This work describes the development of highly efficient human DNA separation with functionalized mesoporous silica (FMS) materials. To demonstrate the electrostatic interaction effect between the target DNA molecules and FMS, three aminofunctionality types comprised of a mono-, a di-, and a tri-amine functional group were introduced on the inner surfaces of mesoporous silica particles. Systematic characterization of the synthesized materials was achieved by solid-state 29Si and 13C-NMR techniques, BET, FT-IR, and XPS. The DNA separation efficiency was explored via the function of the amino-group number, the amount used, and the added NaCl concentration. The DNA adsorption yields were high in terms of the use of triaminofunctionalized FMS at the 10 ng/L level, and the DNA desorption efficiency showed the optimum level at over 3.0 M NaCl concentration. The use of FMS in a DNA separation process provides numerous advantages over the conventional silica-based process. 相似文献

Optically pure (+)-calvine, (+)-2-epicalvine, (2S,6S)-(6-pentylpiperidin-2-yl)acetic acid methyl ester and (2R,6S)-(6-pentylpiperidin-2-yl)acetic acid methyl ester, four piperidine alkaloids isolated from ladybird beetles of the genus Calvia (Coccinellidae), were synthesised from a common precursor using cyclisative Pd(II)/Cu(II)-catalysed carboamination-(methoxy)carbonylation tandem reaction of alkenylamines as a key step. The first single-crystal X-ray analysis of (+)-calvine confirmed its proposed absolute configuration to be (2S,6S) corresponding to that of natural product. 相似文献

1,3‐Dioxolanyl‐substituted 1,2‐oxazines, such as syn‐ 1 and anti‐ 1 , rearrange under Lewis acidic conditions to provide bicyclic products 2 – 5 . Subsequent reductive transformations afforded enantiopure 3‐aminopyran derivatives such as 7 and 9 or their protected diastereomers 16 and 18 , which can be regarded as carbohydrate mimetics. An alternative sequence of transformations including selective oxidation of the primary hydroxyl groups in 21 and 24 led to two protected β‐amino acid derivatives with carbohydrate‐like backbone (sugar amino acids). Treatment of bicyclic ester 23 with samarium diiodide cleaved the N? O bond and furnished the unusual β‐lactam 27 in excellent yield. Alternatively, γ‐amino acid derivative 29 was efficiently prepared in a few steps. Fairly simple transformations gave azides 32 and 35 or alkyne 30 which are suitable substrates for the construction of oligosaccharide mimetics such as 34 by copper iodide catalyzed cycloadditions. With this report we demonstrate that enantiopure rearrangement products 2 – 5 are protected precursors of a variety of polyfunctionalized pyran derivatives with great potential for chemical biology. 相似文献

Human insulin in zinc suspension was used as a model protein to test the effect of shear on the settling rate of proteins,
a possible inference for protein denaturation. The rate of settling was determined directly in a spectropho-tometer. Shear
effects are important in retaining the activity of proteins and are present in bubble, foam, and droplet protein fractionation
processes. A sim pletest, such as that conducted here, mayeven be useful for monitoring changes in protein structure caused
by commercial shipping of the protein. The settling ratefor insulin was continously monitored in theoriginal bottle by spectrophotometric
absorbance changes as a function of time. A settling curve was determined following each shear experiment, which included
shaking the “worked” insulin solution in a vortex mixer for different lengths of time. It was determined, when comparing long
shaking times with short ones, that the initial settling rate was less for the long-term shaking of the insulin samples and
greater for the short-term shaking. The secondary effects of light and heat, along with shaking, a pparently did not produce

Summary The aim of our study was to monitor Cyclosporine A (CA) during a long-term period and to determine whether the whole-blood
levels, estimated by HPLC and radio-immunoassay (RIA), provide a relevant indicator of the toxic effect of CA, of the interference
of endogenous substances with CA and of interactions of erythromycin and ketoconazole with CA metabolims. The ratios of CA
concentrations, estimated by RIA and by HPLC were calculated in order to analyse changes in CA metabolism. A decrease in this
ratio was observed in these patients during periods of erythromycin and ketoconazole therapy deteriorating hepatic function.
We observed unusual higher CA concentrations measured by HPLC compared with RIA in heart transplant patient with marked renal
insiffuciency after one-year follow-up period of treatment of CA. The comparison of CA whole-blood levels estimated using
HPLC and RIA makes it possible to analyse the individual biotransformation of CA to its metabolites, and interference of endogenous
