A new cyclic diarylheptanoid, acerogenin M (1), has been isolated along with nine known diarylheptanoids, 2-10, and two known phenolic compounds, 11 and 12, from a MeOH extract of the stem bark of Acer nikoense MAXIM. (Aceraceae). The structure of 1 was determined on the basis of a spectroscopic method. Upon evaluation of the inhibitory effects on 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced inflammation (1 microg/ear) in mice of nine of the compounds (2-6, 8, 10-12), six (2, 4-6, 8, 10) showed a marked anti-inflammatory effect with a 50% inhibitory dose (ID(50)) of 0.26-0.81 mg per ear. In addition, upon an evaluation against the Epstein-Barr virus early antigen (EBV-EA) activation induced by TPA for all of the compounds, all exhibited moderate inhibitory effects against EBV-EA induction (IC(50) values of 356-534 mol ratio/32 pmol TPA).  相似文献

The present work characterizes the gas-phase stacking interactions between four aromatic amino acid residues (histidine, phenylalanine, tyrosine, and tryptophan) and adenine or 3-methyladenine due to the proposed utilization of these interactions by enzymes that repair DNA alkylation damage. The MP2 potential energy surfaces of the stacked dimers are considered as a function of four variables (vertical displacement, angle of rotation, horizontal displacement, and tilt angle) using a variety of basis sets. It is found that the maximum stacking interaction energy decreases with the amino acid according to TRP > TYR approximately HIS > PHE for both nucleobases. However, the magnitude of the stacking interaction significantly increases upon alkylation (by 50-115%). Comparison of the stacking energies calculated using our surface scans to those estimated from experimental crystal structures indicates that the stacking interactions within the active site of 3-methyladenine DNA glycosylase can account for 65-75% of the maximum possible stacking interaction between the relevant molecules. The decrease in stacking in the crystal structure arises due to significant differences in the relative orientations of the nucleobase and amino acid. Nevertheless, alkylation is found to significantly increase the stacking energy when the crystal structure geometries are considered. Our calculations provide computational support for suggestions that alkylation enhances the stacking interactions within the active site of DNA repair enzymes, and they give a measure of the magnitude of this enhancement. Our results suggest that alkylation likely plays a more important role in substrate identification and removal than the nature of the aromatic amino acid that interacts with the substrate via stacking interactions.  相似文献

Alchemical free energy calculations are becoming a useful tool for calculating absolute binding free energies of small molecule ligands to proteins. Here, we find that the presence of multiple metastable ligand orientations can cause convergence problems when distance restraints alone are used. We demonstrate that the use of orientational restraints can greatly accelerate the convergence of these calculations. However, even with this acceleration, we find that sufficient sampling requires substantially longer simulations than are used in many published protocols. To further accelerate convergence, we introduce a new method of configuration space decomposition by orientation which reduces required simulation lengths by at least a factor of 5 in the cases examined. Our method is easily parallelizable, well suited for cases where a ligand cocrystal structure is not available, and can utilize initial orientations generated by docking packages.  相似文献

Uniformly sized, molecularly imprinted polymers (MIPs) of bisphenol A (BPA), one of many potential endocrine disruptors, were prepared by selective surface modification and immobilized at intervals of functional monomers with 4,4′‐methylenebisphenol as a pseudotemplate. MIPs for BPA were prepared with 4‐vinyl pyridine immobilized at the most effective interval and with ethylene glycol dimethacrylate monomer as a functional crosslinker. The prepared MIPs were surface‐modified with both polar and ionic monomers with different modification methods and then evaluated to reveal their selectivity and retention characteristics. Some of the modified MIPs showed significant selectivity for BPA retention when they were used as high‐performance liquid chromatography (HPLC) stationary phases, in comparison with ordinary MIPs. This effect of molecular imprinting was retained even after the surface modification of MIPs. The MIPs employed as pretreatment media for a column‐switching HPLC system provided a detection limit as low as 1 ng/L (ppt) by electrochemical detection. Actual samples, including Suwannee River natural organic matter (NOM), were analyzed for BPA, and BPA was quantitatively detected in NOM even with the combination with widely used UV detection because of the effective removal of interference afforded by an effective surface modification of the MIPs. © 2005 Wiley Periodicals, Inc. Shape–memory polymers (SMPs) are smart materials that can be designed to retain a metastable state and upon activation, recover a preprogrammed shape. In this study, poly(methyl acrylate) (PMA) is blended with poly(ethylene glycol) diacrylate (PEGDA) of various molecular weights in various concentrations and subsequently exposed to ionizing radiation. PEGDA sensitizes the radiation crosslinking of PMA, lowering the minimum absorbed dose for gelation and increasing the rubbery modulus, after crosslinking. Minimum dose for gelation, as determined by the Charlesby–Pinner equation, decreases from 25.57 kGy for unblended PMA to 2.06 kGy for PMA blended with 10.00 mole% PEGDA. Moreover, increase in the blend concentration of PEGDA increases the crosslinking density of the resulting networks. Sensitizer length, namely Mn of PEGDA, also affects crosslinking and final mechanical properties. Increase in the length of the PEGDA molecule at a constant molar ratio increases the efficacy of the molecule as a radiation sensitizer as determined by the increase in gel fraction and rubbery modulus across doses. However, at a constant weight ratio of PEGDA to PMA, shorter PEGDA chains sensitize more crosslinking because they have more reactive ends per weight fraction. Sensitized samples of PMA with PEGDA were tested for shape–memory properties and showed shape fixity of greater than 99%. Samples had a glass transition temperature near 28 °C and recovered between 97% and 99% of the induced strain when strained to 50%.  相似文献

We have determined that a cationic palladium(II)/(R)-BINAP complex is able to catalyze enantioselective double C–S bond-forming reactions between dithiols and dibenzyl dibromides leading to planar-chiral dithiaparacyclophanes. Although the yields and ee values of the palladium(II)-catalyzed syntheses of dithiaparacyclophanes did not exceed our previously reported cationic rhodium(I)/(S)-BINAPHANE complex-catalyzed ones, the palladium(II)-catalyzed reactions allowed the use of commercially available and inexpensive (R)-BINAP as a ligand. On the other hand, an almost racemic product was obtained by using a cationic rhodium(I)/(R)-BINAP complex as a catalyst.  相似文献

The local layer structure of surface stabilized electroclinic liquid crystals has been analysed by time-resolved synchrotron X-ray microdiffraction. At a low applied electric field, the initial bookshelf structure starts to respond above a threshold voltage. With a low to medium applied field of triangular form, the layer structure transforms reversibly between the bookshelf (low field) and the compound chevron (high field), in which the vertical and horizontal chevrons alternate along the layer. When the horizontal chevron component appears, a stripe texture can be seen in an optical micrograph. With increasing field, the horizontal chevron becomes a dominant structure while the vertical chevron still remains. The layer spacing changes in correlation with the chevron angle during the field application. At high field, surface molecules partly rearrange, resulting in alignment deterioration at the interface. The layer response time for an a.c. square wave field is of the order of a few µs to ten µs, which is close to the optical response. The appearance of the compound chevron is discussed in conjunction with the anchoring effect.  相似文献
