A simple High performance liquid chromatographic (HPLC) method for the specific determination of the molecular weight and concentration of hyaluronic acid (HA) in complex mixtures has been developed. Hyaluronate-binding proteins isolated from bovine cartilage labelled by 125I or fluoresceinisothiocyanate were used as specific markers. The specific binding affinities of the markers were compared and were found to have association constants of 1.6 x 10(7) M-1 and 1.2 x 10(7) M-1 respectively. The HA levels and molecular weight distributions can be easily determined in the range 10-500 ng/mL in complex mixtures by the use of markers, molecular sieving HPLC columns and appropriate detectors. It has been demonstrated clearly that the method is useful for the highly specific determination of the parameters in complex biological samples such as serum and synovial fluids and is recommended for clinical applications.  相似文献

Dislocation motion in the real lattice of alloys is highly complex. In a certain temperature range the dynamic strain ageing phenomena have been reported. In this paper the influence of mobile solute atoms (as obstacles) on the motion of dislocation is analysed. Both processes are assumed to be thermally activated. A new model based on this assumption is proposed. The dislocation velocity and the friction stress (due to solute-dislocation interactions) are calculated. A change in the friction stress caused by solute mobility is discussed.  相似文献

We consider a sequence of curved rods which consist of isotropic material and which are clamped on the lower base or on both bases. We study the asymptotic behaviour of the stress tensor and displacement under the assumptions of linearized elasticity when the cross‐sectional diameter of the rods tends to zero and the body force is given in the particular form. The analysis covers the case of a non‐smooth limit line of centroids. We show how the body force and the choice of the approximating curved rods can affect the strong convergence and the limit form of the stress tensor for the curved rods clamped on both bases. We study percolation in the following random environment: let Z be a Poisson process of constant intensity on ℝ2, and form the Voronoi tessellation of ℝ2 with respect to Z. Colour each Voronoi cell black with probability p, independently of the other cells. We show that the critical probability is 1/2. More precisely, if p>1/2 then the union of the black cells contains an infinite component with probability 1, while if p<1/2 then the distribution of the size of the component of black cells containing a given point decays exponentially. These results are analogous to Kesten's results for bond percolation in ℤ2. The result corresponding to Harris' Theorem for bond percolation in ℤ2 is known: Zvavitch noted that one of the many proofs of this result can easily be adapted to the random Voronoi setting. For Kesten's results, none of the existing proofs seems to adapt. The methods used here also give a new and very simple proof of Kesten's Theorem for ℤ2; we hope they will be applicable in other contexts as well. Ammonium uranate was precipitated from uranyl nitrate solution using gaseous ammonia, then filtered, washed with demineralized water and dried. The influence of pH and ammonia flow rate on their composition and structure were examined by X-ray diffraction analysis, making use of additional information obtained from infrared analysis.  相似文献

Reduced charge montmorillonites (RCM) were prepared by the thermal treatment of lithiumsaturated montmorillonite. Samples prepared by mild thermal treatment with lithium contained more water sorbed than the original montmorrilonite. When RCMs were prepared, part of the lithium cations reacted with hydroxyl groups in the octahedral sheet and released protons, which reacted with the structure. Acid treatment probably enhanced the surface area. which was reflected in the amount of water sorbed. Deprotonation of hydroxyl groups was proved by the measurements of the ignition loss. The heating of lithium saturated montmorillonite at higher temperatures brough about the collapse of the interlayers and a decrease in the amount of water sorbed.  相似文献

Oxyethylene macrocyclic compounds (crown-ethers) act as ligands of intracrystalline cations of certain layered silicates as montmorillonites. Stable intercalation materials are formed which are used to prepare organic-inorganic membranes by encapsulating these intercalation compounds with a poly-butadiene thin coating. Electrochemical Impedance Spectroscopy (EIS) is used to study the resulting composite membranes in contact with aqueous electrolytes. From the impedance plots, the ionic resistance of the membranes is obtained. The thickness of the polybutadiene coating is an important factor determining the ability of ions to pass across the membrane. Marked differences in the ionic resistance are observed as a function of the nature of the interlayer macrocyclic compound. For non-intercalated montmorillonite membranes, the ionic resistance is strongly reduced, whereas for some crown-ether intercalated materials such as 18-crown-6 and dibenzo 24-crown-8, iono-selective membranes are obtained. Concerning the nature of the electrolyte, cations exhibiting greater hydration energies show higher difficulties to pass through the membrane and, consequently, the ionic resistance increases.  相似文献

A method is described for the determination of the antitumor agent iphosphamide and seven of its metabolites in the plasma of cancer patients by multiple ion monitoring (MIM) GC-MS, mainly using the electron capture chemical ionization mode, of stable methyl and/or trifluoroacetyl derivatives. The metabolites determined were 2- and 3-dechloroethyliphosphamide, 4-ketoiphosphamide, carboxyiphosphamide, iphosphamide mustard, and two previously undetected metabolites, chloroethylamine and 1,3-oxazolidine-2-one. The isolation of the acidic and neutral metabolites was performed by solid phase extraction on to C18 adsorbent at pH 4. The weakly acidic iphosphamide mustard, isolated under these conditions with a yield of ca 50%, was measured as a stable methyltrifluoroacetyl derivative, in contrast to the corresponding phosphoramide mustard of the isomer cyclophosphamide which decomposes during derivatization. Chloroethylamine and 1,3-oxazolidine-2-one were isolated with high yield by liquid extraction with ethyl acetate at pH 10. Selective measurement of several metabolite derivatives with similar retention times was performed by multiple ion monitoring MS of specific ion masses, using a methyl phenyl siloxane capillary column previously employed in the study of cyclophosphamide metabolites. Quantitation of metabolites in patient plasma samples could be performed in the concentration range 3 ng to 20 μg per ml of original plasma.  相似文献

First-derivative spectrophotometric method with a “zero-crossing” technique of measurement has been used for the quantification of two-components mixtures of cephalothin and clavulanic acid. As the absorption bands of these drugs overlap, both direct and derivative spectrophotometric methods have been investigated and evaluated by a rigorous statistical analysis of the experimental data. The first-derivative spectrophotometric method was found to be more accurate, direct and reproducible. Beer's law was valid over the concentration range 2.0–28.0 mg/l for both compounds. The detection limits of cephalothin and clavulanic acid, at a 0.05 level of significance, were calculated to be 0.13 and 0.15 mg/l. The method was applied for determining these antibiotics in mixtures, some of them containing inject-able dosage forms of cephalothin, and so to determine both compounds in saline and glucosed physiological sera.  相似文献
