strongly impact on cell physiology. Surface micropatterning allows the control of geometry while deformable substrates of tunable stiffness are well suited for the control of the mechanics. We developed a new method to micropattern extracellular matrix proteins on poly-acrylamide gels in order to simultaneously control cell geometry and mechanics. Microenvironment geometry and mechanics impinge on cell functions by regulating the development of intra-cellular forces. We measured these forces in micropatterned cells. Micropattern geometry was streamlined to orient forces and place cells in comparable conditions. Thereby force measurement method could be simplified and applied to large-scale experiment on chip. We applied this method to mammary epithelial cells with traction force measurements in various conditions to mimic tumoral transformation. We found that, contrary to the current view, all transformation phenotypes were not always associated to an increased level of cell contractility.  相似文献

A series of neoglycoconjugates derived from deoxynojirimycin has been prepared by click connection with functionalised adamantanes. They have been assayed as glycosidase inhibitors, as inhibitors of the glycoenzymes relevant to the treatment of Gaucher disease, as well as correctors of the defective ion-transport protein involved in cystic fibrosis. We have demonstrated that it is possible to selectively either strongly inhibit ER-α-glucosidases and ceramide glucosyltransferase or restore the activity of CFTR in CF-KM4 cells by varying the length of the alkyl chain linking DNJ and adamantane.  相似文献

The calix[4]arene-imidazolium salts 5-(3-butyl-1-imidazolylium)-25,26,27,28-tetrabenzyloxy-calix[4]arene bromide (cone) (2), and 5,11-bis(3-alkyl-1-imidazolylium)-25,26,27,28-tetrabenzyloxycalix[4]arene diiodide (cone) (R = methyl, 3a; R = n-butyl, 3b) have been synthesised. Reaction of 2 in dioxane with PdCl(2) in the presence of CsCO(3) and KBr (80 °C, 24 h) gives the carbene complex trans-[PdBr(2)(calix-monocarbene)(2)] (14), containing two N-heterocyclic carbene ligands derived from 2 (yield: 63%). Repeating the reaction in pyridine instead of dioxane gives the mixed pyridine-carbene complex trans-[PdBr(2)(calix-carbene)(pyridine)] (15) in 75% yield. Treatment of the bis-imidazolium salt 3a with [Pd(OAc)(2)] affords a chelate complex, trans-[PdI(2){calix-bis(carbene)}] (16), in which a metallo-(bis-carbene) fragment caps the upper rim of the calixarene basket. Complex 16, as well as its analogue 17, obtained from 3b, display apparent C(s)-symmetry in solution. This is not the case in the solid state, a single X-ray diffraction study carried out for 16 revealing a pinced cone structure for the calixarene skeleton, which reduces the symmetry to C(1). The chelate complex 17 shows poor activity in Suzuki-Miyaura cross-coupling of phenyl boronic acid and p-tolyl halides, an observation that suggests the presence of a strained metallocyclic unit preventing easy stereochemical rearrangement to an active species. Unlike 17, complexes 14 and 15 show good activities in cross-coupling. A comparative study using the carbene precursor 1-butyl-3-(2,6-diisopropylphenyl)imidazolium bromide (18), which is devoid of the receptor fragment, strongly suggests that the carbene ligands of 14 and 15 operate typically as bulky NHC-ligands.  相似文献

Core/shell and core/shell/shell particles comprised of the Prussian blue analogues K(j)Ni(k)[Cr(CN)(6)](l)·nH(2)O (A) and Rb(a)Co(b)[Fe(CN)(6)](c)·mH(2)O (B) have been prepared for the purpose of studying persistent photoinduced magnetization in the heterostructures. Synthetic procedures have been refined to allow controlled growth of relatively thick (50-100 nm) consecutive layers of the Prussian blue analogues while minimizing the mixing of materials at the interfaces. Through changes in the order in which the two components are added, particles with AB, ABA, BA, and BAB sequences have been prepared. The two Prussian blue analogues were chosen because B is photoswitchable, and A is ferromagnetic with a relatively high magnetic ordering temperature, ~70 K, although it is not known to exhibit photoinduced changes in its magnetic properties. Magnetization measurements on the heterostructured particles performed prior to irradiation show behavior characteristic of the individual components. On the other hand, after irradiation with visible light, the heterostructures undergo persistent photoinduced changes in magnetization associated with both the B and A analogues. The results suggest that structural changes in the photoactive B component distort the normally photoinactive A component, leading to a change in its magnetization.  相似文献

Four different regioselective double capping reactions were applied either to α- or β-cyclodextrin (CD) scaffolds. The first, which relied on the use of a rigid, bulky dialkylating reagent containing two trityl-like subunits, gave access to an A,B,D,E-tetrafunctionalised β-CD regioisomer in large scale reactions. Two further capping reactions, involving the dianions PhP(2-) and S(2- , led to the synthesis of new C(1)-symmetrical β-cyclodextrins in which pairs of neighbouring glucose units are linked by very short spacers. The last double capping reaction described allowed the high-yield preparation of unprecedented α- and β-cyclodextrins containing two sulfate handles. Proximal capping turned out to be favoured for each of the above difunctional reagents. The structural characterisation of the capped species was achieved by thorough NMR investigations as well as by single-crystal X-ray diffraction studies.  相似文献

The cyclodextrin-tetraphosphine hybrid coined α-Cytep allows turnover numbers up to 340,000,000,000 and turnover frequencies up to 1,000,000,000 h(-1) to be reached in Suzuki-Miyaura reactions. These exceptional figures are clearly linked to the outstanding longevity of the reactive species induced by the ligand α-Cytep and illustrates the rising potential of cyclodextrins in catalytic applications.  相似文献
