Preparing to read the ubiquitin code: characterization of ubiquitin trimers by top‐down mass spectrometry

The profound effects of ubiquitination on the movement and processing of cellular proteins depend exquisitely on the structures of monoubiquitin and polyubiquitin modifications. Unconjugated polyubiquitins also have a variety of intracellular functions. Structures and functions are not well correlated yet, because the structures of polyubiquitins and polyubiquitin modifications of proteins are difficult to decipher. We are moving towards a robust strategy to provide that structural information. In this report electron transfer dissociation mass spectra of six synthetic ubiquitin trimers (multiply branched proteins with molecular masses exceeding 25 600 Da) are examined using an Orbitrap Fusion Lumos instrument to determine how top‐down mass spectrometry can characterize the chain topology and linkage sites in a single, facile workflow. Design,development and first experiments on the X‐ray imaging beamline at Indus‐2 synchrotron source RRCAT,India

A full‐field hard X‐ray imaging beamline (BL‐4) was designed, developed, installed and commissioned recently at the Indus‐2 synchrotron radiation source at RRCAT, Indore, India. The bending‐magnet beamline is operated in monochromatic and white beam mode. A variety of imaging techniques are implemented such as high‐resolution radiography, propagation‐ and analyzer‐based phase contrast imaging, real‐time imaging, absorption and phase contrast tomography etc. First experiments on propagation‐based phase contrast imaging and micro‐tomography are reported.     

Synthesis of Thioglycosides from Propargyl Glycosides Exploiting Alkynophilic Gold Catalyst

Various thioglycosides were synthesized from stable propargyl glycosides using catalytic quantity of AuBr₃ and various thiol aglycons. The current protocol enables preparation of thioglycosides from propargyl glycosides in an efficient and simple manner. The reaction is fast, catalytic, and mild. Non-ideality in Born-free energy of solvation in alcohol-water and dimethylsulfoxide-acetonitrile mixtures: Solvent size ratio and ion size dependence

Recent extension of mean spherical approximation (MSA) for electrolyte solution has been employed to investigate the non-ideality in Born-free energy of solvation of a rigid, mono-positive ion in binary dipolar mixtures of associating (ethanol-water) and non-associating (dimethylsulfoxide-acetonitrile) solvents. In addition to the dipole moments, the solvent size ratio and ion size have been treated in a consistent manner in this extended MSA theory for the first time. The solvent-solvent size ratio is found to play an important role in determining the non-ideality in these binary mixtures. Smaller ions such as Li⁺ and Na⁺ show stronger non-ideality in such mixtures compared to bigger ions (for example, Cs⁺ and Bu₄N⁺). The partial solvent polarization densities around smaller ions in tertiary butanol (TBA)-water mixture is found to be very different from that in other alcohol-water mixtures as well as to that for larger ions in aqueous solutions of TBA. Non-ideality is weaker in mixtures consisting of solvent species possessing nearly equal diameters and dipole moments and is reflected in the mole fraction dependent partial solvent polarization densities.
