A series of combretastatin A-4 based chalcones ( 14a-l ) were designed, synthesized and these compounds examined for inhibitory effects on the proliferation of human lung (A549), breast (MCF-7), melanoma (A375), and colon (HT-29) carcinoma cells. Compounds 14b , 14c , 14e , 14h , and 14i (tri/dimethoxy, methyl, and mono/dinitro derivatives) have exhibited the most potent antiproliferative activity with IC50 < 2 μM and the hexa methoxy derivative 14b , the most promising one, which displayed the potent inhibitory activities in MCF-7 (IC50: 10 nM), A375 (IC50: 12 nM), and A549 (IC50: 65 nM) cell lines, and is 18 times more potent than the CA-4. Compound 14b represents a new scaffold and the results provide insights into further development of anticancer agents.  相似文献

A detailed study of cosmological models with constant deceleration parameterq is undertaken in the framework of Brans-Dicke theory. These models are divided into two categories: (i) singular models with expansion driven by big-bang impulse, (ii) non-singlar models with expansion driven by creation of matter particles. Prigogine's hypothesis of creation of matter out of gravitational energy is analysed and extended to BD cosmology. To accommodate the creation of new particles, the universe is regarded as an open thermodynamical system and the energy conservation equation is modified with the incorporation of a creation pressure termp c in the energy-momentum tensor . The exact solutions of the field equations of BD theory with are obtained using the power law relation=KR , which leads to models with constantq. The behaviour of the solutions is investigated for different range of values ofa. The role played by the BD scalar field and creation of matter particles in the expansion of the universe is investigated. The rotation around the amide bond in N,N-diethyl-m-toluamide (m-DEET) has been studied extensively and often used in laboratory instructions to demonstrate the phenomenon of chemical exchange. Herein, we show that a simple modification to N,N-diethyl-o-toluamide (o-DEET) significantly alters the dynamics of the restricted rotation around the amide bond due to steric interactions between the ring methyl group and the two N-ethyl groups. This alters the classic two-site exchange due to restricted rotation around the amide bond, to a three-site exchange, with the third conformation trapped at a higher-energy state compared to the other two. This often overlooked phenomenon is elucidated using variable-temperature NMR, two-dimensional exchange spectroscopy and molecular modeling studies.  相似文献

In this report a textile azo dye Remazol orange was degraded and detoxified by bacterium Pseudomonas aeruginosa BCH in plain distilled water. This bacterial decolorization performance was found to be pH and temperature dependent with maximum decolorization observed at pH 8 and temperature 30 °C. Bacterium tolerated higher dye concentrations up to 400 mg?l?1. Effect of initial cell mass showed that higher cell mass concentration can accelerate decolorization process with maximum of 92 % decolorization observed at 2.5 g?l?1 cell mass within 6.5 h. Effect of various metal ions showed Mn has inducing effect whereas Zn strongly inhibited the decolorization process at 5 mM concentration. Analysis of biodegradation products carried out with UV–vis spectroscopy, HPTLC and FTIR confirmed the decolorization and degradation of Remazol orange. Possible route for the degradation of dye was proposed based on GC-MS analysis. During toxicological scrutiny in Allium cepa root cells, induction in the activities of superoxide dismutase (SOD), guaiacol peroxidase (GPX) and inhibition of catalase (CAT) along with raised levels of lipid peroxidation and protein oxidation in dye treated samples were detected which conclusively indicated the generation of oxidative stress. Less toxic nature of the dye degraded products was observed after bacterial treatment.  相似文献

Quantitative measurement of perchlorate in biological fluids is of importance to assess its toxicity and to study its effects on the thyroid gland. Whenever possible, urine samples are preferred in toxicologic/epidemiologic studies because sample collection is non-invasive. We present here a pretreatment method for the determination of perchlorate in urine samples that lead to a clean matrix. Urine samples, spiked with isotopically labeled perchlorate, are exposed to UV to destroy/decompose organic molecules and then sequentially treated with an H+-form cation exchange resin to remove protolyzable compounds, with ammonia to raise the pH to 10-11 and finally passed through a mini-column of basic alumina to remove the color and other organic matter. After filtration through a 0.45 μm syringe filter, the sample thus prepared can be directly injected into an ion chromatograph (IC). We use ion association-electrospray ionization-mass spectrometry (ESI-MS) to detect and quantify perchlorate. The proposed sample preparation method leads to excellent limits of detection (LOD's) for perchlorate since there is essentially no dilution of sample and the matrix effects are eliminated. Results of urine samples from both men and women volunteers are reported for perchlorate, as well as for iodide and thiocyanate, which are generally present at much higher concentrations and for which a “dilute and shoot” approach is adequate. The limit of detection (S/N = 3) for iodide, thiocyanate and perchlorate by the present method was 0.40, 0.10 and 0.080 μg l−1, respectively.  相似文献

The electrochemical behavior of Hg2+ was investigated in poly(Eriochrome Black T)-modified carbon paste electrode (CPE) using cyclic voltammetry (CV). Poly(Eriochrome Black T) was prepared in an alkaline medium on the surface of the CPE using a solution of Eriochrome Black T with the CV technique. The electrochemical impedance study revealed a better charge transfer kinetics at the modified electrode. The effects of variation of the experimental conditions, such as the concentration of electrolytes, pH, deposition time, and the deposition potential, which maximize current efficiency were studied. The optimum response of Hg2+ was observed in 1.0 M KCl solution. The differential pulse anodic stripping voltammetric technique was employed successfully to detect Hg2+, which gave a good linear response at low concentration levels of Hg2+. The detection limit was found to be 2.2?×?10?10 M (S/N?=?3), which is comparable with that achieved in multiwall carbon nanotube-modified electrode. The remarkable electroanalytical performance of the modified electrode makes it amenable to employ it successfully as an electrochemical sensor for the determination of hazardous pollutant Hg2+ in environmental samples.  相似文献
