Organoid is an emerging frontier technology in the field of life science, in which pluripotent stem cells or tissue-derived differentiated/progenitor cells form 3D structures according to their multi-directional differentiation potential and self-assembly ability. Nowadays, although various types of organoids are widely investigated, their construction is still complicated in operation, uncertain in yield, and poor in reproducibility for the structure and function of native organs. Constructing a biomimetic microenvironment for stem cell proliferation and differentiation in vitro is recognized as a key to driving this field. This review reviews the recent development of engineered biomimetic microenvironments for organoids. First, the composition of the matrix for organoid culture is summarized. Then, strategies for engineering the microenvironment from biophysical, biochemical, and cellular perspectives are discussed in detail. Subsequently, the newly developed monitoring technologies are also reviewed. Finally, a brief conclusion and outlook are presented for the inspiration of future research. 相似文献

Multifunctional polymer unimolecular micelles, which are used as templates to fabricate stable gold nanoparticles (GNPs) in one‐step without external reductant, have been designed and prepared. Amphiphilic 21‐arm star‐like block copolymers β‐cyclodextrin‐{poly(lactide)‐poly(2‐(dimethylamino) ethyl methacrylate)‐poly[oligo(2‐ethyl‐2‐oxazoline)methacrylate]}21 [β‐CD‐(PLA‐PDMAEMA‐PEtOxMA)21] and the precursors are synthesized by the combination of ring‐opening polymerization (ROP) and activators regenerated by electron transfer atom transfer radical polymerization (ARGET ATRP). The tertiary amine groups of PDMAEMA block reduce the counterion to zerovalent gold in situ, and these gold atoms combine mutually to form final GNPs. GNPs with relatively small size and narrow size distribution can be obtained in longer DMAEMA block copolymer, larger molar ratio of DMAEMA to HAuCl4 and smaller absolute concentrations of both polymer and HAuCl4. These results showed that the unimolecular micelles can be used as templates for preparing and stabilizing GNPs in situ without any external reducing agents and organic solvents, suggesting that the nanocomposite systems are latent nanocarriers for further biomedical application. © 2015 Wiley Periodicals, Inc. This paper focuses on chaos induced by weak A-coupled-expansion of non-autonomous discrete systems in compact subsets of metric spaces and in bounded and closed subsets of complete metric spaces, separately. A new concept of weak A-coupled-expansion for non-autonomous discrete systems, whose condition is weaker than that of A-coupled-expansion, is introduced, and several new criteria of chaos induced by weak A-coupled-expansion of non-autonomous discrete systems are established. By applying some close relationships between chaotic dynamical behaviours of the original system and its induced systems, two criteria of chaos are established. We have synthesized dual-fluorophore-doped core-shell silica nanoparticles used as ratiometric pH sensor. The nanoparticles were prepared with a reverse microemulsion technique by simultaneously encapsulating two different fluorophores, the pH-sensitive dye fluorescein as a pH indicator and the pH-insensitive dye phenosafranine as an internal reference for fluorescence ratiometric measurement, into silica shell. The nanoparticles prevent the fluorescence dyes leaching from the silica matrix when immersed inside water. The hydrophilic silica shells were made by hydrolysing and polymerizing tetraethoxysilane (TEOS) in water-in-oil microemulsion. The fluorescence intensity ratio of the two dyes varied linearly as a function of pH in the range from 4.0 to 8.0. The sensor was also applied to measure pH of real water samples. The results are in good agreements with that using the conventional glass electrode method. The as-prepared fluorescent nanoparticles showed rapid response, excellent stability and high reproducibility as pH sensors. 相似文献

VIS-NIR spectroscopy combined with multivariate analysis after the appropriate spectral data pre-treatment has been proved to be a very powerful tool for judgment of the relative pattern of the objects that have very similar properties. In this study, seventy transgenic tomatoes with antisense LeETR2 and 94 of their parents, non-transgenic ones were measured in VIS-NIR diffuse reflectance mode. Principal component analysis (PCA), discriminant analysis (DA) and partial least-squares discriminant analysis (PLSDA) were applied to classify tomatoes with different genes into two groups. Calibrations were developed using PLS regression with the leave-one-out cross-validation technique. The results show that differences between transgenic and non-transgenic tomatoes do exist and excellent classification can be obtained after optimizing spectral pre-treatment. The correct classifications for transgenic and non-transgenic tomatoes were both 100% using PLSDA after derivative spectral pre-treatment. The raw spectra with PLSDA model after the second derivative pre-treatment had the best satisfactory calibration and prediction abilities, with rc = 0.97964, root mean square error of calibration (RMSEC) = 0.099, rcv = 0.97963, root mean square error of cross-validation (RMSECV) = 0.0993 and a factor. The results in the present study show VIS-NIR spectroscopy together with chemometrics techniques could be used to differentiate transgenic tomato, which offers the benefit of avoiding time-consuming, costly and laborious chemical and sensory analysis. 相似文献

A series of 2,2'-bipyrroles has been efficiently synthesized using an improved synthetic approach based on Pd(0)-catalyzed homocoupling of various 2-iodopyrroles. This new synthetic approach takes place at room temperature and in the presence of water. Functional groups such as formyl, ester, and nitrile are able to survive these reaction conditions.
