Tuberculosis (TB) has been described as a global health crisis since the second half of the 1990s. Mycobacterium tuberculosis (Mtb), the etiologic agent of TB in humans, is a very successful pathogen, being the main cause of death in the population among infectious agents. In 2019, it was estimated that around 10 million individuals were contaminated by this bacillus and about 1.2 million succumbed to the disease. In recent years, our research group has reported the design and synthesis of quinoline derivatives as drug candidates for the treatment of TB. These compounds have demonstrated potent and selective growth inhibition of drug-susceptible and drug-resistant Mtb strains. Herein, a new synthetic approach was established providing efficient and rapid access (15 min) to a series of 4-alkoxy-6-methoxy-2-methylquinolines using ultrasound energy. The new synthetic protocol provides a simple procedure utilizing an open vessel system that affords the target products at satisfactory yields (45–84%) and elevated purities (≥95%). The methodology allows the evaluation of a larger number of molecules in assays against the bacillus, facilitating the determination of the structure–activity relationship with a reduced environmental cost. 相似文献

A two-dimensional liquid chromatography system coupled to ion-trap tandem mass spectrometer (2DLC-IT-MS/MS) was employed for the simultaneous quantification of pantoprazole and lansoprazole enantiomers fractions. A restricted access media of bovine serum albumin octyl column (RAM-BSA C(8)) was used in the first dimension for the exclusion of the humic substances, while a polysaccharide-based chiral column was used in the second dimension for the enantioseparation of both pharmaceuticals. The results described here show good selectivity, extraction efficiency, accuracy, and precision with detection limits of 0.200 and 0.150 μg L(-1) for the enatiomers of pantoprazole and lansoprazole respectively, while employing a small amount (1.0 mL) of native water sample per injection. This work reports an innovative assay for monitoring work, studies of biotic and abiotic enantioselective degradation and temporal changes of enantiomeric fractions. 相似文献

In this study, the kinetic for the bioconversion of sucrose to fructooligosaccharides (FOS) by free cells of Aspergillus oryzae N74 was modeled. In addition, the effect of immobilized glucose isomerase (IGI) on FOS production yield was evaluated and considered in the kinetic model. The selected kinetic models were based on a proposed reaction mechanism described by elementary rate equations and modified Michaelis?CMenten kinetic equations. The use of IGI allowed to increase the FOS production yield (FOSYield) and to decrease the glucose/fructose (G/F) ratio. At shake flask scale, the FOSYield was increased in 4.7?% (final yield 58.3?%), while the G/F ratio was reduced 6.2-fold. At bench scale, the FOSYield was increased in 2.2?% (final yield 57.3?%), while the G/F ratio was reduced 4.5-fold. The elementary rate equation model was the one that best adjusted experimental data for FOS production using either the fungus biomass or the mixture fungus biomass?CIGI, with an overall average percentage error of 7.2. Despite that FOS production yield was not highly improved by the presence of IGI in the reaction mixture, it favored the reduction of residual glucose in the mixture, avoiding the loss of material owe to glucose transformation to fructose that can be used in situ for FOS production by the fructosyltransferase. 相似文献

Sambucus nigra flowers (elderflower) have been widely used in traditional medicine for the relief of early symptoms of common cold. Its chemical composition mainly consists of polyphenolic compounds such as flavonoids, hydroxycinnamic acids, and triterpenes. Although the antioxidant properties of polyphenols are well known, the aim of this study is to assess the antioxidant and protective potentials of Sambucus nigra flowers in the human neuroblastoma (SH-SY5Y) cell line using different in vitro approaches. The antioxidant capacity is first evaluated by the oxygen radical absorbance capacity (ORAC) and the free radical scavenging activity (DPPH) methods. Cell viability is assessed by the crystal violet method; furthermore, the intracellular ROS formation (DCFH-DA method) is determined, together with the effect on the cell antioxidant defenses: reduced glutathione (GSH) and antioxidant enzyme activities (GPx, GR). On the other hand, mTORC1 hyperactivation and autophagy blockage have been associated with an increase in the formation of protein aggregates, this promoting the transference and expansion of neurodegenerative diseases. Then, the ability of Sambucus nigra flowers in the regulation of mTORC1 signaling activity and the reduction in oxidative stress through the activation of autophagy/mitophagy flux is also examined. In this regard, search for different molecules with a potential inhibitory effect on mTORC1 activation could have multiple positive effects either in the molecular pathogenic events and/or in the progression of several diseases including neurodegenerative ones. 相似文献

Thermodynamic characterisation of the adsorption process (at a low temperature) of dihydrogen on the zeolite Li-ZSM-5 was carried out by means of variable-temperature infrared spectroscopy, with the simultaneous measurement of temperature and equilibrium pressure. Adsorption renders the H–H stretching mode infrared active, at 4092 cm−1. The standard adsorption enthalpy and entropy resulted to be ΔH0=−6.5(±0.5) kJmol−1 and ΔS0=−90(±5) Jmol−1 K−1, respectively. The adsorption enthalpy is significantly larger than the liquefaction heat, and this fact renders Li-ZSM-5 a potential cryoadsorbent for hydrogen storage. 相似文献

Positive-ion mass spectral fragmentations of seven mycosporine-like amino acids (MAAs) are reported and discussed. The MAAs studied are small compounds composed of a cycloheximine ring substituted with amino acid or amino alcohol units. Techniques used include electron impact (EI) and electrospray ionization (ESI) with tandem mass spectrometry (MS/MS). ESI-MS/MS showed unusual small radical losses, generally resulting from the loss of a methyl group with the exception of shinorine and porphyra for which the initial losses were 30 and 44 Da, respectively. As expected from structural similarities, porphyra, shinorine and palythinol displayed similar fragmentation patterns, while palythenic acid and palythene fragmented in a similar manner. Overall, the ESI-MS/MS fragmentations at m/z <200 exhibited a distinctive pattern for all seven MAAs with characteristic ions at m/z 137, 168, 186, and 197 or 199. Several ions were observed for each of the MAAs analyzed, and together provide a useful and potentially diagnostic pattern for identification of MAAs and as an aid in structure elucidation of novel MAAs. For GC/EI-MS analysis, trimethylsilyl (TMS) derivatives were made. The EI-MS fragmentation patterns of TMS-MAAs showed many features typical of TMS-derivatized alpha-amines. The precursor TMS-MAA ion was not detected, but a [M-90](+ radical) ion was the highest-mass intense peak observed for palythine, palythinol and shinorine, while palythene gave a [M-116](+ radical) ion. Besides determining the number of acidic hydrogens, EI-MS of TMS-derivatized MAAs will aid in structure elucidation of novel MAAs. 相似文献

The partial least square technique (PLS) was applied to the monoclonal antibody (Mab) CB.Hep-1 immunosorbent manufacturing to determine the influence of cyanate ester concentration, ligand concentration and target ligand density on Mab coupling efficiency, elution capacity, Hepatitis B surface antigen purity and ligand leakage (output variables). Results demonstrated that cyanate ester concentration, ligand concentration and density do not have an influence on output variables in assessed ranges. Conversely, the eluted antigen purity was significantly influenced by cyanate ester concentration and ligand concentration. In conclusion, the PLS application allows for the identification of critical variables and improvement of established chromatographic processes. 相似文献
