Summary A method is described for the determination of the enantiomeric purity (enantiomeric excess) of the anticholinergic drug oxyphenonium. The method for this quaternary ammonium compound is based on the direct HPLC analysis with a chiral stationary phase. Partial filling multiple injection affinity capillary electrophoresis (PFMIACE) is used to determine binding constants between vancomycin (Van) from Streptomyces orientalis, teicoplanin (Teic) from Actinoplanes teicomyceticus and ristocetin (Rist) from Nocardia lurida to d-Ala-d-Ala terminus peptides and carbonic anhydrase B (CAB, E.C.4.2.1.1) to arylsulfonamides. Two variations of PFMIACE are described herein. In the first technique, the capillary is partially filled with ligand at increasing concentrations, a non-interacting standard, three or four separate plugs of receptor each separated by small plugs of buffer, a plug containing a second non-interacting standard, and then electrophoresed in buffer. Upon continued electrophoresis, equilibrium is established between the ligand and receptors causing a shift in the migration time of the receptors with respect to the non-interacting standards. This change in migration time is utilized for estimating multiple binding constants (Kb) for the same interaction. In the second technique, separate plugs of sample containing non-interacting standards, peptide one, buffer, and peptide two, were injected into the capillary column. The capillary is partially filled with a series of buffers containing an antibiotic at increasing concentrations and electrophoresed. Peptides migrate through the column at similar electrophoretic mobilities since their charge-to-mass ratios are approximately the same but remain as distinct zones due to the buffer plug between peptides. Upon electrophoresis, the plug of antibiotic flows into the peptide plugs affecting a shift in the migration time of the peptides with respect to the non-interacting standards occurs due to formation of the of the antibiotic-peptide complex. The shift in the migration time of the peptides upon binding to the antibiotic is used for the Scatchard analysis and measurement of a Kb. The PFMIACE technique expands the functionality and potential of ACE as an analytical tool to examine receptor-ligand interactions. In PFMIACE, a smaller amount of sample is required in the assay compared to both conventional ACE and MIACE. A series of bissilylated arenium ions 1 with different substitution patterns on the aryl ring have been synthesized by hydride abstraction from 2-aryl-substituted 2,6-dimethyl-2,6-disilaheptanes (2) via transient silylium ions. The arenium ions have been identified by their characteristic NMR chemical shifts, (delta(29)Si=19.1-25.6, delta(13)C(ipso) =89.0-102.4, delta(13)C(ortho)=160.9-182.0, delta(13)C(meta)=132.5-146.9, delta(13)C(para)=150.2-169.9) supported by quantum mechanical calculations of structures, energies, and magnetic properties at the B3LYP/6-311G(d,p)//B3LYP/6-31G(d) + DeltaZPVE level of theory. The calculations clearly reveal the charge dispersing and stabilizing effect of the silyl substituents in arenium ions 1. The bissilylated benzenium ion 1a is more stable than the parent benzenium ion (C(6)H(7)(+)) by 37.6 kcalmol(-1). The synthesized arenium ions 1 are stable in solution at room temperature for periods ranging from a few hours to days. This unusual stability is attributed to: 1) the thermodynamic stabilization of the arenium ion by two beta-silyl substituents and 2) the essentially non-nucleophilic reaction conditions (the use of the weakly coordinating [B(C(6)F(5))(4)](-) anion and aromatic hydrocarbons as solvents). Addition of stronger nucleophiles than aromatic hydrocarbons (for example, acetonitrile) results in desilylation of the arenium ion 1 and recovery of the 2-aryl-2,6-disilaheptane moiety. 相似文献

In this article, we report on the synthesis and thermotropic behaviour of methacrylic polymers containing 4′-(4-alkyloxyphenyl)azobenzene mesogens attached to the backbone through n-alkyloxy spacers of 6 or 10 methylene groups. Polymerisations were carried out via free radicals using azobisisobutyronitrile (AIBN) as initiator. Chemical structures of polymers and their precursors were characterised by 1H NMR spectroscopy. Thermogravimetric analysis showed that azopolymers are thermally stable up to temperatures around 300°C. The thermotropic liquid-crystalline (LC) behaviour was studied by differential scanning calorimetry (DSC), polarized optical microscopy (POM) and X-ray diffraction (XRD). Results indicate that all synthesised mesogens, monomers and polymers developed two or more orthogonal LC phases in wide temperature ranges. Mesogens and monomers developed nematic and smectic phases, whereas polymers exhibited only smectic phase. In polymers, the arrangement of mesogens depends on the relative length of the spacer and the terminal flexible chain; two distinct structural models were proposed based on chemical interactions and steric constrains. The trans–cis photoisomerisation of monomers and polymers in solution was also studied. High cis-isomer contents (>80%) were reached in relative short irradiation times despite the steric constrains imposed by the polymer backbone. 相似文献

A new nickel(II) σ‐aryl complex, trans‐chloro(9‐phenanthrenyl)bis(triphenylphosphine)nickel(II), was used as a precatalyst for the Suzuki–Miyaura coupling reactions of aryl chlorides. The catalytic conditions were optimized by investigating the cross‐coupling of p‐chloroanisole with phenylboronic acid. The results show that this complex is efficient for both electron‐rich and electron‐deficient aryl chlorides, though it gives better yields for activated arylboronic acids than deactivated ones. All isolated cross‐coupled biaryl products have been characterized by 1H and 13C NMR, and their spectral data are consistent with those reported. Side products from the coupling of arylboronic acid with the precatalyst complex have also been isolated and characterized, which is helpful for understanding the coupling mechanism. This study presents a novel approach for the chemical representation of lignin for modelling the reaction kinetics of lignin in lignocellulosic biomass. This methodology relies on the definition of dimeric pseudo-components containing phenolic functionalities, i.e., p-hydroxyphenyl, guaiacyl and syringyl groups, as measured in real biomass and native lignin through wet chemistry and spectroscopic techniques. The reactivities of the lignin pseudo-components are modelled through a series of lumped unidirectional reactions, whose product formation and reaction rate constants are optimised to replicate a comprehensive experimental dataset gathered from several works available in the literature. The new kinetic model contributes to the state-of-the-art by providing a more accurate depiction of the conversion rates, selectivity of char vs. volatiles, and aromatic composition in condensable products in line with the inherent reactivity of lignin functionalities and the empirical observations of lignin depolymerisation and thermal degradation at low (<1?K/s) and high heating rates (>50?K/s). 相似文献
