1,1-Bis(dimethylamino)ethylene (ketene N,N-acetal) (1) reacted with isocyanates to give either 1 : 1 adduct 3,3-bis(dimethylamino)acrylamides (3) or 1 : 2 adduct bis(dimethylamino)methylenemalonamides (4), depending on the amount of the charged isocyanate. 3 was obtained selectively in the case of isocyanate/1 = 1, while 4 was exclusively yielded in the case of isocyanate/1 = 2. Isothiocyanate showed similar reaction behavior as isocyanate. Polyaddition of 1 with diisocyanates afforded polyamides bearing a bis(dimethylamino)methylenemalonamide group with higher molecular weight. The obtained novel polyamides are soluble in various organic solvents, and reacted with diacid chloride to give crosslinked polymer quantitatively. © 1999 John Wiley & Sons, Inc. Inhibition of fructose absorption may suppress adiposity and adiposity-related diseases caused by fructose ingestion. Eucalyptus leaf extract (ELE) inhibits intestinal fructose absorption (but not glucose absorption); however, its active compound has not yet been identified. Therefore, we evaluated the inhibitory activity of ELE obtained from Eucalyptus globulus using an intestinal fructose permeation assay with the human intestinal epithelial cell line Caco-2. The luminal sides of a cell monolayer model cultured on membrane filters were exposed to fructose with or without the ELE. Cellular fructose permeation was evaluated by measuring the fructose concentration in the medium on the basolateral side. ELE inhibited 65% of fructose absorption at a final concentration of 1 mg/mL. Oenothein B isolated from the ELE strongly inhibited fructose absorption; the inhibition rate was 63% at a final concentration of 5 μg/mL. Oenothein B did not affect glucose absorption. In contrast, the other major constituents (i.e., gallic acid and ellagic acid) showed little fructose-inhibitory activity. To our knowledge, this is the first report that oenothein B in ELE strongly inhibits fructose absorption in vitro. ELE containing oenothein B can prevent and ameliorate obesity and other diseases caused by dietary fructose consumption. 相似文献

solutions of gelling polymers and colloids such as seaweed polysaccharides (agarose, carrageenans), microbial polysaccharides
(gellan, curdlan), plant polysaccharides (methylcellulose), globular proteins (casein, glycinin, β-conglycinin), fibrous proteins
(gelatin, fibrin), and polyvinyl alcohol, which are related to foods, cosmetics, biomedical and pharmaceutical applications,
are described. Some gelation processes at a constant temperature have been treated successfully by an equation of first order
kinetics or by other modified equations, and the molecular mechanism of gel formation is discussed briefly. For water-soluble
polymers, the criterion of the gel or sol based on the frequency dependence of storage and loss moduli gives valuable informations.
TSR and DSC are complementary, and the combination of these methods has been proved to be useful.

Chemical conversion of the extract of natural resources is a very attractive way to expand the chemical space to discover bioactive compounds. In order to search for new medicines to treat parasitic diseases that cause high morbidity and mortality in affected countries in the world, the ethyl acetate extract from the rhizome of Alpinia galanga (L.) has been chemically converted by epoxidation using dioxirane generated in situ. The biological activity of chemically converted extract (CCE) of A. galanga (L.) significantly increased the activity against Leishmania major up to 82.6 ± 6.2 % at 25 μg/mL (whereas 2.7 ± 0.8% for the original extract). By bioassay-guided fractionation, new phenylpropanoids (1–6) and four known compounds, hydroquinone (7), 4-hydroxy(4-hydroxyphenyl)methoxy)benzaldehyde (8), isocoumarin cis 4-hydroxymelein (9), and (2S,3S,6R,7R,9S,10S)-humulene triepoxide (10) were isolated from CCE. The structures of isolated compounds were determined by spectroscopic analyses of 1D and 2D NMR, IR, and MS spectra. The most active compound was hydroquinone (7) with IC50 = 0.37 ± 1.37 μg/mL as a substantial active principle of CCE. In addition, the new phenylpropanoid 2 (IC50 = 27.8 ± 0.34 μg/mL) also showed significant activity against L. major compared to the positive control miltefosine (IC50 = 7.47 ± 0.3 μg/mL). The activities of the isolated compounds were also evaluated against Plasmodium falciparum, Trypanosoma brucei gambisense and Trypanosoma brucei rhodeisense. Interestingly, compound 2 was selectively active against trypanosomes with potent activity. To the best of our knowledge, this is the first report on the bioactive “unnatural” natural products from the crude extract of A. galanga (L.) by chemical conversion and on its activities against causal pathogens of leishmaniasis, trypanosomiasis, and malaria. 相似文献

Mechanistic studies are performed on the alkane hydroxylation with m-CPBA (m-chloroperbenzoic acid) catalyzed by nickel(II) complexes, NiII(L). In the oxidation of cycloalkanes, NiII(TPA) acts as an efficient catalyst with a high yield and a high alcohol selectivity. In the oxidation of adamantane, the tertiary carbon is predominantly oxidized. The reaction rate shows first-order dependence on [substrate] and [NiII(L)] but is independent on [m-CPBA]; vobs=k2[substrate][NiII(L)]. The reaction exhibited a relatively large kinetic deuterium isotope effect (KIE) of 6.7, demonstrating that the hydrogen atom abstraction is involved in the rate-limiting step of the catalytic cycle. Furthermore, NiII(L) supported by related tetradentate ligands exhibit apparently different catalytic activity, suggesting contribution of the NiII(L) in the catalytic cycle. Based on the kinetic analysis and the significant effects of O2 and CCl4 on the product distribution pattern, possible contributions of (L)NiII−O. and the aroyloxyl radical as the reactive oxidants are discussed. 相似文献

We report the potential of a small Ca2+-dependent deoxyribozyme as a novel biomaterial to distinguish RNA foldings. It is found that an immobilized deoxyribozyme using avidin-biotin interaction cleaves the target site within only single-stranded RNAs. The RNA cleavage reaction is also detected using the deoxyribozyme SPR sensor chip. Furthermore, we develop a novel NAPzyme (nucleic acid peptide deoxyribozyme) with its RNA cleavage function in the absence of divalent metal ions. 相似文献
