The role of intracellular magnesium ions is of high interest in the fields of pharmacology and cellular biology. To accomplish the dynamic and three-dimensional imaging of intracellular Mg2+, there is a strong desire for the development of optimized Mg2+ fluorescent probes. In this paper we describe the design, synthesis, and cellular application of the three novel Mg2+ fluorescent probes KMG-101, -103, and -104. The compounds of this series feature a charged beta-diketone as a binding site specific for Mg2+ and a fluorescein residue as the fluorophore that can be excited with an Ar+ laser such as is widely used in confocal scanning microscopy. This molecular design leads to an intensive off-on-type fluorescent response toward Mg2+ ions. The two fluorescent probes KMG-103 and -104 showed suitable dissociation constants (Kd,Mg2+ = 2 mM) and nearly a 10-fold fluorescence enhancement over the intracellular magnesium ion concentration range (0.1-6 mM), allowing high-contrast, sensitive, and selective Mg2+ measurements. For intracellular applications, the membrane-permeable probe KMG-104AM was synthesized and successfully incorporated into PC12 cells. Upon application of the mitochondria uncoupler FCCP to the probe-incorporated cells, the resulting increase in the free magnesium ion concentration could be followed over time. By using a confocal microscope, the intracellular 3D magnesium ion concentration distributions were satisfactorily observed.  相似文献

An immobilized urease reactor can be used with ion chromatography for the simultaneous determination of urea, and sodium, potassium and ammonium ions. The conversion of urea to ammonium ion was found to be 76.5%. The calibration graph for urea was linear over the range 1 × 10?5?1 × 10?3 M (RSD 3%). The effects of the beta-1,3-glucan, LELFD, obtained from liquid-cultured mycelium of Grifola frondosa, on the growth of syngeneic tumors and immune responses in mice were examined. In Meth A or IMC solid tumor systems, LELFD administered intraperitoneally (i.p.) or intralesionally (i.l.) exhibited significant antitumor effects. However, the growth of L1210 and P388 leukemias was unaffected by the injection of LELFD. The injection of LELFD i.p. enhanced the activities of natural killer cells and macrophages in mice. LELFD also enhanced the antibody response when it was injected i.p. with sheep red blood cells into mice. Furthermore, it was found that LELFD could activate the alternative complement pathway.  相似文献

This process enables the synthesis of highly substituted products with notable functional group tolerance. A new microbial electrode using immobilizedClostridium butyricum was prepared for biochemical oxygen demand (BOD) estimation of wastewaters. The current of the electrode was decreased with time until a steady state was reached. The steady state current was in all cases attained within 30-40 min at 37°C, and the maximum current output was obtained at 37°C and pH between 6.2 and 7.0. A linear relationship was obtained between the steady state current and BOD. The steady state current values were reproducible within ±7% of the relative error. The BOD of industrial wastewaters can be estimated by using the microbial electrode. Relative error of the BOD estimation of industrial wastewaters was within ± 10%. The current output of the microbial electrode was almost constant for 30 days.  相似文献

Amino acid hydantoins were detected directly in amounts of 0.2–0.5 μg with tert.-butyl hypochlorite. The amino acid hydantoins were separated by thin-layer chromatography using the following solvent systems: (I) chloroform-ethanol-formic acid (2:1:1); (II) n-butyl acetate-ethanol-formic acid (12:2:1); (III) dichloroethane-ethanol-formic acid (80:20:1); and (IV) n-butanol-acetic acid-water (15:2:5). The terminal amino groups of several proteins were detected successfully.  相似文献

We report on a spreading behavior of phospholipid membranes that arise from a lump of phospholipid (a lipid source) on topographically patterned substrates immersed in an aqueous solution. Microgrooves with well-defined shapes were prepared on Si111 surfaces by anisotropic etching in an alkaline solution. A spreading front that consists of membrane lobes and a single lipid bilayer was observed on the patterned silicon substrates by utilizing fluorescence interference contrast (FLIC) microscopy. FLIC images indicate that the membrane lobes span the microgrooves, while the underlying single lipid bilayer spread along the surface of the microgrooves. In fact, fluorescent polystyrene nanoparticles could be encapsulated in the microgrooves that were completely covered with the membrane lobes. The groove-spanning behavior of membrane lobes is discussed in terms of a balance between adhesion and bending energies of lipid bilayers.  相似文献
