Capillary electrophoresis (CE) was employed for the determination of thermodynamic acidity constants (pKa) and actual ionic mobilities of polycationic antimicrobial peptides (AMPs). The effective electrophoretic mobilities of AMPs were measured by CE in a series of the background electrolytes within a wide pH range (2.00–12.25), at constant ionic strength (25 mM) and ambient temperature, using polybrene coated fused silica capillaries to suppress sorption of cationic AMPs to the capillary wall. Eventually, Haarhoff–Van der Linde peak fitting function was used for the determination of correct migration times of some AMPs peaks that were distorted by electromigration dispersion. The measured effective mobilities were corrected to 25°C. Mixed acidity constants, , and actual ionic mobilities, mi, of AMPs were determined by the nonlinear regression analysis of pH dependence of their effective mobilities. The values were recalculated to thermodynamic pKas using the Debye–Hückel theory. Thermodynamic pKa of imidazolium group of histidine residues was found to be in the range 3.72–4.98, pKa of α‐NH3+ group was in the range 6.14–6.93, and pKa of ε‐NH3+ group of lysine spanned the interval 7.26–9.84, depending on the particular amino acid sequence of the AMPs. Actual ionic mobilities of AMPs with positive charges from one to six elementary units achieved values (9.8 – 36.5) × 10?9 m2V?1s?1.  相似文献

New series of liquid crystals with an alkyl-lactate chiral chain and four aromatic rings in the mesogenic core and chloro-substitution were synthesized. For all the compounds studied, SmA and SmC* phases, existing over very broad temperature ranges, were detected and characterized. Subject of this work is the analysis of molecular dynamics (MD) trajectories of neurophysins I (NPI) and II (NPII) and their complexes with the neurophyseal nonapeptide hormones oxytocin (OT) and vasopresssin (VP), respectively, simulated in water. NPs serve in the neurosecretory granules as carrier proteins for the hormones before their release to the blood. The starting data consisted of two pairs of different trajectories for each of the (NPII/VP)2 and (NPI/OT)2 heterotetramers and two more trajectories for the NPII2 and NPI2 homodimers (six trajectories in total). Using essential dynamics which, to our judgement, is equivalent to factor analysis, we found that only about 10 degrees of freedom per trajectory are necessary and sufficient to describe in full the motions relevant for the function of the protein. This is consistent with these motions to explain about 90% of the total variance of the system. These principal degrees of freedom represent slow anharmonic motional modes, clearly pointing at distinguished mobility of the atoms involved in the protein's functionality.  相似文献

Spatially resolved measurements of vibrational and rotational temperature determined from the N2(C) nitrogen bands intensities have been performed by means of optical scanner of original construction. It has been found that radial variations of studied bands are independent of pressure and discharge current under our experimental conditions, i.e. in the pressure range (100–300) Pa and for discharge current up to 40 mA. Moreover, it has been found that vibrational as well as rotational temperatures stay almost constant in the radial direction. No radial changes of both temperatures can be explained by good thermal conductivity of the positive column of DC glow discharge. Rotational temperature in the oxygen DC glow discharge is determined from atmospheric A-band of molecular oxygen. Two methods of estimation of this parameter taking into account the quality of the detected spectra are analysed and compared. Radial profile of the rotational temperature is measured. No radial variations of rotational temperature have been observed in the positive column under our experimental conditions.  相似文献

Summary The aim of our study was to monitor Cyclosporine A (CA) during a long-term period and to determine whether the whole-blood levels, estimated by HPLC and radio-immunoassay (RIA), provide a relevant indicator of the toxic effect of CA, of the interference of endogenous substances with CA and of interactions of erythromycin and ketoconazole with CA metabolims. The ratios of CA concentrations, estimated by RIA and by HPLC were calculated in order to analyse changes in CA metabolism. A decrease in this ratio was observed in these patients during periods of erythromycin and ketoconazole therapy deteriorating hepatic function. We observed unusual higher CA concentrations measured by HPLC compared with RIA in heart transplant patient with marked renal insiffuciency after one-year follow-up period of treatment of CA. The comparison of CA whole-blood levels estimated using HPLC and RIA makes it possible to analyse the individual biotransformation of CA to its metabolites, and interference of endogenous substance in special clinical situations.  相似文献

Comonotonicity provides a convenient convex upper bound for a sum of random variables with arbitrary dependence structure. Improved convex upper bound was introduced via conditioning by Kaas et al. [Kaas, R., Dhaene, J., Goovaerts, M., 2000. Upper and lower bounds for sums of random variables. Insurance: Math. Econ. 27, 151-168]. In this paper, we unify these results in a more general context using the concept of conditional comonotonicity.
