We have studied the characterization of thermophilic cytochrome P450 (P450st)‐didodecyldimethylammonium bromide (DDAB) films by using UV‐vis absorption, resonance Raman spectroscopy, and electrochemical methods. The observed Raman spectrum indicated near‐native conformation of the heme iron in DDAB film on the surface of a glass slide, while on the surface of a plastic‐formed carbon (PFC) electrode, the conformation of P450st‐DDAB was very similar to that of heme‐DDAB film, suggesting the release of heme from P450st in DDAB films on PFC electrodes. When NaBr was added as salt to the casting solution, the result of Raman spectrum indicated near‐native conformation of P450st in DDAB film even on the PFC electrode, but no redox potential of P450st which has near native structure was observed. This study suggests the essential experimental conditions when working with heme protein‐DDAB films as, in some cases, heme iron from proteins is released on the surface of the electrode.  相似文献

Various aryl‐, alkenyl‐, and/or alkyllithium species reacted smoothly with aryl and/or benzyl ethers with cleavage of the inert C?O bond to afford cross‐coupled products, catalyzed by commercially available [Ni(cod)2] (cod=1,5‐cyclooctadiene) catalysts with N‐heterocyclic carbene (NHC) ligands. Furthermore, the coupling reaction between the aryllithium compounds and aryl ammonium salts proceeded under mild conditions with C?N bond cleavage in the presence of a [Pd(PPh3)2Cl2] catalyst. These methods enable selective sequential functionalizations of arenes having both C?N and C?O bonds in one pot.  相似文献

Infrared photodissociation spectra of Al(+)(CH(3)OH)(n) (n = 1-4) and Al(+)(CH(3)OH)(n)-Ar (n = 1-3) were measured in the OH stretching region, 3000-3800 cm(-1). For n = 1 and 2, sharp absorption bands were observed in the free OH stretching region, all of which were well reproduced by the spectra calculated for the solvated-type geometry with no hydrogen bond. For n = 3 and 4, there were broad vibrational bands in the energy region of hydrogen-bonded OH stretching vibrations, 3000-3500 cm(-1). Energies of possible isomers for the Al(+)(CH(3)OH)(3),4 ions with hydrogen bonds were calculated in order to assign these bands. It was found that the third and fourth methanol molecules form hydrogen bonds with methanol molecules in the first solvation shell, rather than a direct bonding with the Al(+) ion. For the Al(+)(CH(3)OH)(n) clusters with n = 1-4, we obtained no evidence of the insertion reaction, which occurs in Al(+)(H(2)O)(n). One possible explanation of the difference between these two systems is that the potential energy barriers between the solvated and inserted isomers in the Al(+)(CH(3)OH)(n) system is too high to form the inserted-type isomers.  相似文献

A detailed computational study is performed on the singlet potential energy surface (PES) for possible isomerization and dissociation reactions of CH(3)CHO at the DFT (B3LYP/6-311++G(d,p)) and CCSD(T)/cc-pVTZ//B3LYP/6-311++G(d,p) levels. The pathways around the equilibrium structures can be discovered by the scaled hypersphere search (SHS) method, which enables us to make a global analysis of the PES for a given chemical composition. Fourteen isomers inclusive of 11 single-molecules and three "non-stabilized" oxygen-based ylides, 5 energetically favored complexes, and 79 interconversion transition states have been found on the singlet PES. Four lowest lying isomers with thermodynamic stability are also kinetically stable with respect to metastable intermediates. It was revealed that vinyl alcohols, which could be generated by the tautomerization of acetaldehyde, could undergo dissociation to form acetylene and water. In addition, recombination channels between some fragments, such as H(2)CO + (1)CH(2) and (1)CHOH + (1)CH(2), are energetically accessible via collision complex or oxygen-based ylides. Most of available unimolecular decompositions are found to be responsible for favorable hydrogen abstraction processes.  相似文献

Novel thermosensitive polyion complex (PIC) micelles were prepared in an aqueous medium based on the complexation of a pair of oppositely charged block ionomers, poly(2-isopropyl-2-oxazoline)-b-poly(amino acid)s (PiPrOx-b-PAA), containing thermosensitive PiPrOx segments. The controlled synthesis of PiPrOx-b-PAA was achieved via the ring-opening anionic polymerization of N-carboxyanhydrides (NCA) of either eta-benzyloxycarbonyl-l-lysine (Lys(Z)-NCA) or beta-benzyl-l-aspartate (BLA-NCA) with omega-amino-functionalized PiPrOx macroinitiators and the subsequent deprotection reaction under acidic or basic conditions. Gel permeation chromatography (GPC) and 1H NMR spectroscopy revealed that the syntheses of two block ionomers, poly(2-isopropyl-2-oxazoline)-b-poly(l-lysine) [PiPrOx-P(Lys)] and poly(2-isopropyl-2-oxazoline)-b-poly(aspartic acid) [PiPrOx-P(Asp)], proceeded almost quantitatively to give samples with a narrow molecular weight distribution (Mw/Mn 相似文献

The reaction of a mixture of 2-(1-naphthyl)benzothiazoline (HL1) and 2,6-diphenylbenzo[1,2-d:4,5-d']bisthiazoline (H3L2) with nickel(II) acetate tetrahydrate yielded three kinds of square-planar nickel(II) complexes: one nickel(II) complex with innocent ligands ([Ni(L1)2] (1c)) and two nickel(II) complexes with non-innocent ligands ([Ni(L1-L1)] (1a) and [Ni(L1-L2)] (1b)). The complex 1c has two bidentate-N,S ligands, which are formed via ring opening of HL1. On the other hand, the two complexes 1a and 1b contain a tetradentate-N2S2 ligand, which is created via ring opening of HL1 and H3L2, followed by bond formation between imino carbon atoms. Complexes 1a and 1b show very intense absorptions in the near-infrared (NIR) region, characteristic of square-planar complexes with non-innocent ligands. The third nickel(II) complex having a non-innocent tetradentate-N2S2 ligand ([Ni(L2-L2)] (2)) was prepared from H3L2 and nickel(II) acetate tetrahydrate. The electronic spectrum of 2 exhibits a very intense absorption at 981 nm (epsilon = 3.6 x 10(4) M-1 cm-1), which is significantly red-shifted compared with those of 1a (837 nm and 4.4 x 10(4) M-1 cm-1) and 1b (885 nm and 4.5 x 10(4) M-1 cm-1), indicating the presence of an extended pi delocalization. The reaction of 2,6-bis(3,5-dichlorophenyl)benzo[1,2-d:4,5-d']bisthiazoline (H3L3) with nickel(II) acetate tetrahydrate also led to the formation of a nickel(II) complex with a non-innocent ligand ([Ni(L3-L3)] (3)). While complex 3 is analogous to 2, its electrical conductivity is much higher than that of 2. The molecular structures of 1b, 1c, 2, and 3 were determined by X-ray crystallography.  相似文献

Phototropin is a blue-light photoreceptor in plants that mediates phototropism, chloroplast relocation, stomata opening and leaf expansion. Phototropin molecule has two photoreceptive domains named LOV1 (light-oxygen-voltage) and LOV2 in the N-terminus and a serine/threonine kinase domain in the C-terminus, and acts as a blue light-regulated kinase. Each LOV domain binds a flavin mononucleotide as a chromophore and undergoes unique cyclic reactions upon blue-light absorption that comprises a cysteinyl-flavin adduct formation through a triplet-excited state and a successive adduct break to revert to the initial ground state. The molecular reactions underlying the photocycle are reviewed and one of the probable molecular schemes is presented. Adduct formation alters the secondary protein structure of the LOV domains. This structural change could be transferred to the linker between the kinase domain and involved in the photoregulation of the kinase activity. The structural changes as well as the oligomeric structures seem to differ between LOV1 and LOV2, which may explain the proposed roles of each domain in the photoregulation of the kinase activity. The photoregulation mechanism of phototropin kinase is reviewed and discussed in reference to the regulation mechanism of protein kinase A, which it resembles.  相似文献

Polyion complexes (b‐PICs) are prepared by mixing single‐ or double‐stranded oligo RNA (aniomer) with poly(ethylene glycol)‐b‐poly(l ‐lysine) (PEG‐PLL) (block catiomer) to clarify the effect of aniomer chain rigidity on association behaviors at varying concentrations. Here, a 21‐mer single‐stranded RNA (ssRNA) (persistence length: 1.0 nm) and a 21‐mer double‐stranded RNA (small interfering RNA, siRNA) (persistence length: 62 nm) are compared. Both oligo RNAs form a minimal charge‐neutralized ionomer pair with a single PEG‐PLL chain, termed unit b‐PIC (uPIC), at low concentrations (<≈0.01 mg mL−1). Above the critical association concentration (≈0.01 mg mL−1), ssRNA b‐PICs form secondary associates, PIC micelles, with sizes up to 30–70 nm, while no such multimolecular assembly is observed for siRNA b‐PICs. The entropy gain associated with the formation of a segregated PIC phase in the multimolecular PIC micelles may not be large enough for rigid siRNA strands to compensate with appreciably high steric repulsion derived from PEG chains. Chain rigidity appears to be a critical parameter in polyion complex association.
