Latticelike smectic liquid crystal phase in a rigid-rod helical polyisocyanide with mesogenic pendants

We report a unique macromolecule consisting of a rodlike helical polyisocyanide backbone with a narrow molecular weight distribution and rigid mesogenic chiral pendants linked via a flexible spacer that exhibits lyotropic nematic and latticelike new smectic (lat-Sm) liquid crystal phases at different concentrations. The unprecedented lat-Sm phase is associated with the smectic ordering of both the stiff polymer backbone and the rigid-rod side groups. A detailed investigation of the films using X-ray scattering and atomic force microscopy revealed a novel tilted smectic layer structure of the polymer backbone aligned perpendicular to the smectic layer of the mesogenic pendants, which arrange in an antiparallel overlapping interdigitated manner.     

Single-molecule force spectroscopy for studying kinetics of enzymatic dextran elongations

Catalytic elongation of dextran by a single molecule of dextransucrase (DSase) was directly monitored by observing the movements of the positions of a rupture peak, which represented the adhesive force between an isomaltoheptaose (dextran 7-mer)-immobilized probe and a DSase-immobilized mica surface. This was initiated with the addition of sucrose monomers. From the histograms of the rupture peaks after elongation reactions on each individual enzyme and the continuous peak shift of certain single enzymes, the catalytic elongation rate constant (k(cat)) was ascertained to be 1.2-2.7 s(-1).     

Experimental detection of proteolytic activity in a signal peptide peptidase of Arabidopsis thaliana

Background

The ubiquitin ligase COP1, COnstitutively Photomorphogenic 1, functions in many biological responses in mammalian cells, but its downstream pathway remains unclear.

Results

Here, we identified FIP200, a key regulator of mammalian autophagy, as a novel COP1-interacting protein by yeast two-hybrid screening. The interaction was confirmed by a GST-pulldown assay. Split-GFP analysis revealed that interaction between COP1 and FIP200 predominantly occurred in the cytoplasm and was enhanced in cells treated with UV irradiation. Different forms of FIP200 protein were expressed in cultured mammalian cells, and ectopic expression of COP1 reduced one of such forms.

Conclusions

These data suggest that COP1 modulates FIP200-associated activities, which may contribute to a variety of cellular functions that COP1 is involved in.

Partially coherent Fresnel diffraction by a slit aperture. III. Fresnel diffraction patterns

The intensity distribution in the Fresnel diffraction pattern of a slit aperture illuminated with partially coherent light is obtained, as a function of the coherence condition across the aperture, by evaluating the formula developed in a previous paper. It is shown that the Fresnel diffraction pattern is affected by the coherence condition of illumination across the aperture and its form.     

Exafs study of FeCl3-doped polyacetylene

FeK absorption spectra of FeCl₃-doped polyacetylene [CH(FeCl₃)_y]_x having y values of 0.024, 0.087 and 0.13, were measured by use of synchrotron radiation. The analyses of observed EXAFS data showed that Fe atom is surrounded by about four Cl atoms with Fe-Cl distance of 2.18–2.20 Å. It is concluded that the dopant exists mainly in the form of FeCl₄^-, and the following reaction takes place in the FeCl₃ doping process. 2FeCl₃ + e^- → FeCl₄^- + FeCl₂     

Evidence for muon neutrino oscillation in an accelerator-based experiment

We present results for nu(mu) oscillation in the KEK to Kamioka (K2K) long-baseline neutrino oscillation experiment. K2K uses an accelerator-produced nu(mu) beam with a mean energy of 1.3 GeV directed at the Super-Kamiokande detector. We observed the energy-dependent disappearance of nu(mu), which we presume have oscillated to nu(tau). The probability that we would observe these results if there is no neutrino oscillation is 0.0050% (4.0 sigma).     

Determination of distance of intra-molecular hydrogen bonding in (Ala-Gly)15 with silk I form after removal of the effect of MAS frequency in REDOR experiment

It is important to know the structure of silk I (Bombyx mori silk structure before spinning in the solid state) in order to understand the mechanism of fiber formation at the atomic level. In this study, 15N-dephased, 13C-observe REDOR has been carried out to determine the atomic distance of intra-molecular hydrogen bond between the 13C=O carbon of the 14th Gly residue and the 15N nitrogen of the 17th Ala residue of (AG)(6)A[1-13C]GAG[15N]AG(AG)(6) with silk I form after removal of the effect of MAS frequency on the re-coupling. The distance was determined to be 4.3A, which confirmed the intra-molecular hydrogen bonding formation between these two atomic sites.     

Novel fluorescent isoquinoline pigments, panaefluorolines A-C from the cultured mycobiont of a lichen, Amygdalaria panaeola

Novel fluorescent substances, named panaefluorolines A-C (1-3), were isolated from the cultured mycobiont of a lichen, Amygdalaria panaeola. These structures were elucidated on the basis of spectroscopic data, especially 2D NMR.
