Probe electrospray ionization (PESI) has recently been developed, in which the electrospray was generated from a solid needle instead of by using a capillary. In this paper, the characteristics of probe electrospray ionization were studied based on the measurement of spray current, optical microscopy, and PESI mass spectrometry. In the experiment, the solid needle was moved up and down a vertical axis, and a small amount of sample was repeatedly loaded to the needle when the tip of the needle touched the surface of the liquid sample at the lowest position. After the application of high voltage, a liquid droplet was formed on the tip of the solid needle probe, with its size was determined by the size of the needle tip. The liquid flow rate to the tip, as indicated by the spray current, depends on the voltage applied to the needle as well as the loaded liquid amount. Stable electrospray can be maintained until the total consumption of liquid sample. The kilohertz current pulsation takes place in the case of overloading the sample to the needle. The influences of the applied voltage and the liquid flow rate on the PESI mass spectra were also examined.  相似文献

We previously reported that transferrin (Tf)-modified liposomes (Tf-L) additionally modified with a cholesterylated pH-sensitive fusogenic peptide (Chol-GALA) can release an encapsulated aqueous phase marker to cytosol via endosomal membrane fusion. However, further obstacles need to be overcome to bring the Tf-L to the level of a viral-like gene delivery system. In this study, we developed a novel packaging method to encapsulate condensed plasmid DNA into PEgylated Tf-L (Tf-PEG-L) to form a core–shell-type nanoparticle. The most difficult challenge was to provide a mechanism of escape for the condensed core from endosome to cytosol in the presence of polyethylene glycol (PEG). We hypothesized that a membrane-introduced Chol-GALA and a PEgylated GALA would interact synergistically to induce membrane fusion between liposome and endosome. By simultaneously incorporating Chol-GALA into the membrane of Tf-PEG-L and GALA at tips of PEG chains, a condensed core was released into cytosol, and transfection acitivty increased 100-fold. We concluded that topological control was responsible for the synergistic effect of GALA derivatives introduced on Tf-PEG-L. During translation, the biosynthesis of polypeptides is dynamically regulated. The translation rate along messenger RNA (mRNA), which is dependent on the codon, structure, and sequence, is not always constant. However, methods for measuring the duration required for polypeptide elongation on an mRNA of interest have not been developed. In this work, we used a quartz crystal microbalance (QCM) technique to monitor mRNA translation in an Escherichia coli cell-free translation system in real time. This method permitted us to evaluate the translation of proteins of interest fused upstream of a streptavidin-binding peptide (SBP) fusion protein. The translation of mRNA encoding the SBP fusion protein alone was observed as a mass increase on a streptavidin-modified QCM plate. Addition of the protein of interest resulted in a delay in the mass change corresponding to the traveling time of the ribosome along the coding region of the protein of interest. With this technique, the lengths of coding sequences, codon usages, influences of unique sequences, and various protein-coding sequences were evaluated. The results showed that the traveling time of the translating ribosome depends on the length of the coding region translated but is also affected by the sequence itself. Differences in the time lags for various proteins imply that mRNA coding sequences may regulate gene expression.  相似文献

Cobalt-containing metal oxides [perovskites (LaCoO(3), NdCoO(3), YCoO(3), La(0.7)Sr(0.3)CoO(3)), spinel (Co(3)O(4)) and wolframite (CoWO(4))] have been examined as catalysts for photocatalytic water oxidation with Na(2)S(2)O(8) and [Ru(bpy)(3)](2+) as an electron acceptor and a photosensitizer, respectively. Catalysts with the perovskite structure exhibited higher catalytic activity as compared with the catalysts with the spinel and wolframite structures. LaCoO(3), which stabilizes Co(III) species in the perovskite structure, exhibited the highest catalytic activity in the photocatalytic water oxidation compared with CoWO(4), Co(3)O(4) and La(0.7)Sr(0.3)CoO(3) which contain Co(II) or Co(IV) species in the matrices. The high catalytic reactivity of LaCoO(3) possessing perovskite structure was maintained in NdCoO(3) and YCoO(3) which exclusively contain Co(III) species. Thus, the catalytic activity of Co ions can be controlled by the additional metal ions, which leads to development of highly reactive and robust catalysts for the photocatalytic water oxidation.  相似文献

Tetrahydroquinolines (THQs), a new class of nonsteroidal selective androgen receptor (AR) modulators, have two indispensable functional groups, that is, a hydroxyl group for AR binding and a nitro group for agonistic activity. Interestingly, switching the nitro to a cyano group, the compound acts as an antagonist. To understand this phenomenon, molecular dynamics simulations were applied for dihydrotestosterone (DHT) and representative THQs complexes with AR. Upon ligand binding, the hydroxyl group formed a tight hydrogen-bond (H-bond) with Asn705 on Helix 3 (H3). The immobilization of Asn705 on H3 is helpful in the formation of tight H-bonds with Asp890 on loop 11-12, and this immobilization consequently leads to a stabilization of H12. The difference in the DHT carbonyl isosteres affected the presence or absence of the H-bonds between the hydroxyl group of THQ and Thr877 and the distortion of H12, which is caused by the methyl group of THQ. Thus, the binding, agonist, and antagonist functions were controlled by subtle structural changes in THQ.  相似文献

We have designed new cleavable cross-linkers for biomolecules containing the 2,6-bis(oxazolinyl)pyridine (pybox) lanthanum complex. These species can effectively donate multiple charges to afford cross-linker ions for target biomolecules. The cross-linkers are cleaved easily by adding water. Moreover, we were able to detect chain structures of target molecules, including biomolecules and carbon clusters, such as fullerene C60, by the addition of some MS ionic probes. Cold-spray ionization mass spectrometry demonstrated the applications of multiply charged ionic probes as cleavable cross-linkers and polymerization reagents.  相似文献

The structure of Rh-Cr mixed-oxide (Rh(2)(-)(y)Cr(y)O(3)) nanoparticles dispersed on (Ga(1)(-)(x)Zn(x))(N(1)(-)(x)O(x)) is characterized by electron microscopy and X-ray spectroscopy. The Rh(2)(-)(y)Cr(y)O(3) nanoparticle is an efficient cocatalyst for photocatalytic overall water splitting on the (Ga(1)(-)(x)Zn(x))(N(1)(-)(x)O(x)) solid solution and is loaded onto the catalyst by impregnation from an aqueous solution containing Na(3)RhCl(6).2H(2)O and Cr(NO(3))(3).9H(2)O followed by calcination in air. Impregnation of the (Ga(1)(-)(x)Zn(x))(N(1)(-)(x)O(x)) with 1 wt % Rh and 1.5 wt % Cr followed by calcination at 623 K for 1 h provides the highest photocatalytic activity. Structural analyses reveal that the activity of this photocatalyst is strongly dependent on the generation of trivalent Rh-Cr mixed-oxide nanoparticles with optimal composition and distribution.  相似文献

An effective screening method combining parallel synthesis and solid-state CD measurements was established to identify achiral aromatic sulfonamides that show spontaneous resolution with rapidity. We found that 4 of the 12 achiral sulfonamides crystallized as chiral crystals through this method. The chirality of each sulfonamide was discriminated by solid-state CD spectra and Flack parameter in an X-ray analysis. Correspondence between the observed Cotton effect and the absolute configuration could be confirmed by time-dependent DFT calculations. [structure: see text]  相似文献

[Structure: see text] Bis(phosphine)boronium salts 3a-c were designed and prepared as key building blocks for the synthesis of highly congested diphosphinobenzenes. The preparation of sterically hindered ortho-phenylene-bridged diphosphines 1a-c was achieved by the reaction of the bis(phosphine)boronium salts 3a-c with difluorobenzenechromium complex 2 and subsequent removal of the BH2 group. The steric nature of diphosphine 1a was revealed in single-crystal X-ray analysis of its Rh complex.  相似文献
