Mg(2.56)V(1.12)W(0.88)O(8) crystals were grown from a MgO/V(2)O(5)/WO(3) melt. X-ray single-crystal diffraction studies revealed that it is orthorhombic with space group Pnma, a = 5.0658(5) A, b = 10.333(1) A, c = 17.421(2) A, Z = 6, and is isostructural with Mg(2.5)VMoO(8). Raman spectra are reported, and the assignment of the Raman bands is made by comparing the metal-oxygen vibrations of VO(4)/WO(4) tetrahedra in Mg(2.5)VWO(8) with the metal-oxygen vibrations of VO(4)/MoO(4) tetrahedra in Mg(2.5)VMoO(8). The stretching vibrations appearing at 1016 and 1035 cm(-)(1) are assigned to Mo=O and W=O double bonds, respectively, associated with the Mg(2+) cation vacancies.  相似文献

Unsolvated magnesium formate crystallizes upon reaction of the metal nitrate with formic acid in DMF at elevated temperatures. Single-crystal XRD studies reveal the formation of [Mg3(O2CH)6 [symbol: see text] DMF], 1, a metal-organic framework with DMF molecules filling the channels of an extended diamondoid lattice. The DMF molecules in 1 can be entirely removed without disruption to the framework, giving the guest-free material alpha-[Mg3(O2CH)6], 2. Compound 2 has been characterized by both powder and single-crystal XRD studies. Thermogravimetric analyses of 1 show guest loss from 120 to 190 degrees C, with decomposition of the sample at approximately 417 degrees C. Gas sorption studies using both N2 and H2 indicate that the framework displays permanent porosity. The porosity of the framework is further demonstrated by the ability of 2 to uptake a variety of small molecules upon soaking. Single-crystal XRD studies have been completed on the six inclusion compounds [Mg3(O2CH)6 [symbol: see text] THF], 3; [Mg3(O2CH)6 [symbol: see text] Et2O], 4; [Mg3(O2CH)6 [symbol: see text] Me2CO], 5; [Mg3(O2CH)6 [symbol: see text] C6H6], 6; [Mg3(O2CH)6 [symbol: see text] EtOH], 7; and [Mg3(O2CH)(6) [symbol: see text] MeOH], 8. Analyses of the metrical parameters of 1-8 indicate that the framework has the ability to contract or expand depending on the nature of the guest present.  相似文献

To model thioether-copper coordination chemistry including oxidative reactivity, such as occurs in the copper monooxygenases peptidylglycine -hydroxylating monooxygenase (PHM) and dopamine beta-hydroxylase (DbetaH), we have synthesized new tridentate N2S ligands LSEP and LSBz [LSEP = methyl(2-phenethylsulfanylpropyl)(2-pyridin-2-ylethyl)amine; LSBz = (2-benzylsulfanylpropyl)methyl(2-pyridin-2-ylethyl)amine)]. Both copper(I) and copper(II) complexes have been prepared, and their respective O2 and H2O2 chemistry has been studied. Under mild conditions, oxygenation of [(LSEP)CuI]+ (1a) and [(LSBz)CuI]+ (2a) leads to ligand sulfoxidation, thus exhibiting copper monooxygenase activity. A copper(II) complex of this sulfoxide ligand product, [(LSOEP)CuII(CH3OH)(OClO3)2], has been structurally characterized, demonstrating Cu-Osulfoxide ligation. The X-ray structure of [(LSEP)CuII(H2O)(OClO3)]+ (1b) and its solution UV-visible spectral properties [S-CuII LMCT band at 365 nm (MeCN solvent); epsilon = 4285 M-1 cm-1] indicate the thioether sulfur atom is bound to the cupric ion in both the solid (CuII-S distance: 2.31 A) and solution states. Reaction of 1b with H2O2 leads to sulfonation via the sulfoxide; excess hydrogen peroxide gives mostly sulfone product. These results may provide some insight into recent reports concerning protein methionine oxidation, showing the potential importance of copper-mediated oxidation processes in certain biological settings.  相似文献

Geometry optimization calculations were carried out on the (approximate)X (1)A(1) state of SCl(2) and the (approximate)X(2)B(1), (approximate)A(2)B(2), (approximate)B(2)A(1), (approximate)C(2)A(1), (approximate)D(2)A(2), and (approximate)E (2)B(2) states of SCl(2) (+) at the restricted-spin coupled-cluster single-double plus perturbative triple excitation [RCCSD(T)] level with basis sets of up to the augmented correlation-consistent polarized quintuple-zeta [aug-cc-pV(5+d)Z] quality. Effects of core electron correlation, basis set extension to the complete basis set limit, and relativistic contributions on computed minimum-energy geometrical parameters and/or relative electronic energies were also investigated. RCCSD(T) potential energy functions (PEFs) were calculated for the (approximate)X (1)A(1) state of SCl(2) and the low-lying states of SCl(2)(+) listed above employing the aug-cc-pV(5+d)Z basis set. Anharmonic vibrational wave functions of these neutral and cationic states of SCl(2), and Franck-Condon (FC) factors of the lowest four one-electron allowed neutral photoionizations were computed employing the RCCSD(T)aug-cc-pV(5+d)Z PEFs. Calculated FC factors with allowance for the Duschinsky rotation and anharmonicity were used to simulate the first four photoelectron (PE) bands of SCl(2). The agreement between simulated and observed He I PE spectra reported by Colton et al. [J. Electron Spectrosc. Relat. Phenom. 3, 345 (1974)] and Solouki et al. [Chem. Phys. Lett. 26, 20 (1974)] is excellent. However, our FC spectral simulations indicate that the first observed vibrational component in the first PE band of SCl(2) is a "hot" band arising from the SCl(2)(+)(approximate)X(2)B(1)(0,0,0)<--SCl(2)(approximate)X (1)A(1)(1,0,0) ionization. Consequently, the experimental adiabatic ionization energy of SCl(2) is revised to 9.55+/-0.01 eV, in excellent agreement with results obtained from state-of-the-art ab initio calculations in this work.  相似文献

We present a parallel implementation of the integral equation formalism of the polarizable continuum model for Hartree-Fock and density functional theory calculations of energies and linear, quadratic, and cubic response functions. The contributions to the free energy of the solute due to the polarizable continuum have been implemented using a master-slave approach with load balancing to ensure good scalability also on parallel machines with a slow interconnect. We demonstrate the good scaling behavior of the code through calculations of Hartree-Fock energies and linear, quadratic, and cubic response function for a modest-sized sample molecule. We also explore the behavior of the parallelization of the integral equation formulation of the polarizable continuum model code when used in conjunction with a recent scheme for the storage of two-electron integrals in the memory of the different slaves in order to achieve superlinear scaling in the parallel calculations.  相似文献

In the last decade, mass spectrometry has been employed by more and more researchers for identifying the proteins in a macromolecular complex as well as for defining the surfaces of their binding interfaces. This characterization of protein-protein interfaces usually involves at least one of several different methodologies in addition to the actual mass spectrometry. For example, limited proteolysis is often used as a first step in defining regions of a protein that are protected from proteolysis when the protein of interest is part of a macromolecular complex. Other techniques used in conjunction with mass spectrometry for determining regions of a protein involved in protein-protein interactions include chemical modification, such as covalent cross-linking, acetylation of lysines, hydrogen-deuterium exchange, or other forms of modification. In this report, both limited proteolysis and chemical modification were combined with several mass spectrometric techniques in efforts to define the protein surface on the HIV core protein, p24, recognized by two different monoclonal human antibodies that were isolated from HIV+ patients. One of these antibodies, 1571, strongly inhibits the CD4+ T cell proliferative response to a known epitope (PEVIPMFSALSEGATP), while the other antibody, 241-D, does not inhibit as strongly. The epitopes for both of these antibodies were determined to be discontinuous and localized to the N-terminus of p24. Interestingly, the epitope recognized by the strongly inhibiting antibody, 1571, completely overlaps the T cell epitope PEVIPMFSALSEGATP, while the antibody 241-D binds to a region adjacent to the region of p24 recognized by the antibody 1571. These results suggest that, possibly due to epitope competition, antibodies produced during HIV infection can negatively affect CD4+ T cell-mediated immunity against the virus.  相似文献

Self-assembled supramolecular host-guest complexes have been characterized by electrospray ionization mass spectrometry. The spectra obtained by use of a Q-TOF instrument equipped with a Z-spray ion source show primarily the 3- and 4- charge states of the assemblies. The assemblies have the general formula [guest subset Ga4L6]11- where L represents the chelating bidentate catechol ligand 1,5-bis(2',3'-dihydroxy-benzamido)naphthalene and guests are tetramethyl ammonium (Me4N+), tetraethyl ammonium (Et4N+), tetra-n-propyl ammonium (Pr4N+) and decamethylcobaltocenium (Cp*2Co+) cations. For the first time, the mass spectrum of the empty assembly [Ga4L6]12- is reported. This article also reports that provided the electrospray ion source is capable of preserving noncovalent interactions, it is possible to observe host-guest complexes containing both weak binding guests as well as sterically demanding guests in the mass spectra. The present data suggest that electrospray mass spectrometry is a powerful tool for characterization of supramolecular host-guest complexes.  相似文献

The determination of the enantiomeric composition of samples by chemometric modeling of spectral data was investigated for samples of N,N′-bis-(-methylbenzyl) sulfamide and tryptophan methyl ester hydrochloride. Multivariate regression models (PLS-1) were developed from spectral data obtained on solutions containing N,N′-bis-(-methylbenzyl)sulfamide or tryptophan methyl ester hydrochloride in the presence of sodium dodecyl sulfate and mixed cyclodextrin host molecules. The regression models were subsequently used to predict the enantiomeric composition of laboratory-prepared test samples of N,N′-bis(-methylbenzyl)sulfamide or tryptophan methyl ester hydrochloride. The capability of the models to accurately predict the enantiomeric composition was evaluated in terms of the root-mean-square percent relative error (RMS %R.E.) as calculated from the results obtained with independently prepared validation sets of samples. It was found that the presence of SDS in most cases either had little effect on the predictive ability of the model or it actually reduced the predictive ability of the model. Moreover, it was found that the use of mixed CDs, either in the presence or absence of SDS, reduced the predictive ability of the regression model when compared with results obtained with individual CDs.  相似文献

While protein-polypeptide and nucleic acid-polypeptide interactions are of significant experimental interest, quantitative methods for the characterization of such interactions are often cumbersome. Here we described a relatively simple means of optically monitoring such interactions using excimer-based peptide beacons (PBs). The design of PBs is based on the observation that, whereas short peptides are almost invariably unfolded and highly dynamic, they become rigid when complexed with macromolecular targets. Using this binding-induced folding to segregate two pyrene moieties and therefore inhibit excimer formation, we have produced PBs directed against both anti-HIV antibodies and the retroviral transactive response (TAR) RNA hairpin. For both polypeptides, target recognition is accompanied by a roughly 2-fold decrease in excimer emission, thus allowing the detection of their respective targets at concentrations of a few nanomolar. Because excimer emission requires the formation of a tight, precisely oriented pyrene dimer, even relatively trivial binding-induced segregation reduces fluorescence significantly. This suggests that the PB approach will be suitable for monitoring a wide range of peptide-macromolecule recognition events. Moreover, the synthesis of excimer-based PBs utilizes commercially available modified pyrenes in a simple and well-established protocol, making the approach well suited for routine laboratory applications.  相似文献
