Treatment of trans-(Et 2O) 2MoCl 4 with 2 or 3 equiv of Na(silox) (i.e., NaOSi (t) Bu 3) afforded (silox) 3MoCl 2 ( 1-Mo) or (silox) 3MoCl ( 2-Mo). Purification of 2-Mo was accomplished via addition of PMe 3 to precipitate (silox) 3ClMoPMe 3 ( 2-MoPMe 3), followed by thermolysis to remove phosphine. Use of MoCl 3(THF) 3 with various amounts of Na(silox) produced (silox) 2ClMoMoCl(silox) 2 ( 3-Mo). Alkylation of 2-Mo with MeMgBr or EtMgBr afforded (silox) 3MoR (R = Me, 2-MoMe; Et, 2-MoEt). 2-MoEt was also synthesized from C 2H 4 and (silox) 3MoH, which was prepared from 2-Mo and NaBEt 3H. Thermolysis of WCl 6 with HOSi ( t )Bu 3 afforded (silox) 2WCl 4 ( 4-W), and sequential treatment of 4-W with Na/Hg and Na(silox) provided (silox) 3WCl 2 ( 1-W, tbp, X-ray), which was alternatively prepared from trans-(Et 2S) 2WCl 4 and 3 equiv of Tl(silox). Na/Hg reduction of 1-W generated (silox) 3WCl ( 2-W). Alkylation of 2-W with MeMgBr produced (silox) 3WMe ( 2-WMe), which dehydrogenated to (silox) 3WCH ( 6-W) with Delta H (double dagger) = 14.9(9) kcal/mol and Delta S (double dagger) = -26(2) eu. Magnetism and structural studies revealed that 2-Mo and 2-MoEt have triplet ground states (GS) and distorted trigonal monopyramid (tmp) and tmp structures, respectively. In contrast, 2-W and 2-WMe possess squashed-T d (distorted square planar) structures, and the former has a singlet GS. Quantum mechanics/molecular mechanics studies of the S = 0 and S = 1 states for full models of 2-Mo, 2-MoEt, 2-W, and 2-WMe corroborate the experimental findings and are consistent with the greater nd z (2) /( n + 1)s mixing in the third-row transition-metal species being the dominant feature in determining the structural disparity between molybdenum and tungsten. 相似文献

The Ser/Thr kinase CK2 (previously called casein kinase 2) is composed of two catalytic chains (CK2 alpha) attached to a dimer of noncatalytic subunits (CK2 beta). CK2 is involved in suppression of apoptosis, cell survival, and tumorigenesis. To investigate these activities and possibly affect them, selective CK2 inhibitors are required. An often-used CK2 inhibitor is 5,6-dichloro-1-beta-D-ribofuranosylbenzimidazole (DRB). In a complex structure with human CK2 alpha, DRB binds to the canonical ATP cleft, but additionally it occupies an allosteric site that can be alternatively filled by glycerol. Inhibition kinetic studies corroborate the dual binding mode of the inhibitor. Structural comparisons reveal a surprising conformational plasticity of human CK2 alpha around both DRB binding sites. After local rearrangement, the allosteric site serves as a CK2 beta interface. This opens the potential to construct molecules interfering with the CK2 alpha/CK2 beta interaction. 相似文献

Compounds 1-3, composed of two guanidiniocarbonylpyrrole moieties linked by oligoamide bridges and differing in number and type of basic groups, were prepared. The sites and degree of protonation of 1-3 depend strongly on the pH value. The interactions of these compounds with several double-stranded (ds) DNA and dsRNA were investigated by means of UV/Vis and CD spectroscopy as well as isothermal titration microcalorimetry (ITC). These studies revealed that the binding of 1-3 to the polynucleotides is driven by three factors, the presence of aliphatic amino groups, the protonation state of the compounds, and the steric properties of the polynucleotide binding site, that is, the shape and structure of their grooves. The results obtained by all applied methods consistently indicated that receptors 1-3 bind to the minor groove of DNA, but, by contrast, to the major groove of RNA. Additionally, it was shown by atomic force microscopy (AFM) imaging that upon interaction of compound 2 with calf thymus (ct) DNA induced aggregation of the DNA occurs, leading to pronounced changes in its secondary structure. 相似文献

The analytical performance of an axial inductively-coupled-plasma time-of-flight mass spectrometer (ICP–TOFMS) as a detector
for fast transient chromatographic signals resulting from the coupling to capillary gas chromatography (CGC) was investigated.
A cryotrapping GC–ICP–TOFMS method for the determination of volatile metal(loid) compounds (VOMs) in gases was used and the
suitability of the TOF mass analyzer for multi-elemental speciation analysis and multi-isotope ratio determinations was studied
in terms of accuracy and precision. Isotope ratios 118Sn/120Sn and 121Sb/123Sb have been determined in in-house gas standard atmospheres in Tedlar bags at two different levels (100 pg and 1 ng) for
different elemental species (SnH4, MeSnH3, Me2SnH2, Me3SnH, BuSnH3, SbH3, and MeSbH2). A limitation arising from counting statistics in both detection modes could be shown. A solution containing rhodium (10
ng mL–1) and cadmium (40 ng mL–1) was introduced simultaneously to the GC outlet. Rhodium acts as a continuous internal standard and Cd is used for mass-bias
correction (by measuring the 111Cd/113Cd ratio). The detection system in both pulse counting and analog mode was examined. The best attainable precision was established
for Me2SnH2 (analog mode, 12 replicates, 1 ng, RSD 0.34%, accuracy 0.31%) whereas most other species ranged between 0.4 and 0.5% RSD
if higher concentrations were used. The limitations of the pulse counting system are clearly seen, with peak heights of more
than 2000 counts reaching saturation (for an integration time of 100 ms), which reduces the accuracy of isotope ratio determinations.
A dozen VOM could be detected in an aged landfill gas sample; several unidentified Sn compounds were present. Although their
isotope ratios are within the confidence value of the standards, it is not yet clear if the acquired precision is good enough
to identify isotopic fractionation of metal(loid)s through biovolatilization processes. With the precision achieved, the combination
of cryotrapping GC and ICP–TOFMS is a powerful tool for monitoring volatile multi-element species in multi-tracer experiments
and isotope dilution methodology.

The gpdA-promoter-controlled exocellular production of glucose oxidase (GOD) by recombinant Aspergillus niger NRRL-3 (GOD3-18) during growth on glucose and nonglucose carbon sources was investigated. Screening of various carbon substrates
in shake-flask cultures revealed that exocellular GOD activities were not only obtained on glucose but also during growth
on mannose, fructose, and xylose. The performance of A. niger NRRL-3 (GOD3-18) using glucose, fructose, or xylose as carbon substrate was compared in more detail in bioreactor cultures.
These studies revealed that gpdA-promoter-controlled GOD synthesis was strictly coupled to cell growth. The gpdA-promoter was most active during growth on glucose. However, the unfavorable rapid GOD-catalyzed transformation of glucose
into gluconic acid, a carbon source not supporting further cell growth and GOD production, resulted in low biomass yields
and, therefore, reduced the advantageous properties of glucose. The total (endo- and exocellular) specific GOD activities
were lowest when growth occurred on fructose (only a third of the activity that was obtained on glucose), whereas utilization
of xylose resulted in total specific GOD activities nearly as high as reached during growth on glucose. Also, the portion
of GOD excreted into the culture fluid reached similar high levels (≅ 90%) by using either glucose or xylose as substrate,
whereas growth on fructose resulted in a more pelleted morphology with more than half the total GOD activity retained in the
fungal biomass. Finally, growth on xylose resulted in the highest biomass yield and, consequently, the highest total volumetric
GOD activity. These results show that xylose is the most favorable carbon substrate for gpdA-promoter-controlled production of exocellular GOD. 相似文献

Injection of carbon dioxide (CO2) into saline aquifers confined by low- permeability cap rock will result in a layer of CO2 overlying the brine. Dissolution of CO2 into the brine increases the brine density, resulting in an unstable situation in which more-dense brine overlies less-dense brine. This gravitational instability could give rise to density-driven convection of the fluid, which is a favorable process of practical interest for CO2 storage security because it accelerates the transfer of buoyant CO2 into the aqueous phase, where it is no longer subject to an upward buoyant drive. Laboratory flow visualization tests in transparent Hele-Shaw cells have been performed to elucidate the processes and rates of this CO2 solute-driven convection (CSC). Upon introduction of CO2 into the system, a layer of CO2-laden brine forms at the CO2-water interface. Subsequently, small convective fingers form, which coalesce, broaden, and penetrate into the test cell. Images and time-series data of finger lengths and wavelengths are presented. Observed CO2 uptake of the convection system indicates that the CO2 dissolution rate is approximately constant for each test and is far greater than expected for a diffusion-only scenario. Numerical simulations of our system show good agreement with the experiments for onset time of convection and advancement of convective fingers. There are differences as well, the most prominent being the absence of cell-scale convection in the numerical simulations. This cell-scale convection observed in the experiments may be an artifact of a small temperature gradient induced by the cell illumination. 相似文献
