A stripping chronopotentiometric method, using a rotating gold disk electrode for mercury measurements in sea water is described. Compared with a same method using a stationary gold film electrode, this method has a eight times higher sensitivity and a detection limit of 5 ng l−1 after 10 min deposition time. Moreover, the time needed for gold plating is eliminated. Compared with other electrochemical methods capable of measuring mercury at low concentrations, the present method is more simplified with no degassing step and no need to use a medium-exchange procedure before the stripping step. These characteristics render the method easily practicable on board oceanographic vessels for ‘in situ’ measurements.  相似文献

The cluster anion [HRu3(CO)11]- (1) reacts with dicyclohexylphosphine in THF solution to give the anionic derivative [HRu3(CO)8(PCy2)2]- (2), protonation of which yields the neutral cluster H2Ru3(CO)8(PCy2)2 (3) and, in the presence of excess phosphine, HRu3(CO)7(PCy2)3 (4). In protic methanol as reaction medium, the reaction of 1 with HPCy2 gives directly the neutral complex H2Ru3(CO)6(PCy2)2(HPCy2)2 (5), together with 4. The single-crystal structure X-ray analysis of 3 shows a closed triangular Ru3 framework. The electron count is in accordance with the EAN rule, but the structure analysis of 5 reveals an open, almost linear Ru3 skeleton, which is electron-deficient with respect to the EAN rule.  相似文献

The parallel solution-phase synthesis of two libraries of product-like compounds derived from a 1-aryltetralin privileged structure is described. The N-aryl picropodophyllone γ-lactams were synthesized from methyl ester thuriferic acid via InCl3-tandem aza-Michael addition-cyclization reaction of anilines. Bromo-aryl compounds from this library were subjected to a ligandless palladium Suzuki cross-coupling to give the expected N-biaryl picropodophyllin γ-lactams.  相似文献

We provide an unsupervised adaptive sampling strategy capable of producing μs-timescale molecular dynamics (MD) simulations of large biosystems using many-body polarizable force fields (PFFs). The global exploration problem is decomposed into a set of separate MD trajectories that can be restarted within a selective process to achieve sufficient phase-space sampling. Accurate statistical properties can be obtained through reweighting. Within this highly parallel setup, the Tinker-HP package can be powered by an arbitrary large number of GPUs on supercomputers, reducing exploration time from years to days. This approach is used to tackle the urgent modeling problem of the SARS-CoV-2 Main Protease (Mpro) producing more than 38 μs of all-atom simulations of its apo (ligand-free) dimer using the high-resolution AMOEBA PFF. The first 15.14 μs simulation (physiological pH) is compared to available non-PFF long-timescale simulation data. A detailed clustering analysis exhibits striking differences between FFs, with AMOEBA showing a richer conformational space. Focusing on key structural markers related to the oxyanion hole stability, we observe an asymmetry between protomers. One of them appears less structured resembling the experimentally inactive monomer for which a 6 μs simulation was performed as a basis for comparison. Results highlight the plasticity of the Mpro active site. The C-terminal end of its less structured protomer is shown to oscillate between several states, being able to interact with the other protomer, potentially modulating its activity. Active and distal site volumes are found to be larger in the most active protomer within our AMOEBA simulations compared to non-PFFs as additional cryptic pockets are uncovered. A second 17 μs AMOEBA simulation is performed with protonated His172 residues mimicking lower pH. Data show the protonation impact on the destructuring of the oxyanion loop. We finally analyze the solvation patterns around key histidine residues. The confined AMOEBA polarizable water molecules are able to explore a wide range of dipole moments, going beyond bulk values, leading to a water molecule count consistent with experimental data. Results suggest that the use of PFFs could be critical in drug discovery to accurately model the complexity of the molecular interactions structuring Mpro.

We study an unsteady nonlinear fluid–structure interaction problem which is a simplified model to describe blood flow through viscoelastic arteries. We consider a Newtonian incompressible two-dimensional flow described by the Navier–Stokes equations set in an unknown domain depending on the displacement of a structure, which itself satisfies a linear viscoelastic beam equation. The fluid and the structure are fully coupled via interface conditions prescribing the continuity of the velocities at the fluid–structure interface and the action–reaction principle. We prove that strong solutions to this problem are global-in-time. We obtain, in particular that contact between the viscoelastic wall and the bottom of the fluid cavity does not occur in finite time. To our knowledge, this is the first occurrence of a no-contact result, and of the existence of strong solutions globally in time, in the frame of interactions between a viscous fluid and a deformable structure.  相似文献

Differential scanning calorimetry of high molar mass poly(4‐vinylphenylboronic acid, pinacol ester)s evidenced unusual reactive events above 120 °C, resulting in a high glass‐transition temperature of 220 °C. A reversible ring‐opening reactivity of pinacol boronates is proposed, involving a nucleophilic attack on the sp2 boron and subsequent bridging between boron atoms by interconnected pinacol moieties to form a densely crosslinked network with high Tg. FTIR, solid‐state NMR investigations, and rheology studies on the polymer as well as double‐tagging analyses on molecular model structures and theoretical calculations further support this hypothesis and indicate a ring‐opening inducing crosslinking. When diluted in an apolar solvent such as toluene, the polymer network can be resolubilized via ring closing, thus recovering the entropically favored linear chains featuring cyclic boronate esters.  相似文献

BACKGROUND: Normally, gelsolin functions in plasma as part of the actin-scavenging system to assemble and disassemble actin filaments. The Asp 187-->Asn (D187N) Asp 187-->Tyr (D187Y) gelsolin mutations facilitate two proteolytic cuts in the parent protein generating a 71-residue fragment that forms amyloid fibrils in humans, putatively causing Finnish type familial amyloidosis (FAF). We investigated the role of the D187N mutation in amyloidogenicity using biophysical studies in vitro. RESULTS: Both the recombinant wild-type and D187N FAF-associated gelsolin fragments adopt an ensemble of largely unfolded structures that do not self-associate into amyloid at pH 7. 5. Incubation of either fragment at low pHs (6.0-4.0) leads to the formation of well-defined fibrils within 72 hours, however. CONCLUSIONS: The D187N mutation has been suggested to destabilize the structure of the gelsolin parent protein (specifically domain 2), facilitating two proteolytic cleavage events. Our studies demonstrate that generating the largely unstructured peptide is not sufficient alone for amyloid formation in vitro (on a time scale of months). A drop in pH or an analogous environmental change appears necessary to convert the unstructured fragment into amyloid fibrils, probably through an associative mechanism. The wild-type gelsolin fragment will make amyloid fibrils from pH 6 to 4 in vitro, but neither the wild-type fragment nor fibrils have been observed in vivo. It is possible that domain 2 of wild-type gelsolin is stable in the context of the whole protein and not susceptible to the proteolytic degradation that affords the 71-residue FAF-associated peptide.  相似文献
