Synthesis of 6-mono- and 5,6-disubstituted 1,2,3-triazolo[4,5-d]pyrimidin-7-ones

The reactions of N-substituted 4-amino-3-benzyl-1,2,3-triazole-5-carboxamides with phosphorus oxochloride and dimethylformamide at 80 °C or with triethyl orthoacetate and acetic anhydride at 160 °C afforded 6-mono- or 5,6-disubstituted 1,2,3-triazolo[4.5-d]pyrimidin-7-ones in 30—85% and 65—90% yields, respectively.     

Hydrogenation on Granular Palladium-containing Catalysts: I. Hydrogenation of Tertiary Acetylene Alcohols

Commercial granular palladium catalyst (0.5% of Pd on -Al₂O₃) was modified by treating with zinc acetate (type 1) or successively with zinc acetate and ammonia (type 2). The treatment significantly increased the hydrogenation selectivity for a triple bond into a double bond: to 85.3-93.1% with the type 1 catalyst and to 96.3-97.8% with the type 2 catalyst. A construction of an autoclave with a fixed bed of the granular catalyst is described.     

Synthesis of 3-amino-4-methyl-6-trifluoromethylthieno[2,3-b]pyridine-2-carboxanilide and its crystal and molecular structure

4-Methoxy-1,1,1-trifluoropent-3-en-2-one reacts with cyanothioacetamide to give 3-cyano-4-methyl-6-trifluoromethylpyridine-2(1H)-thione, which was transformed into 3-amino-4-methyl-6-trifluoromethylthieno[2,3-b]pyridine-2-carboxanilide by the reaction with chloroacetanilide. A straightforward preparation of benzo[f]naphtho[b][1,4]oxazepines from TNT

2,4,6-Trinitrotoluene readily reacts with 1-nitroso-2-naphthol to afford 9,11-dinitrobenzo[f]naphtho[2,1-b][1,4]oxazepine. The nitro groups of the latter undergo displacement by O- and S-nucleophiles with preferential substitution of the 11-NO₂ (peri-nitro group). The structures of the substitution products are confirmed by X-ray diffraction and ¹H NMR NOE experiments.
