1-(2′-Deoxy-2′-fluororibofuranosyl)pyrimidines were synthesized and incorporated into an RNA oligonucleotide to give 5′-r[CfGCf(UfUfCfG)GCfG]-3′ (Cf: short form of C = 2′-deoxy-2′-fluorocytidine; Uf: short form of U = 2′-deoxy-2′-fluorouridine). The oligomer was investigated by means of UV, CD, and NMR spectroscopy to address the question of how F-labels can substitute 13C-labels in the ribose ring. Through-space (NOE) and through-bond (scalar couplings) experiments were performed that make use of the ameliorated chemical-shift dispersion induced by 19F as an alternative heteronucleus. A comparison of the structures of fluorinated vs. unmodified oligomer is given. It turns out that the fluorinated oligonucleotide exists in a 14:3 equilibrium between a hairpin and a duplex conformation, in contrast to the unmodified oligonucleotide which predominantly adopts the hairpin conformation. Furthermore, the fluorinated hairpin structure adopts two distinct conformations that differ in the sugar conformation of the U and C nucleoside units, as detected by the 19F-NMR chemical shifts. The role of the 2′-OH group as stabilizing element in RNA secondary structure is discussed. 相似文献

Abstract Various trimeric co-oligomers combining 2-phenyl-1,3,4-thiadiazole mesogenic moieties with a biphenyl mesogenic moiety were synthesized and their mesomorphic behaviour investigated by polarizing microscopy, calorimetry and X-ray scattering. Such co-oligomeric structures provide an opportunity to combine different mesogenic units. Poly(ether amide)s were prepared by polycondensation of 1,13‐diamino‐4,7,11‐trioxatridecane (DTT) with the bisimidazolide or with the bis N‐hydroxysuccinimide ester of sebacic acid. Four different solvents and three different temperatures were compared. The highest molecular weights were obtained with the bisimidazolide in dimethylsulfoxide (DMSO) at 60°C. MALDI‐TOF mass spectra revealed the existence of cyclic oligoamides and polyamides in all samples. The molar fraction of cycles considerably increased with higher molecular weights of the entire sample. The polycondensations were repeated under optimum conditions in the presence of α‐cyclodextrin to prepare polydisperse catenanes consisting of α‐cyclodextrin threaded on cyclic polyamides. Yet, despite broad variation of the reaction conditions, only cylic polyamides free of cyclodextrin were isolated. Furthermore, a pseudorotaxane was prepared from DTT and α‐cyclodetrin and polycondensed with bis‐(4‐chlorophenyl)sebacate. Again, only cyclic polyamides free of cyclodextrin were detectable. 相似文献

An efficient procedure for the oxidation of primary and secondary alcohols to aldehydes and ketones, respectively, with molecular oxygen under ambient conditions has been achieved. By applying catalytic amounts of Pd(OAc)2 in the presence of tertiary phosphine oxides (O?PR3) as ligands, a variety of substrates are selectively oxidized without formation of ester byproducts. A surface-assisted laser desorption/ionization (SALDI) source is coupled to the Orbitrap mass analyzer; the instrumental approach is tested for the analysis of rhenium (Re) and osmium (Os) complexes with 8-mercaptoquinoline. Silicon (Si) material obtained by laser treatment of monocrystalline Si is used as SALDI substrate. All studied complexes are detected as radical cations, with no protonated molecules. The comparison of SALDI, matrix-assisted laser desorption/ionization (MALDI), and direct laser desorption/ionization (LDI) on metal plates in the same instrumental setup demonstrated that the detection of the studied complexes using SALDI provides the highest sensitivity. The ability to analyze samples rapidly, high purity of spectra, and good analytical parameters make SALDI coupled to the Orbitrap mass analyzer a potentially powerful tool for the detection of Re and Os complexes and related organic, UV-absorbing compounds. NMR studies of synthetic polymers and biomacromolecules, which provide insight into the conformation and dynamics of these materials, can benefit strongly from the increased sensitivity offered by dynamic nuclear polarization (DNP) and other hyperpolarizing methods. In this study 1H DNP nuclear spin hyperpolarization of two polybutadiene samples, representing a supercooled liquid and an entangled polymer melt, is demonstrated at 0.35 T magnetic field strength and at temperatures between −80 and +50 °C. Electron spin polarization transfer from the α,γ‐bisdiphenylene‐β‐phenylallyl radical to the sample nuclei is achieved by the Overhauser and solid effect. DNP signal enhancements are studied, varying the electron spin resonance offset, microwave power, and sample temperature. The influence of spin relaxation times, line widths, and molecular dynamics are discussed. The results show promising, up to 15‐fold NMR signal enhancements using noncryogenic temperatures and an inexpensive setup that is less technically demanding than current high‐field DNP setups.

The synthesis and detailed characterization of a new Ru polypyridine complex containing a heteroditopic bridging ligand with previously unexplored metal-metal distances is presented. Due to the twisted geometry of the novel ligand, the resultant division of the ligand in two distinct subunits leads to steady state as well as excited state properties of the corresponding mononuclear Ru(II) polypyridine complex resembling those of prototype [Ru(bpy)3]2+ (bpy=2,2'-bipyridine). The localization of the initially optically excited and the nature of the long-lived excited states on the Ru-facing ligand spheres is evaluated by resonance Raman and fs-TA spectroscopy, respectively, and supported by DFT and TDDFT calculations. Coordination of a second metal (Zn or Rh) to the available bis-pyrimidyl-like coordination sphere strongly influences the frontier orbitals, apparent by, for example, luminescence quenching. Thus, the new bridging ligand motif offers electronic properties, which can be adjusted by the nature of the second metal center. Using the heterodinuclear Ru−Rh complex, visible light-driven reduction of NAD+ to NADH was achieved, highlighting the potential of this system for photocatalytic applications. 相似文献

μ-Opioid receptors (μ-ORs) play a critical role in the modulation of pain and mediate the effects of the most powerful analgesic drugs. Despite extensive efforts, it remains insufficiently understood how μ-ORs produce specific effects in living cells. We developed new fluorescent ligands based on the μ-OR antagonist E-p-nitrocinnamoylamino-dihydrocodeinone (CACO), that display high affinity, long residence time and pronounced selectivity. Using these ligands, we achieved single-molecule imaging of μ-ORs on the surface of living cells at physiological expression levels. Our results reveal a high heterogeneity in the diffusion of μ-ORs, with a relevant immobile fraction. Using a pair of fluorescent ligands of different color, we provide evidence that μ-ORs interact with each other to form short-lived homodimers on the plasma membrane. This approach provides a new strategy to investigate μ-OR pharmacology at single-molecule level. 相似文献

A resistant starch (RS) mixture (MIX) consisting of fibre of potatoes (FP) and wrinkled pea starch (WPS), and high amylose maize starch (HAMS) were supplemented in adults to evaluate their effects on fat oxidation by means of a 13CO2-breath test. Sixteen subjects received a regular diet either without or with the supplementation of MIX and HAMS in randomised order. After administration of a [U-13C]algal lipid mixture, exhaled air was collected over 14?h in 0.5- and 1-h intervals. The 13C abundances were measured by nondispersive infrared spectroscopy. In comparison to the dry run (DR), supplementation with MIX and with HAMS increased the cumulative percentage dose recovery: (DR: 16.7?%, MIX: 16.9?%, HAMS: 18.0?%), but without statistical significance. The colonic degradation of MIX and HAMS to short-chain fatty acids tends to lower the formation of carbohydrate-derived acetyl-CoA and contributes to a postprandial lipid oxidation increase by using fat-derived acetyl-CoA as a compensatory fuel source. 相似文献
