Binding of pyrene (PyH) and its derivatives to humic acid (HA) has been studied by fluorescence spectroscopy. The nature of the interaction between HA and pyrene derivatives are extensively investigated by employing three derivatives ranging from anionic to cationic compounds: 1-pyrenebutylic acid (PyA), 1-pyrenemethanol (PyM), and 1-pyrenebutyltrimethylammonium bromide (PyB). Binding constants between HA and PyX (X=H, A, M, B) are obtained by steady-state fluorescence quenching techniques, and it is found that PyB has a markedly large binding constant among the pyrene family. This is attributed to a strong electrostatic interaction between cationic PyB and anionic HA. The result suggests that an electrostatic interaction plays a dominant role in binding of pyrenes to humic acid. The importance of electrostatic interaction was also confirmed by a salt effect on the binding constant. Influence of collisional quenching on the binding constant, which causes overestimation of the binding constant, was examined by time-resolved fluorescence spectroscopy as well as temperature effect in steady-state fluorescence measurements. In searching for tyrosinase inhibitors from plants using L-3,4-dihydroxyphenylalanine (L-DOPA) as a substrate, quercetin was found to be partially oxidized to the corresponding o-quinone under catalysis by mushroom tyrosinase (EC 1.14.18.1). Simultaneously, L-DOPA was also oxidized to dopaquinone and both o-quinones were further oxidized, respectively. The remaining quercetin partially formed adducts with dopaquinone through a Michael type addition. In general, flavonols form adducts with dopaquinone as long as their 3-hydroxyl group is free. Quercetin enhanced melanin production per cell in cultured murine B16-F10 melanoma cells, but this effect may be due in part to melanocytotoxicity. The concentration leading to 50% viable cells lost was established as 20 microM and almost complete lethality was observed at 80 microM. 相似文献

A sensitive and highly selective high-performance liquid chromatography (HPLC) method was developed for the determination of vitamin K homologues including phylloquinone (PK), menaquinone-4 (MK-4) and menaquinone-7 (MK-7) in human plasma using post-column peroxyoxalate chemiluminescence (PO-CL) detection following on-line ultraviolet (UV) irradiation. The method was based on ultraviolet irradiation (254 nm, 15 W) of vitamin K to produce hydrogen peroxide and a fluorescent product at the same time, which can be determined with PO-CL detection. The separation of vitamin K by HPLC was accomplished isocratically on an ODS column within 35 min. The method involves the use of 2-methyl-3-pentadecyl-1,4-naphthoquinone as an internal standard. The detection limits (signal-to-noise ratio = 3) were 32, 38 and 85 fmol for PK, MK-4 and MK-7, respectively. The recoveries of PK, MK-4 and MK-7 were greater than 82% and the inter- and intra-assay R.S.D. values were 1.9-5.4%. The sensitivity and selectivity of this method were sufficient for clinical and nutritional applications. 相似文献

Effects of co-solutes on the physical property of mannitol and sorbitol in frozen solutions and freeze-dried solids were studied as a model of controlling component crystallinity in pharmaceutical formulations. A frozen mannitol solution (500 mM) showed a eutectic crystallization exotherm at -22.8 degrees C, whereas sorbitol remained amorphous in the freeze-concentrated fraction in the thermal scan. Various inorganic salts reduced the eutectic mannitol crystallization peak. Trisodium and tripotassium phosphates or citrates prevented the mannitol crystallization at much lower concentrations than other salts. They also raised transition temperatures of the frozen mannitol and sorbitol solutions (T(g)': glass transition temperature of maximally freeze-concentrated amorphous phase). Crystallization of some salts (e.g., NaCl) induced crystallization of mannitol at above certain salt concentration ratios. Thermal and near-infrared analyses of cooled-melt amorphous sorbitol solids indicated increased intermolecular hydrogen-bonding in the presence of trisodium phosphate. The sodium phosphates and citrates should prevent crystallization of mannitol in frozen solutions and freeze-dried solids by the intense hydrogen-bonding and reduced molecular mobility in the amorphous phase. 相似文献

Elution behavior of diaminopimelic acid and related diamino acids using the advanced Marfey's method
The advanced Marfey's method consists of a chromatography technique for the separation of amino acids into each enantiomer by derivatization with 1-fluoro-2,4-dinitrophenyl-5-L-leucinamide (L-FDLA), and a detection method using liquid chromatography/mass spectrometry (LC/MS) which can determine the non-empirically the absolute configuration of various amino acids including the non-protein ones. However, this method has not been applied to the determination of the absolute configuration of an amino acid with a "meso" configuration such as diaminopimelic acid (A2pm). In the present study, this method was successfully applied to determine the absolute configurations of diaminosuccinic acid (DAS), A2pm, cystine (Cys), selenocystine (SeCys) and homocystine (HomoCys) using a racemization procedure and the DL-FDLA method, and the resulting elution behavior was summarized as follows: (1) the LL- and meso-isomers were eluted prior to the DD-isomer except for one case; (2) the LL- and meso-isomers are closely eluted and the elution was occasionally reversed; (3) the retention time for both the L- and D-derivatives of the meso-isomer was not changed; (4) the complementary use of the two solvent systems using CH3CN and MeOH was effective to obtain a chromatogram with a high resolution; (5) the abnormality, such as the elution order and peak shape, was observed in the elution behavior of DAS. 相似文献

A series of dendritic ligands with a 2,2'-bipyridine core was synthesized through the coupling of 4,4'-dihydroxy-2,2'-bipyridine with poly(aryl ether) dendrons. The corresponding dendritic Cu(OTf)2 catalysts were used for Diels-Alder and three-component condensation reactions. The dendritic Cu(OTf)2-catalyzed Diels-Alder reaction proceeded smoothly, and these dendritic catalysts could be recycled without deactivation by reprecipitation. Three-component condensation reactions such as Mannich-type reactions also proceeded not only in dichloromethane but also in water. A simultaneous determination of aspirin (ASA) and its metabolite, salicylic acid (SA), in human serum by a semi-micro column HPLC-UV was developed. A relatively small size of serum sample (100 microL) containing ASA and SA was cleaned up by a simple solid phase extraction. A good separation of ASA and SA could be achieved within 25 min using a semi-micro ODS column with an eluent of MeOH/0.7 mm phosphoric acid solution (pH 2.5) = 50:50 (v/v). The calibration curves for ASA and SA showed good linearity (r = 0.999) with the detection limits 114 and 38 ng/mL at a signal-to-noise ratio of 3, respectively. ASA and SA in patients' sera administered with low-dose enteric-coated aspirin were determined, and the concentration ranges obtained for ASA and SA were 1.2-2.2 and 0.5-57.3 microg/mL, respectively. 相似文献

To reliably study soil–wheel interactions, an indoor traction measurement system that allows creation of controlled soil conditions was developed. This system consisted of: (i) single wheel tester (SWT); (ii) mixing-and-compaction device (MCD) for soil preparation; (iii) soil bin; (iv) traction load device (TLD). The tire driving torque, drawbar pull, tire sinkage, position of tire lug, travel distance of the SWT and tire revolution angle were measured. It was observed that these measurements were highly reproducible under all experimental conditions. Also relationships of slip vs. sinkage and drawbar pull vs. slip showed high correlation. The tire driving torque was found to be directly influenced by the tire lug spacing. The effect of tire lug was also discussed in terms of tire slip. 相似文献

Microstructural changes, such as the density of grain boundary (GB) and dislocation density, due to isochronal annealing in severely plastic-deformed commercial purity aluminium up to 523 K was evaluated using electrical resistivity measurements and scanning transmission electron microscopy. Eventually, the GB density decreases from about 7.2 × 106 to about 2 × 106 m?1, whilst the dislocation density decreases from an initial value of around 1.3 × 1014 m?2 down to around 4 × 1013 m?2. 相似文献
