The low temperature effect has been investigated on a set of three different silica-gel micro columns with FC-78 as eluent. An open tubular micro glass capillary, a packed micro glass capillary, and a packed micro teflon column were examined. Temperature dependences of separation factors and theoretical plate numbers were determined. It appears that a decrease in column temperature improves chromatographic selectivity while column efficiency decreases. The compensation temperatures were determined for these systems. The results suggest that the retention mechanism operative in the low temperature range is almost the same as that in the normal temperature range. Miniaturized sample preparation technique for complex sample matrices has been developed with a polymer-coated fibrous extraction medium. Several hundreds of fine fibrous materials were packed longitudinally into a fused-silica capillary followed by a polymeric coating on it to prepare the extraction capillary. The extraction capillary was installed in a liquid chromatograph as a sample loop of the injection valve. The on-line coupled sample preparation/separation system demonstrated a good validity for the analysis of phthalates in real river and wastewater samples. The lowest limits of quantification for several phthalates were less than 1 ng/ml. The effect of polymeric coating to the filaments on the extraction power was also investigated. 相似文献

Summary Deuterated compounds have been used as mobile phases for microcolumn high-performance liquid chromatography (HPLC) employing flow-cell Fourier transform infrared spectroscopy for detection. Separations were carried out on adsorption, reversed-phase, non-aqueous size-exclusion and aqueous size-exclusion chromatographic columns. Due to the IR transparency of deuterated compounds in a C–H stretching region they represent nearly ideal eluents in terms of universal detection. In addition, due to the shift in the absorption wavenumber following deuteration, deuterated solvents allow FTIR detection of solutes in other regions, where otherwise it would be prohibited, or sensitivity sacrified by interfering solvent absorption. 相似文献

The low temperature effect has been investigated in reversed phase micro high performance liquid chromatography with various aqueous mobile phase systems by an approach based on enthalpy-entropy compensation. The compensation temperatures, Tc , were determined for these systems, and the results show that the retention mechanism in a lower temperature column is similar to that in a normal temperature column in the reversed phase mode at mobile phase water contents above 9.8%. Preparative-grade bonded β- and γ-cyclodextrin stationary phases were used as the packing material of liquid chromato-graphic analytical microcolumns. Although the resulting columns are characterized by relatively low efficiency, the high selectivity of the cyclodextrin phases nevertheless allows their successful use for the separation of different classes of isomeric compounds that are difficult to resolve on conventional LC stationary phases. Examples of baseline (or almost baseline) separations of a number of isomeric compounds, including isomeric polycyclic aromatic hydrocarbons, are presented to demonstrate the analytical potential of such columns. Retention behavior of the separated isomers is discussed based on the structure of the solute molecule and the possibility of its inclusion into the molecular cavity of cyclodextrin stationary phases. 相似文献
